Abstract:Inspired by biomineralization, monodispersed spherical calcites coated with a layer of polymer were obtained with the cooperative role of two polymers: carboxymethyl chitosan for size control and chondroitin sulfate for shape control. These calcites can be regarded as mesocrystals aggregated by nanoparticles with a mesoporous structure, and therefore have potential application as drug delivery systems.

Abstract:High quality CaF2: Yb, Er@mesoporous Si02 core-shell nanoparticles were prepared by coating silica shell onto solvothermal synthesized CaF2: Yb, Er nanoparticles through reverse microemulsion and liquid crystal template processes. The phase identification, morphology and upconversion luminescence properties of CaF2: Yb, Er@Si02 core-shell nanoparticles were investigated in detail. The results show that CaF2: Yb, Er nanocrystals are of cubic structure and Si02 shell is mesoporous. The core-shell structure has little effect on the upconversion properties of CaF2: Yb, Er nanoparticles. And the nanocomposites with upconversion luminescent CaF2: Yb, Er nanocrystals core and a mesoporous silica shell are potential photocontrollable drug vehicles.

Abstract:The past decades have witnessed great advances in nanotechnology since tremendous efforts have been devoted for the design, synthesis, and application of nano particles. However, for most mineral materials such as calcium sulfate, it is still a challenge to prepare their nanoparticles, especially with uniform size and high monodispersity. In this work, we report a route to regulate the morphology and structure of a-calcium sulfate hemihydrate (alpha-HH) and successfully synthesize and stabilize its mesocrystals for the first time. The ellipsoidal mesocrystals in length of 300-500 nm are composed by a-HH nanoparticles arranged in the same crystallographic fashion and interspaced with EDTA. The time-dependent experiments indicate the a-HH aggregates evolve from irregular structure to mesocrystal structure with the subsequent growth of subunits and then partially fuse into single crystals. Disorganizing the mesocrystal structure before the emergence of fusion reaps alpha-HH nanorods in a length of 30-80 nm and a width of 10-20 nm with high monodispersion. This ingenious concept paves an alternative way for nanoparticle preparation and is readily extended to other inorganic systems.
Synthesis of Size-Controlled Acid-Resistant Hybrid Calcium Carbonate Microparticles As Templates for Fabricating “Micelles-Enhanced” Polyelectrolyte Capsules by the LBL Technique

Abstract:Size-controlled, low-dispersed calcium carbonate microparticles were synthesized in the presence of the amphiphilic block copolymer polystyrene-b-poly(acrylic acid) (PS-b-PAA) by modulating the concentration of block copolymer in the reactive system. This type of hybrid microparticles have acid-resistant properties. By investigating the aggregation behaviors of PS-b-PAA micelles by transmission electron microscopy (TEM), the mechanism of hybrid calcium carbonate formation illustrated that the block copolymer served not only as "pseudonuclei" for the growth of calcium carbonate nanocrystals, but also forms the supramicelle congeries, a spherical framework, as templates for calcium carbonate nanocrystal growth into hybrid CaCO(3) particles. Moreover, this pilot study shows that the hybrid microparticle is a novel candidate as a template for fabricating multilayer polyelectrolyte capsules, in which the block copolymer is retained within the capsule interior after core removal under soft conditions. This not only facilitates the encapsulation of special materials, but also provides "micelles-enhanced" polyelectrolyte capsules.

Abstract:We report on the transformation of calcium carbonate microparticles (CCMs) to calcium phosphate microparticles (CPMs) with modulated morphologies and phases. In this additive-free transformation process, the use of templates or surfactants was circumvented, thereby eliminating potential contamination of the final products. The hydroxycarbonate apatite (HCAP) microspheres in high yield were more stable than CaCO3 microspheres in aqueous solution and had higher specific surface areas, which suggested that they had higher loading capability than their CaCO3 counterpart. In addition, the products had good biocompatibility because they were free from extraneous surfactants or stabilizers and, hence, did not require further purification. In order to examine the loading efficiency of these microspheres, carboxymethyl chitosan (CMC) and doxorubicin, which were both excellent biomedical materials, were taken as model high molecular weight and low molecular weight probes, respectively, to investigate the encapsulation capacity of CPMs. The CPMs showed high encapsulation efficiency for both molecules, with an impressive 40% loading efficiency of the adsorbed CMC biomacromolecules in the porous microparticles.

