Study on the Mechanism of Anti-Fas Antibody in Mice with Viral Myocarditis

Abstract:Objective To investigate the effect and mechanism of anti-Fas antibody in experimental coxsackievirus B3 myocarditis. Methods A total of 80 BALB/c mice were used in the experiments, which were divided into normal control group(G1), CVB3 control group(G2), IgG control group(G3) and anti-Fas antibody therapy group(G4) randomly. The mice in G2, G3, G4 were inoculated with 0.15 ml TCID_ 50 coxsackie virus B3(CVB3) and the mice in G1 with 0.15 ml Eagle. The mice in G3, G4 were inoculate with IgG(0.1 mg/kg) and anti-Fas antibody (0.1 mg/kg) on day 0 and day 3 post inoculation(p.i.), respectively. Eight mice in each group were sacrificed on day 10 p.i. Light microscopy, electronic microscopy and terminal transferase-mediated dUTP-biotin nick end labeling(TUNEL) assay were used to quantify inflammation, necrosis and apoptosis in myocardium. The expression of active caspase-3 in myocardium was determined by immunohistochemistry. Caspase-3 mRNA and CVB3 mRNA were analyzed by reverse-transcription polymerase chain reaction(RT-PCR) and real-time quantitative polymerase chain reaction(RQ-PCR), respectively. Results (1) Caspase-3 activation and apoptosis were seen in the myocardium of mice with myocarditis. The dynamic changes of them all showed significantly positive correlation with the changes of myocardial histopathologic scores(r=0.81, P0.01; r=0.73, P0.05). (2) The myocardial histopathologic scores, average percentages of apoptotic cardiomyocytes, expression of active caspase-3 protein and mRNA and expression of CVB3 mRNA in myocardium of mice in G4, were significantly reduced than that in the myocardium of mice in G2(P0.01, P0.01, P0.01, P0.05, P0.05, respectively) and G3(P0.01, P0.01, P0.05, P0.05, P0.05, respectively). Conclusion Myocyte apoptosis is a key mechanism of myocardial damage in viral myocarditis. Anti-Fas antibody can reduce expression of active caspase-3 protein and mRNA, viral replication, cardiomyocyte apoptosis and myocardial injury in the experimental CVB3 myocarditis.
