Abstract:Three alkaloids, Evodiamine, Rutaecarpine and Dehydroevodiamine, were isolated fromthe fruits of Evndia rutaecarpa (Juss) var. bodinaieri (Dode) Huang. In this paper, all ofthe iH and 13C NMR chemical shifts of these compounds were assigned through iD NMRand 2D NMR with Pulse Field Gradient (PFG) techniques (gCOSY, gNOESY, gHMQC andgHMBC).

Abstract:Aim To investigate the NMR spectroscopy of amlodipine and risperidone.Methods 1D NMR and 2D NMR experimental techniques of gCOSY, gHSQC and gHMBC were wsed.Results The assignments of the 1H and 13C NMR data for the two drugs were performed and confirmed by the evidence of JHF and JCF.Conclusion The structures of amlodipine and risperidone were confirmed by careful analysis of regular 1D and 2D NMR spectroscopy.

Abstract:High-resolution magic-angle-spinning(MAS) H-1 NMR spectroscopic and pattern recognition(PR) based methods were applied to compare the different acute biochemical effects between La (NO3)(3)- and Ce (NO3)(3)-treated rats. Male Wistar rats were treated with various doses(2, 10, 50 mg/kg body weight) of La(NO3)(3) and Ce(NO3)(3), and MAS H-1 NMR spectra of the intact liver and kidney tissues were analyzed by using principal components analysis to extract metabolic information. The target lesion of La (NO3)(3) to liver and Ce(NO3)(3) to both liver and kidney were found by MAS H-1 NMR-PR methods. This work illustrated that the combination of NMR and pattern recognition technique is a powerful method to study the biochemical effects induced by xenobiotics.

Abstract:When being put into CDCl3 (slightly acidic condition), the double bond of steroid drug (LIVIAL) will rearrange 24 hours. This paper shows that the 2D NMR techniques were successfully performed to determine the structures of compound 1 and compound 2. All of the 1H and 13C NNR chemical shifts were assigned by 1D and 2D NMR. Their stereochemistry was discussed, too. The structures of compound 1 and compound 2 were simulated by computer molecular modeling. Their results agree with those of 2D NMR.

Abstract:Aim To investigate the structure of mogroside Iva isolated from traditional Chinese medicine fructus mo-mordicae [ fruits of Siraitia grosvenori (Swingle) C. Jeffery] and summarize the NMR characteristics of the structure. Meth-ods Common extraction, separation and purification methods were used. Various NMR techniques including 1 H NMR,13 CNMR, DEPT, 1H-1H COSY, HSQC, HMBC, NOESY and molecular model simulated by computer were used to elucidatethe structure. Results 1 H and 13 C NMR signals of mogroside Ⅳa were assigned, and spectroscopic basis was obtained for identification of such type of compounds. Conclusion 1D and 2D NMR techniques including 1 H-1 H COSY, HSQC, HM-BC, NOESY spectra are powerful tools for structure analysis. The structure determined by NMR methods is identical with energy minimized conformation simulated by computer.

Abstract:A combination of 1D and 2D NMR techniques, including HMQC, HSQC, 1H-1H COSY and HMBC, was used to provide completely assigned 1H and 13C NMR data for the structures of three 3-[substituted methylidene]-1H,3H-naphtho[1,8-cd]-pyran-1-ones—3-[(4-methoxyphenyl)methylidene]-1H,3H-naphtho[1,8-cd]-pyran-1-one, 3-[(4-fluorophenyl)methylidene]-1H,3H-naphtho-[1,8-cd]-pyran-1-one and 3-[(thiophen-3-yl)methylidene]-1H,3H-naphtho[1,8-cd]-pyran-1-one. The heteronuclear coupling constants 1–4JCF and 3–4JHF were correspondingly determined. Additionally, IR and Raman spectral data were provided in support of the assigned structures.

Abstract:Molecular dynamics (MD) simulations and chemical shifts from machine learning are used to predict 15 N, 13 C and 1 H chemical shifts for the amorphous form of the drug irbesartan. The molecules are observed to be highly dynamic well below the glass transition, and averaging over this dynamics is essential to understanding the observed NMR shifts. Predicted linewidths are consistently about 2 ppm narrower than observed experimentally, which is hypothesised to result from susceptibility effects. Previously observed differences in the 13 C shifts associated with the two tetrazole tautomers can be rationalised in terms of differing conformational dynamics associated with the presence of intramolecular interaction in one tautomer. 1 H shifts associated with hydrogen bonding can also be rationalised in terms of differing average frequencies of transient hydrogen bonding interactions.