Abstract:The design and synthesis of highly ordered and oriented hierarchical structures such as mesocrystals is of great interest due to their superior performance. However, the orientation mechanism is not yet fully understood. Here, we report a subcrystallite growth controlled orientation process to synthesize anisotropic alpha-calcium sulfate hemihydrate (alpha-HH) spherical mesocrystals in a Na(2)EDTA-containing ethylene glycol-water system. The spherical mesocrystals consist of rod-like nanoparticles with parallel crystallographic alignment along the c-axis. The time-dependent experiments indicate that the growth of subcrystallites after self-assembly is crucial for mesocrystal formation, which changes the subunits from an irregular shape to a uniform rod-like shape and synchronously aligns them in the same direction. The crystalline growth of subunits mainly results from the supply of lattice ions released from Ca-EDTA complexes. The cross-section observation reveals that the growth and orientation of the subcrystallites start exteriorly and proceed inward step by step. This work deepens the understanding of mesocrystal formation and opens up a potential way for the design and synthesis of complex and highly ordered structures.
Controlled and Sustained Drug Release Performance of Calcium Sulfate Cement Porous TiO2 Microsphere Composites.

Abstract:BACKGROUND:Calcium sulphate cement (CSC) is widely used as an osteoconductive biomaterial in bone repair and regeneration.PURPOSE:In this study, porous TiO2 microspheres were added to CSC to achieve a controlled and sustained drug (gentamicin) release.METHODS:Scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray powder diffraction (XRD), and Brunauer-Emmett-Teller (BET) surface area analysis were conducted to analyse the morphology, phase composition, and surface area of the TiO2 micro-spheres and composite cements. In addition, the injection time, compressive strength, degradation behaviour, and antibacterial ability of the composite cements were examined during in vitro degradation. Gentamicin release profile was recorded using an ultraviolet spectrophotometer.RESULTS:The results revealed the excellent drug loading ability of the TiO2 microspheres. The addition of TiO2 microspheres improved the injectability and compressive strength of the composite cements, the maximum value of which was achieved at a TiO2 loading of 5 wt.%. When immersed in simulated body fluid (SBF), the composite cements doped with TiO2 microspheres were observed to release gentamicin in a stable and sustained manner, especially in the latter stages of in vitro degradation. During degradation, CSC doped with TiO2 microspheres exhibited a typical apatite-like behaviour. Further, antibacterial analysis showed that CSC doped with TiO2 microspheres exhibited long-term antibiotic activity.CONCLUSION:Thus, as an effective sustained-release formulation material, TiO2 microspheres show a great potential for application in bone cements.

Abstract:CaCO3 crystals with different hierarchical structure were fabricated by the precipitation of CaCO3 in an aqueous solution containing guar gum (GG). Through adjusting a range of parameters, the size distribution and micro-structure of CaCO3 were well controlled. Results showed that GG facilitated the formation of the hierarchical hollow sphere through assembling of nanocrystals, while inhibited the growth of CaCO3 crystals and restrained phase transition from calcite to vaterite at high concentration of GG (CGG) condition; Moreover, increasing reaction temperature led to larger particles and stable crystalline phase formed. According to the results, a morphology map was presented. Furthermore, Drug loading and releasing results ensured porous CaCO3 microspheres a promising vector for drug delivery.

Abstract:Mesoporous silica nanoparticles (MSNs) is an attractive candidate as a drug delivery carrier due to their large surface area, high pore volume and t intrinsic biocompatibility. Here, MSNs were synthesized by the hydrolysis and condensation of tetraethyl orthosilicate (TEOS) with cetyltrimethylammonium bromide (CTAB) acting as structural directing agent. A large mesopore with diameter of 3.8 to 5.5 nm of MCM-41style can be obtained via the addition of 1,3,5-trimethylbenzene. Metoprolol tartrate as a selective β1 receptor blocker was embedded on MSNs by the incipient wetness impregnation. The delivery profiles were collected in vitro in SBF at pH 7.4. A close correlation can be observed between the drug release kinetic and the mesopore size and specific surface area of MSNs.

Abstract:The novel kind flowerlike mesoporous spheres of calcium silicate hydrate (CaO center dot SiO2 center dot H2O, CSH) have been synthesized by co-precipitation method on based with water/alcohol/calvital/ tetraethyl orthosilicate (TEOS) system. The synthesized CSH were applied as a carrier to study the loaded behaviors of ibuprofen(IBU),was used to investigate the drug loading behavior of the CSH for different times has been also discussed. The drug loading capacities(DLCs) of CSH loaded for 12h has reached 116.78wt.%. The structural, morphological and textural properties were investigated by SEM, FTIR, TG and BET. The results show that it would be a good candidate as drug carrier for bone repair.

