Abstract:A general procedure to determine the absolute configuration of cyclic secondary amines with Mosher's NMR method is demonstrated, with assignment of absolute configuration of isoanabasine as an example. Each Mosher amide can adopt two stable conformations (named rotamers) caused by hindered rotation around amide C--N bond. Via a three-step structural analysis of four rotamers, the absolute configuration of (-)-isoanabasine is deduced to be (R) on the basis of Newman projections, which makes it easy to understand and clarify the application of Mosher's method to cyclic secondary amines. Furthermore, it was observed that there was an unexpected ratio of rotamers of Mosher amide derived from (R)-isoanabasine and (R)-Mosher acid. This phenomenon implied that it is necessary to distinguish the predominant rotamer from the minor one prior to determining the absolute configuration while using this technique.

Abstract:Aim To investigate the structure of mogroside Iva isolated from traditional Chinese medicine fructus mo-mordicae [ fruits of Siraitia grosvenori (Swingle) C. Jeffery] and summarize the NMR characteristics of the structure. Meth-ods Common extraction, separation and purification methods were used. Various NMR techniques including 1 H NMR,13 CNMR, DEPT, 1H-1H COSY, HSQC, HMBC, NOESY and molecular model simulated by computer were used to elucidatethe structure. Results 1 H and 13 C NMR signals of mogroside Ⅳa were assigned, and spectroscopic basis was obtained for identification of such type of compounds. Conclusion 1D and 2D NMR techniques including 1 H-1 H COSY, HSQC, HM-BC, NOESY spectra are powerful tools for structure analysis. The structure determined by NMR methods is identical with energy minimized conformation simulated by computer.

Abstract:As a salt-type compound, mosapride citrate's metabolism and side effects are correlated with its salt-forming ratio. Several techniques were developed in this work to compare various quantitative nuclear magnetic resonance (qNMR) methodologies and to quantitatively examine the content of raw materials. Among the qNMR techniques, methods for 1H NMR and 19F NMR were developed. Appropriate solvents were chosen, and temperature, number of scans, acquisition time, and relaxation delay parameter settings were optimized. Maleic acid was chosen as the internal standard in 1H NMR, and the respective characteristic signals of mosapride and citrate were selected as quantitative peaks. The internal standard in 19F NMR analysis was 4,4′-difluoro diphenylmethanone, and the distinctive signal peak at −116.15 ppm was utilized to quantify mosapride citrate. The precision, repeatability, linearity, stability, accuracy, and robustness of the qNMR methods were all validated according to the ICH guidelines. By contrasting the outcomes with those from high-performance liquid chromatography (HPLC), the accuracy of qNMR was assessed. As a result, we created a quicker and easier qNMR approach to measure the amount of mosapride citrate and evaluated several qNMR techniques to establish a foundation for choosing quantitative peaks for the qNMR method. Concurrently, it is anticipated that various selections of distinct quantitative objects will yield the mosapride citrate salt-forming ratio.

Abstract:In this study,NMR techniques,including 1H NMR,13C NMR,DEPT,19F NMR,HSQC,and HMBC,were used in combination to assign the 1H and 13C chemical shifts of lansoprazole in room temperature.On the basis of the assignments and the results of variable-temperature experiments,a hypothesis was put forward to explain the existence of several low broad lines in the 1H and 13C NMR spectra of the compound.It was proposed that there exist two inter-converting tautomers at room temperature.The inter-conversion rate increased and the broad lines became narrower as the temperature increased,and vice versa.The low broad lines in the 1H spectra of the compound disappeared at-70 ℃,indicating the existence of only one tautomer and the termination of the inter-conversion process.The 1H spectra of lansoprazole obtained at-70 ℃ were assigned,which were shown to be consistent with the results obtained at in room temperature.

