Abstract:We report a Gauss-Bessel STED microscope for super-resolution deep imaging. Improved lateral resolution at 95μm-depth in agarose phantom of brain tissue is about 110nm, which is nearly 2-fold improvement on resolution comparing to conventional STED imaging.

Abstract:We design and build a stimulated emission depletion (STED) microscopy which is capable of super-resolution imaging in two colors. The enhancement in the lateral resolution is demonstrated experimentally through fluorescent beads and biological samples. Abstract:Stimulated emission depletion (STED) microscopy has become a powerful imaging and localized excitation method, breaking the diffraction barrier for improved spatial resolution in cellular imaging, lithography, etc. Because of specimen-induced aberrations and scattering distortion, it is a great challenge for STED to maintain consistent lateral resolution deep inside specimens. Here we report on deep imaging STED microscopy using a Gaussian beam for excitation and a hollow Bessel beam for depletion (GB-STED). The proposed scheme shows an improved imaging depth of up to about 155 mu m in a solid agarose sample, 115 mu m in polydimethylsiloxane, and 100 mu m in a phantom of gray matter in brain tissue with consistent super resolution, while standard STED microscopy shows a significantly reduced lateral resolution at the same imaging depth. The results indicate the excellent imaging penetration capability of GB-STED, paving the way for deep tissue super-resolution imaging and three-dimensional precise laser fabrication.

Abstract:Here, we propose a novel imaging method, which is called image scanning difference microscopy (ISDM), for superresolution imaging. In ISDM, we implement a detector array composed of 19 avalanche photodiodes (APD) rather than single‐point detector in standard confocal microscopy for reconstructing superresolved images with higher signal‐to‐noise ratio (SNR). Combining with our former proposed fluorescence emission difference (FED) method, we have achieved a lateral resolution of 111 nm (∼λ/6) without the damage of image quality, the highest FED resolution to the best of our knowledge. With its simple setup and remarkable performance, we believe that ISDM can become a versatile observation tool in biology and other fundamental studies.

Abstract:Stimulated emission depletion microscopy (STED) has been proved to be a feasible and straightforward method of breaking the conventional diffraction barrier in the far-field. In this paper, we design and setup a home-built high speed STED microscope. Both the excitation beam (488nm) and the depletion beam (592nm) are provided by continuous wave lasers. By using a pair of galvo mirrors (TILL Yanus IV Digital Scan Head), we realize a pixel dwell time down to 2 μs in the experiment which enables an acquisition speed of 2 frames per second in an imaging field of 5*5 μm with an individual pixel size of 10nm. The image acquisition process is controlled by the software Imspector. In the manuscript, we give a clear description on how to build the microscope and also conduct several experiments to evaluate its performance in practice. A spatial resolution of <100nm, which is well beyond the diffraction barrier has been demonstrated in both nanoparticles and biomedical samples. Featuring a superresolution together with a high imaging speed, our STED microscope has big potential to be widely applied in related scientific researches.

Abstract:The recently proposed fluorescence emission difference (FED) microscopy has been demonstrated to be capable of breaking the diffraction barrier, which restricts the spatial resolution of far-field fluorescence microscopy. In this paper, we report a novel high-speed FED system that can realize real-time super-resolution imaging. By replacing the conventional nanopositioning stage with a galvo mirror, the temporal resolution of FED is improved to nearly one frame per second, which is 100 times faster than that of best existing FED, while the system maintains a super-high spatial resolution of 150 nm, which is far beyond the diffraction barrier. Therefore, the high-speed FED is suitable for large-area observations while avoiding photobleaching. Detailed theoretical analysis, simulations, and experimental real-time resolution tests on 100 nm nanoparticles and biological cells are reported.