Abstract:Mesoporous calcium silicate has emerged as appealing hosts for the exploitation of novel nanocarriers featuring "on demand" drug delivery. The uniform and well-dispersed Eu3+-doped calcium silicate nanospheres have been prepared with monodisperse mesoporous silica nanospheres as template. The as-obtained spherical composite may be potentially applied as a drug carrier due to their advantages such as high biocompatibility, proper particle size (100 nm), large specific surface area, nanosized channels for drug loading and release, superior drug loading capacity and longer efficiency time of the drug release behavior. In particular, the composite exhibits intense red emission at 612 nm. After loading the model drug, the emission intensity decreases evidently. The luminescence intensity change may be potentially applied to monitor or track the drug release process. The mesoporous, biocompatible, and luminescent drug delivery system may find potential applications in fields of controlled drug delivery, cell imaging, and disease diagnosis. (C) 2016 Elsevier Ltd. Abstract:Mesoporous silica microspheres (MSMs) with a pore-size larger than 10nm and a large pore-volume have attracted considerable attention for their application in delivering poorly water-soluble drugs. Here we developed a simple method for large-scale synthesis of MSMs using sodium silicate as silica precursor. The novelty of this approach lies in the use of sucrose solution to achieve large size and volume of nanopores. The highest values of pore size and pore volume are 13.2 nm and 1.97 cm(3)/g, respectively. Importantly, the method is reliable and easily upscalable. The blank and drug-loaded MSMs were characterized by differential scanning calorimetry (DSC) and X-ray powder diffraction (XRPD). Ibuprofen and resveratrol were successfully loaded into the nanopores of MSMs in amorphous and nanocrystalline form and showed high drug-loadings and enhanced dissolution rates. This kind of MSMs appears to be a promising candidate as a new oral drug delivery vehicle providing a rapid drug release.
Synthesis and Drug Releasing Effectiveness of Hollow Mesoporous Silica Nanospheres with Ordered Mesochannels

Abstract:The mesoporous silica solid spheres with different silica condensationdegrees inside and outside were preparedviathe hydrolysis and condensation of tetraethoxysilane (TEOS). The inner part of solid spheres is more easily dissolved rather than the outer part, and the inner part of the solid spheres is dissolved in deionized water at room temperature to obtain the hollow mesoporous silica nanospheres (HMSS) with ordered mesochannels. A facile method for preparing HMSS by water dissoving at room temperature was developed. The performance of HMSS in drug loading and releasing was investigated. The results show that this method can synthesize the HMSS with a well-defined spherical morphology, uniform particle size (i.e., 460nm), high specific surface area (i.e., 1237m2/g) and ordered mesochannels (i.e., 2.7nm), and the superior performance in drug releasing. The loading content of CPT–11 is 14.50%, and the drug releasing amount in 108h at pH 7.2, 5.5 and 4.2can reach 28.49%, 39.22% and 77.72%, respectively.

Abstract:A range of mesoporous silica nanoparticles (MSNs) with controlled microstructural characteristics were successfully prepared via the binary surfactant templated synthesis approach with varied concentration of triblock copolymer Pluronic F127. The relationship between the MSNs structural evolution and the surfactant concentration was extensively discussed. Ibuprofen (IBU) was loaded as drug model to uncover the in vitro drug releasing kinetics. It was found that the quantity of the drug loaded mainly depended on the specific surface area, while the drug releasing rate was dominantly determined by the length and curvature of the mesopores. This study has uncovered the core influential factors of MSNs system on its drug releasing properties, and thus demonstrated a facile approach to prepare MSNs with manipulated structural characteristics for drug delivery applications.