Abstract:A combination of 1D and 2D NMR techniques, including HMQC, HSQC, 1H-1H COSY and HMBC, was used to provide completely assigned 1H and 13C NMR data for the structures of three 3-[substituted methylidene]-1H,3H-naphtho[1,8-cd]-pyran-1-ones—3-[(4-methoxyphenyl)methylidene]-1H,3H-naphtho[1,8-cd]-pyran-1-one, 3-[(4-fluorophenyl)methylidene]-1H,3H-naphtho-[1,8-cd]-pyran-1-one and 3-[(thiophen-3-yl)methylidene]-1H,3H-naphtho[1,8-cd]-pyran-1-one. The heteronuclear coupling constants 1–4JCF and 3–4JHF were correspondingly determined. Additionally, IR and Raman spectral data were provided in support of the assigned structures.

Abstract:The previously unindexed laboratory X-ray powder diffraction data of mosapride dihydrogen citrate dihydrate, an API used to stimulate gastrointestinal motility, has been recorded at room temperature. Using these data, the crystal structure of this API has been refined in space group P2 1 /c (No. 14) with a = 18.707(4) Å, b = 9.6187(1) Å, c = 18.2176(4) Å, β = 114.164(1)°, V = 2990.74(8) Å 3 , and Z = 4. The structure of this material corresponds to the phase associated with CSD Refcode LUWPOL determined at 93 K. The Rietveld refinement, carried out with TOPAS-Academic, proved the single nature of the sample and the quality of the data recorded.

Abstract:N-acylhydrazone compounds have two isomers, E/cis and E/trans, in polar solvents. In this work, 1H and 13C nuclear magnetic resonance (NMR) signals, coupling constants (J), and the spatial structures of the two isomers (E/cis and E/trans) of a novel carbazole-triazinoindole based N-acylhydrazone derivative 3, 2-(5H-[1,2,4] triazine[5,6-b]indol-3-ylthio)-N'-(9-ethylcarbazol-3-ylmethylene)acetylhydrazine, were obtained by one-dimensional and two-dimensional NMR techniques, infrared spectroscopy (IR) and elemental analysis. The ratios of the two isomers were determined.

Abstract:The H-1 and C-13 NMR chemical shifts of the aldose reductase inhibitor 4(S)-2,3-dihydro-6-fluoro-2(R)-methylspiro[chroman-4,4'-imidazolinel-2',5'-dione,methylsorbinil, and its seven synthetic intermediates, have been completely assigned on the basis of DEPT, COSY, g-HSQC and g-HMBC. All C-F coupling constants from one-bond to four-bond in the C-13 NMR spectra and H-F and H-H coupling constants from three-bond to four-bond in H-1 spectra were obtained. Abstract:Alprazolam is an antidepressant drug used extensively in clinic. In this study,1H and 13C chemical shifts of the compound were completely assigned using 2D NMR techniques,including DEPT,1H-1H COSY,13C-1H HSQC and 13C-1H HMBC. Both NMR data and theoretical calculation showed that the methylene protons in the compound are magnetically non-equivalent. The infrared spectra and TOF-MS/MS spectra of alprazolam were also recorded.

Abstract:A new compound,N-phenyl-2-({5-[(2-(phenoxymethyl)-benzimidazol-1-yl)methyl]-1,3,4-oxadiazol-2-yl}thio) acetamide(3),was synthesized by the reaction of 5-{[2-(phenoxymethyl)-benzimidazol-1-yl]methyl}-2-thiol-1,3,4-oxadiazole(2) with N-phenyl-2-chloro-acetamide.The compound 2 was obtained by the reaction of the hydrazide(1) with carbon disulfide in the presence of absolute ethanol and potassium hydroxide.The structure of the new compound 3 was determined by elemental analysis,IR and NMR techniques.The 1H and 13C NMR signals of the two isomers(trans and cis) of compound 3 were assigned using 2D NMR techniques.The corresponding coupling constants and the tautomeric ratios of isomers were calculated.The ratio of the dominant trans isomer was 64.5%,while the cis isomer was 35.5%.

