Study on Regulation of Th Cell Apoptosis in the Acute and Recovery Stage of Viral Myocarditis and Interference with QingXin Ⅱ Decoction

Abstract:Objective:To investigate partly regulating mechanisms of Th cell apoptosis initially,and observe the interference effect of Qing-xin Ⅱ decoction on them.Methods:Make models of VMC mice in acute stage and recovery stage with the methods of injecting coxsackie B3 virus intraperitoneally.Each stage contains three groups:normal group,model group and treatment group.The mice in treated groups were fed with water-solution of Qing-xin Ⅱ decoction,mice in normal group and model group were given the same volume of distilled water,course of treatment was 10 days and 14 days respectively.Then killed all mice,and gathered their spleen cells,after course of treatment was complished.Examined the apoptosis of Th cells by Annexin v/PI double stain flow cytometry and transmisson electron microscope,expression of caspase-3,caspase-9,NF-κB were detected by western blot.Results:(1) Apoptosis rates of spleen Th cells of mice in acute or recovery stage VMC are higher than those in normal groups(P<0.01);compared with model groups,in both stage apoptosis rates of Th cells of mice in treated groups came down significantly(P<0.01,P<0.05).Both in two stages,there were no statistical differences between treated groupe and model group(P>0.05)(2) In both stage,compared with normal group,the expression of caspase-3,caspase-9,NF-κB on Th cell of the mice went up significantly.Compared with model group,the expression of caspase-3,caspase-9,NF-κB in treated group went down significantly.Conclusion:The apoptosis of Th cells and the expression of caspase-3,caspase-9,NF-κB in model group were higher than those in normal group.The above revealed that there were over-apoptosis of Th cells whatever in acute stage or recovery stage of VMC.Qing-xin Ⅱ decoction could play an active role in educing the over-apoptosis of Th cells.