Abstract:We propose a novel fluorescence emission difference microscopy (FED) system based on focusing cylindrical vector beams. In conventional FED, a Gaussian beam and a 0–2π vortex phase plate are used to generate solid and hollow spots. We focus radially polarized and azimuthally polarized cylindrical vector beams to obtain an expanded solid spot and a shrunken hollow spot, taking advantage of the optical properties of cylindrical vector beams to improve the conventional FED performance. Our novel method enhances FED performance because the hollow spot size determines the FED resolution and an expanded solid spot effectively reduces negative side-lobe emergence during image processing. We demonstrate improved performance theoretically and experimentally using an in-house built FED. Our FED achieved resolution of less than λ/4 in test images of 100nm nanoparticles, better than the confocal image resolution by a factor of approximately 1/3. We also discuss detailed simulation analyses and FED imaging of biological cells.

Here we propose and build a novel dual-color FED system that can provide fluorescence imaging with sub-diffraction resolution. The enhancement in the lateral resolution is verified theoretically and experimentally. Compared to standard STED system, FED enjoys the advantages of lower illumination power and higher flexibility. In addition, the dual-color configuration enables the presented system to be suitable for biological investigations.

Abstract:ISM can effectively enhance the signal strength and spatial resolution of traditional confocal microscopy. How ever, its maximum resolution is limited to root 2 times higher than confocal. On the other hand, stimulated emission depletion (STED) microscopy has been proven to be able to greatly improve the spatial resolution of confocal and has been widely used in practical applications. In this article, we present the implementation of STED with array detection and photon reassignment, and for convenient engineering applications, we also implement an integrated and practical array detection and photon reassignment module. We give a brief explanation of theoretical basis of array detection and photon reassignment, and make a comparison of these two methods. In practical applications, we tested the imaging characteristics of the system with a variety of samples. The results show that array detection can effectively improve the imaging signal-to-noise ratio (SNR); meanwhile, photon reassignment can also improve SNR and resolution to some extent under different depletion intensity. We believe that our research will provide effective inspiration and reference for subsequent research and can be widely applied to actual imaging observations.

Abstract:To overcome the diffraction barrier, super-resolution microscopy is contrived and has witnessed scientific developments in varying fields, especially in last few decades, such as stochastic optical reconstruction microscopy, stimulated emission depletion microscopy (STED), mirror-enhanced super-resolution microscopy (MEANS), and fluorescence emission difference microscopy (FED). Recently, saturated competition microscopy (SAC) was developed to realize high sub-diffraction resolution in either fluorescent or non-fluorescent imaging. Compared with STED, SAC features non-constraint in fluorescent dye selection. Nevertheless, the lateral resolution is limited in consideration of photobleaching side effects. Also, the axial resolution enhancement of SAC has not been demonstrated. In this study, a method, combining FED, MEANS, and SAC, is presented to improve the three-dimensional (3D) resolution. The numerical study reveals that the lateral resolution is close to 0.085 lambda. and axial resolution can be enhanced to 0.184 lambda. In addition, the SNR is improved simultaneously. The availability to improve 3D resolution of SAC is believed to be significant for biological imaging in the future. (C) 2020 Optical Society of America

Abstract:A superresolution imaging technique based on wide-field stochastic fluorescent bleaching is studied. A stream of fluorescence images is simply acquired, and single fluorophore is detected by subtracting each frame from the previous one. After image subtractions, a superresolution image is got by fitting images of single fluorescent molecules with a point dispersion function method to localize them. Then this method is used to image the human embryonic kidney cells with the conventional fluorescent probe Alex 488. Compared with the conventional fluorescence microscope, the resolution increases about 40%, showing that it works with the sample with conventional fluorescent probes and can break the diffraction limit. Also it overcomes the weakness of conventional techniques based on the precise localization of single molecules, which needs to activate and quench the fluorescent molecules repeatedly, and can not be applied to conventional fluorescent samples.