Abstract:By using density functional theory (DFT) calculations, we refined the H atom positions in the structures of β-caffeine (C), α-oxalic acid (OA; (COOH)2), α-(COOH)2⋅2H2O, β-malonic acid (MA), β-glutaric acid (GA), and I-maleic acid (ME), along with their corresponding cocrystals of 2:1 (2C--OA, 2C--MA) or 1:1 (C--GA, C--ME) stoichiometry. The corresponding 13C/1H chemical shifts obtained by gauge including projector augmented wave (GIPAW) calculations agreed overall very well with results from magic-angle-spinning (MAS) nuclear magnetic resonance (NMR) spectroscopy experiments. Chemical-shift/structure trends of the precursors and cocrystals were examined, where good linear correlations resulted for all COO1H sites against the H⋯O and/or H⋯N H-bond distance, whereas a general correlation was neither found for the aliphatic/caffeine-stemming 1H sites nor any 13C chemical shift against either the intermolecular hydrogen- or tetrel-bond distance, except for the 13COOH sites of the 2C--OA, 2C--MA, and C--GA cocrystals, which are involved in a strong COOH⋯N bond with caffeine that is responsible for the main supramolecular stabilization of the cocrystal. We provide the first complete 13C NMR spectral assignment of the structurally disordered anhydrous β-caffeine polymorph. The results are discussed in relation to previous literature on the disordered α-caffeine polymorph and the ordered hydrated counterpart, along with recommendations for NMR experimentation that will secure sufficient 13C signal-resolution for reliable resonance/site assignments.

Abstract:NMR experiments were used to identify the two main conformations of Larotrectinib and its synthetic precursor, the nitro compound 4. Conformational analysis by dynamic NMR was performed in different temperature and solvents, combining 2D EXSY and NOE experiments. The results revealed the conformations of Larotrectinib and compound 4 were caused by the barrier to rotation of C11–N17. Meanwhile, the conformational specific structures of compound 4 were postulated based on NOE data and molecular simulation. The complete assignments of 1H and 13C NMR data for compounds 1, 4 and 5 were reported for the first time.

Abstract:Proton NMR signals of the phenolic hydroxyl groups of the substituted catechins were successfully collected. The identities of these signals were elucidated and confirmed by a combination of computational and experimental investigations including GIAO-B3LYP NMR chemical shift calculation, kinetic analysis of acylation, and NOESY correlation. Furthermore, the reliability of chemical shifts of hydrogens of phenolic hydroxyl groups was confirmed by evaluation of the influence of acidic, basic and water residues. Finally, the chemical shift profiles of phenolic hydroxyl groups were developed for instant structural recognition. This study demonstrated the feasibility of application of chemical shifts of hydrogens of phenolic hydroxyl groups as fingerprint for structural determination and the concept could be extended to other polyphenols.

Abstract:The structural elucidation of chiral molecules with more than one stereocenter is usually a tricky problem. In this paper, efficient 1H NMR spectroscopic approaches for assigning the erythro and threo configurations of 1-oxygenated 1,2-diarylpropan-3-ols were developed. By analysis of the chemical shift differences of diastereotopic methylene H2-3 (Δδ3) in CDCl3 or the chemical shift differences of H-1 and H-2 (Δδ1,2) in methanol-d4, deuterated dimethyl sulfoxide, and acetone-d6, the configurations of 1-oxygenated 1,2-diarylpropan-3-ols can be rapidly and conveniently determined.

Abstract:Mogroside Ⅲ was isolated from the fruit of Momordica grosvenori Swingle [Siraitia grosvenorii (Swingle) C. Jeffery]. Mogrol (9β-methyl-19-norlanost-5-ene-3β, 11α, 24β, 25-tetrol) was derived from mogroside Ⅲ by mild enzymatic hydrolysis. One-dimensional and two-dimensional NMR (including gCOSY, gNOESY, gHSQC and gHMBC) techniques were employed to assign the chemical shifts and elucidate the structures of the compounds. The effects of the substitutent groups on the 1H and 13C chemical shifts of the mogroside were also discussed.

Abstract:The infrared spectrum, ultraviolet spectrum, mass spectra, and nuclear magnetic resonance spectra of metoprolol tartrate, an antiarrhythmic medicine, were reported and interpreted comprehensively. The vibrations of functional groups in infrared spectrum, electron transitions in ultraviolet spectrum, and main characteristic fragmentations in mass spectra of metoprolol tartrate were discussed. All the (1)H and (13)C NMR chemical shifts were assigned by means of distoritionless enhancement by polarization transfer-135 and -90, (1)H-(1)H correlation spectroscopy, and (13)C-(1)H correlation spectroscopy via short- and long-range coupling. Abstract:A series of 1,3-oxazino［3,2-d］［1,5］ benzodiazepine-1-one derivatives' stereochemistry was studied by 1H NMR. During the 1H NMR experiments, we found out their unique stereochemistry of seven-membered rings. All the discernible hydrogen's chemical shifts were assigned. Their distorted boat conformations were discovered by the related coupling constants and gNOESY results, which were compared with x-ray diffraction result and the conformation of analogous benzothioazepines.

Abstract:Delineation of protein-ligand interaction modes is key for rational drug discovery. The availability of complex crystal structures is often limited by the aqueous solubility of the compounds, while lead-like compounds with micromolar affinities normally fall into the NMR intermediate exchange regime, in which severe line broadening to beyond the detection of interfacial resonances limits NMR applications. Here, we developed a new method to retrieve low-populated bound-state 1H pseudocontact shifts (PCSs) using paramagnetic relaxation dispersion (RD). We evaluated using a 1H PCS-RD approach in a BRM bromodomain lead-like inhibitor to filter molecular docking poses using multiple intermolecular structural restraints. Considering the universal presence of proton atoms in druglike compounds, our work will have wide application in structure-guided drug discovery even under an extreme condition of NMR intermediate exchange and low aqueous solubility of ligands.