Abstract:The spectral data of peptidomimetics cyclo[-RGD-ψ(triazole)-GD-] were reported, including UV, FT-IR and NMR (i.e.,1H NMR,1H-1H COSY,13C NMR, DEPT, 1H-13C HSQC and1H-13C HMBC). The NMR signals were assigned. The infrared spectrum was analyzed to derive the types of vibrations of the functional groups in the compound. The molecular structure of peptidomimetics cyclo[-RGD-ψ(triazole)-GD-] was determined.

Abstract:NMR experiments were used to identify the two main conformations of Larotrectinib and its synthetic precursor, the nitro compound 4. Conformational analysis by dynamic NMR was performed in different temperature and solvents, combining 2D EXSY and NOE experiments. The results revealed the conformations of Larotrectinib and compound 4 were caused by the barrier to rotation of C11–N17. Meanwhile, the conformational specific structures of compound 4 were postulated based on NOE data and molecular simulation. The complete assignments of 1H and 13C NMR data for compounds 1, 4 and 5 were reported for the first time.

Abstract:The structural elucidation of chiral molecules with more than one stereocenter is usually a tricky problem. In this paper, efficient 1H NMR spectroscopic approaches for assigning the erythro and threo configurations of 1-oxygenated 1,2-diarylpropan-3-ols were developed. By analysis of the chemical shift differences of diastereotopic methylene H2-3 (Δδ3) in CDCl3 or the chemical shift differences of H-1 and H-2 (Δδ1,2) in methanol-d4, deuterated dimethyl sulfoxide, and acetone-d6, the configurations of 1-oxygenated 1,2-diarylpropan-3-ols can be rapidly and conveniently determined.

Abstract:One new compound 6-demethylvermistatin (1), together with two known compounds, the penicillide derivatives (2) and (3) were isolated from the mangrove fungus Guignardia sp. No. 4382 obtained from the South China Sea. Their structures were assigned using high-resolution electron ionization mass spectrometry(HREIMS), H-1 and C-13 NMR spectra, DEPT, and by 2D COSY, HMQC, and HMBC experiments. Abstract:Object: To identify the structure of the new agent nicergoline . Methods: The structure of nicergoline was estublished EA,UV, IR, MS, 1H NMR, 13C NMR and SR. Conclusions: In this paper all the 1H NMR and 13C NMR signals of the nicergoline were assigned unequivocally for the first time by means of two-dimensional NMR techniques (COSY, HMQC andHMBC). Results: The structure of nicergoline was identified by intensive spectral studies.

Abstract:Anew natural product, named phomopsin A, 1-(meta-hydroxyphenyl)-4-hydroxy-3-isoquinolone (1), together with two known compounds cytochalasin H (2) and glucosylceramide (3), was isolated from the mangrove endophytic fungus Phomopsis sp. (ZZF08) obtained from the South China Sea coast. The structures were elucidated by 1D and 2D NMR experiments including COSY, HMQC, and HMBC. According to NMR and single-crystal X-ray diffraction, it was found that some assignments about H-1 and C-13 NMR data for cytochalasin H (2) were probably uncorrected in the previous reports. In our cytotoxicity assays, compound 1 showed moderate cytotoxicity toward KB cells with IC50 at 28.0 mu g ml(-1) and KBv200 cells with IC50 at 16.8 mu g ml(-1), and compound 2 exhibited strong cytotoxicity toward KB cells and KBv200 cells with IC50 less than 1.25 mu g ml(-1). Abstract:1H and 13C NMR spectra of p-nitrophenyl azobenzene derivatives were completely assigned by using ID 1H and 13C methods, which were further confirmed by 2D 1H-1H COSY, 13C-1H HETCOR and 13C-1H long-range HETCOR methods. These results would be a foundation for NMR studies of the liquid crystals containing such side-chains.
