Change and Significance of Expression of N-cadherin in Myocardium of Mice with Acute and Chronic Viral Myocarditis

Abstract:Objective To observe and analyze the change of the expression of N-cadherin in the myocardium of the different phase of viral myocarditis.Methods We established experimental model of VMC and DCM by repetitive coxsickievirus B3(CVB3) infection in Balb/c mice.Then we evaluated cardiac function by means of ultrasonic inspection,examined myocardium histopathology on H-E stain sections and detected the changes of myocardium N-cadherin expression by means of immunohistochemistry,Western-blotting and RT-PCR.at different time-points of the 10th day and the 5th month after the first infection.Results At the 10th day,obvious inflammatory changes were observed in myocardium of experimental VMC mice.The cardiac function of experimental mice was depressed.However,no obvious differences of N-cadherin expression were observed between the VMC and control groups.At the 5th month,histopathological changes were characterized by fibrosis,derangement and loss of myocardium in myocardium tissue of experimental mice.The size of cardiac chambers was enlarged and ventricular wall was thinner in experimental group.The cardiac function of experimental mice was significantly depressed and the N-cadherin expression in myocardium of experimental group down-regulated.Conclusion Increasing N-cadherin expression may be involved in the mechanisms of impaired cardiac function in DCM mice.