Abstract:A facile approach towards the synthesis of a novel hierarchical sieve-like structure of mesoporous silica nanoparticle aggregates (hsMSNA) is reported using a centrifugal method at room temperature, based on the Kelvin-Helmholtz instability and soft-template method. The developed approach is simple and can potentially be applied for scaled-up preparation. Importantly, scanning electron microscopy, transmission electron microscopy, small-angle X-ray diffraction, and nitrogen adsorption-desorption experiments characterize the mesoporous silica as hsMSNA consisting of 40–100nm mesoporous silica nanoparticles piled at 1–2 μm cylindrical pores in sieve-like tissues. Further, various pore sizes and sieve-, mesh-, and vesicular-like structures can be obtained by adjusting the reaction conditions. The Brunauer-Emmett-Teller specific surface area is as large as 500 m2/g with a 47cm3/g pore volume, facilitating easy drug loading and delivery. Cytotoxicity assays show that the samples are not cytotoxic under a high concentration of 200 μg/mL. Finally, the high drug encapsulation efficiency and sustained release behaviors indicats the considerable potential of the hsMSNA as a drug delivery system in the field of nanomedicine.

Abstract:The synthesis of mesoporous β -tricalcium phosphate ( β -TCP) powder was performed by using the microemulsion approach, with hexadecyltrimethyl ammonium bromide (CTAB)/cyclohexane/n-octyl alcohol microemulsion system. The influences of different pH values and calcination temperatures on the phase composition of the β -TCP powder were studied. The in vitro proliferation of bone marrow mesenchymal stem cells (BMSCs) in the suspensions of β -TCP powders with meso-structure was studied. The phase composition, mesoporous structure, powder morphology, cell morphology and the optical density (OD) were characterized through X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), Fourier transform infrared (FTIR) spectroscopy, N 2 adsorption-desorption isotherms, inverted phase contrast microscopy and Multiskan spectrum, respectively. The mesoporous β -TCP powder with specific surface area of 12.85 m 2 /g and the average pore size 7.11 nm was obtained through the microemulsion approach (100 g/L CTAB/250 mL/L cyclohexane/250 mL/L n -octyl alcohol) with a controlled pH of 7.0, after calcinating the powder at 800 °C. It was confirmed that mesoporous β -TCP powder benefits the activity of BMSCs more than the non-mesoporous β -TCP powder.

Abstract:Calcium carbonate/carboxymethyl chitosan (CaCO3/CMC) hybrid microspheres and nanospheres were prepared by the precipitation of calcium carbonate in an aqueous solution containing CMC. Through adjusting the preparation conditions, the size of CaCO3/CMC hybrid particles could be easily controlled at micro- to nanometer ranges with relatively narrow size distributions. The obtained microspheres and nanospheres were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, thermogravimetric analysis, and differential scanning calorimetry. The size and size distribution were measured by a particle size analyzer. Doxorubicin hydrochloride (DOX center dot HCl), a water-soluble anticancer drug, was loaded in the hybrid microspheres and nanospheres with a high encapsulation efficiency. The in vitro drug release showed that the release of DOX center dot HCl from the microspheres and nanospheres could be effectively sustained.

Abstract:Chitosan based dual microspheres are expected to achieve multiple functions in drug delivery area. Herein, the dual microspheres based on thiolated chitosan (CS-TBA)/CaCO3 were fabricated by ionic gelation method. The obtained dual microspheres combine the benefits of CS-TBA for improved water solubility and CaCO3 for mesoporous structure. The microspheres display a sphere morphology with a diameter of 400–500 μm, in which CaCO3 particles with mesoporous structure are encapsulated. In vitro dual drug release shows a controlled manner for different drugs from CS-TBA/CaCO3 microspheres at different pH. The dual CS-TBA/CaCO3 microspheres can serve as a dual-drug delivery system and have potential applications in tissue engineering and regenerative medicine.

Abstract:We report a facile and green chemical solution approach to synthesize monodisperse alpha-calcium sulfate hemihydrate (alpha-HH) nanoellipsoids with a length of 600 nm and a width of 300 nm by simply mixing Ca2+ and SO42- glycerol-water precursor solutions in the presence of Na(2)EDTA. The alpha-HH nanoellipsoid is formed through a Na(2)EDTA-mediated self-assembly of small primary building blocks (alpha-HH domains: similar to 14 nm). The study on the morphological evolution of alpha-HH reveals that the controlled synergy of supersaturation (precursor concentration) and Na(2)EDTA is crucial for the development of alpha-HH into nanoellipsoids. Further thermal annealing of the nanoellipsoid could make the alpha-HH domains transit into calcium sulfate anhydrites and grow up, generating the gaps between them and resulting in a porous structure. This work paves a new way for preparing high-quality alpha-HH nanoellipsoids with a monodisperse nanosize and a porous structure, promising their future application in many fields such as biomedicine.