Abstract:Super-resolution microscopy techniques are versatile and powerful tools for visualizing organelle structures, interactions, and protein functions in biomedical research, and its resolution ability to break the optical diffraction limit provides new analytical frameworks for cell biology on the nanoscale, which is indispensable to life science related fields. However, due to the effect of the diffraction limit, the axial resolution of a super-resolution microscope is more arduous to improve than the lateral resolution, which hinders the realization of sub-hundred-nanometer resolution three-dimensional imaging of cellular structures. Therefore, based on the two main techniques, stimulated emission depletion microscopy and single-molecule localization microscopy, the present paper introduces the principles and characteristics of a variety of existing three-dimensional imaging techniques, and finally discusses the future of that. Finally, we briefly discuss the research trend of the two techniques in the three-dimensional imaging area.

Abstract:A parallel scanning method using optical lattices is proposed theoretically to improve the imaging speed of fluorescence emission difference microscopy (FED), which gives the wide field imaging capability to FED while maintaining all the basic advantages of single point FED. The basic principle of wide field FED (wfFED) is presented briefly and the method of generating optical lattices is discussed. The resolution via two types of optical lattices pattern scanning is also studied. With optical lattices scanning, which is generated by two orthogonally crossed standing waves, the wfFED can be implemented without wide field excitation. This strategy can further improve the wfFED imaging speed and simplify the set-up.

Abstract:We report on the design and implementation of an integrated dual-color stimulated emission depletion (STED) microscopy and fluorescence emission difference (FED) microscopy. This highly integrated system combines the respective advantages of STED and FED microscopies, enabling acquisition of super-resolution STED images with specific fluorophores and high laser power, or resolution-enhanced FED imaging for ordinary fluorescent dyes and other materials with moderate demand for laser power. The functional integration enables researchers to select the more appropriate microscopy according to practical scenario and requirements, giving full play to their respective strengths and satisfying the imaging demands maximally. The system's resolution is characterized by dual-color imaging of fluorescent particles. Its practical applicability is verified through imaging various kinds of samples, including several fixed cells stained with different organic dyes and a given organic material-organometallic halide perovskite.

Abstract:Stimulated emission depletion (STED) microscopy enables the three-dimensional (3D) visualization of dynamic nanoscale structures in living cells, offering unique insights into their organization. However, 3D-STED imaging deep inside biological tissue is obstructed by optical aberrations and light scattering. We present a STED system that overcomes these challenges. Through the combination of two-photon excitation, adaptive optics, red-emitting organic dyes, and a long-working-distance water-immersion objective lens, our system achieves aberration-corrected 3D super-resolution imaging, which we demonstrate 164ɻm deep in fixed mouse brain tissue and 76ɻm deep in the brain of a living mouse. Abstract:We propose an approach that uses a conventional optical microscope to achieve optical super-resolution. This approach will provide a direct, non-invasive, and far-field access in the observation of both metallic and non-metallic objects with a spatial resolution of tens-of-nanometres, achieved in a single snap shot. This method, combines near-field illumination by the micro-fiber and a passive spatial frequency shift by the sample itself, offering a promising approach to surface tomography imaging without localized field enhancement by surface plasmon polaritons. Our results present the potential of breaking the diffractive barrier using a conventional wide-field optical microscope within the visible spectrum.

Abstract:Recent advances in far-field fluorescence microscopy have led to substantial improvements in image resolution, achieving a near-molecular resolution of 20 to 30 nanometers in the two lateral dimensions. Three-dimensional (3D) nanoscale-resolution imaging, however, remains a challenge. We demonstrated 3D stochastic optical reconstruction microscopy (STORM) by using optical astigmatism to determine both axial and lateral positions of individual fluorophores with nanometer accuracy. Iterative, stochastic activation of photoswitchable probes enables high-precision 3D localization of each probe, and thus the construction of a 3D image, without scanning the sample. Using this approach, we achieved an image resolution of 20 to 30 nanometers in the lateral dimensions and 50 to 60 nanometers in the axial dimension. This development allowed us to resolve the 3D morphology of nanoscopic cellular structures.
