Abstract:DNA chip is a new technique in molecular biology. It can be fabricated on silica wafer,glass slide and nylon membrane by high throughput printing or combining the light-directed chemical synthesis. The fluorescent dye molecule can be incorporated in the DNA sample by polymerase chain reaction or other alternative methods.After hybridization of the DNA chip with the labeled sample targets,high-throughput gene sequence and expression information can be retrieved by scanning of the DNA microarray and the analysis of computer .This technology has been applied to sequence DNA,monitoring expression of gene.It has developed into screening gene mutation and polymorphism,and discovered new gene etc.

Abstract:The Human Genome Project has promoted the development of nucleic acid sequence analysis technology.Solid phase synthesis chemistry, photolabile protecting groups and photolithography scanning are integrated to fabricate DNA chip. It is a powerful tool for the rapid screening nucleic acid molecular. The technique can be widely applied to gene polymorphic analysis, detecting mutation, expression monitoring and DNA molecular recognition.The basic principle of fabricating DNA chip was described and the applications in nucleic acid study were reviwed in this paper.

Abstract:The synthesis method of DNA chip is introduced:after substra te been active treated,Light-directed oligonucleotide synthesis or m icroarray printing technique is adopted to synthesize oligonucleotide probe on the surface of substrate,or print oligonucleotide made by off -chip method such as polymerase chain reaction and biochemical synthes is onto the surface of substrate.The amplified and labeled samples are then hybridized with the probes,and the signals should be collected a nd analyzed.The applications in fields such as genome research,gene ex pression monitoring,gene mutation analyses and medicine research are d iscussed.Improvements need to be done and other several realms of DNA chip＇s application are talked too.

Abstract:Based on the nucleotide molecular hybridization technique,Genechips are fabricated either by in situ light-directed chemical synthesis or by automatically spotting after conventional liquid synthe-sis , for binding plenty of oligonucleotide probes with the surface of solid support. Fluorescent or isotope labbeled sample are hybridizated to the chips, and the biological information is achieved by detected and analyzed the to monitoring the expression of genes, searching for gene hybridization signal by computer system. This technology has been successfully applied in Human Genome Project , the study of modern medicine ,the research of animal and plant science and environmental science.

Abstract:The basic principle, technological procedure and types of gene chips were introduced in the article. The probe choice in practical application and processing and hybridization of detected samples were described. Some researchers have used gene chips in hematology, oncology and cellular differentiation. Gene chips can be used to detect the expression of oncogenes, tumor suppression genes, and cell differentiation- and apoptosis-related genes. Abstract:Bioinformatics and gene chip are two new technologies in thefield of Life Science.Bioinformatics is very important to the research and the application of gene chip.It plays a key role in gene chip design,experiment data analysis and data management.The applications of Bioinformatics to gene chip are discussed in this paper in great detail.Some rules for designing gene chip are proposed.A new approach for gene chip design,by which an optimal set of oligonucleotide probes can be generated using dynamic programming,is also proposed.The probes have nearly the same melting temperature Tm,and are of variable length and variable overlap.The method is applied to designing gene chips for DNA resequencing,mutational analysis and gene expression analysis.Two software systems,one for gene chip design and one for data analysis have been developed.

Abstract:The perparation of biochip,detection methods of the hybridization signal and biomedical applications are presented.There are two kinds of preparation methods:one is directly synthesis oligonucleotides on the chip array;another is that using array pin or jet printer method to place the solution of DNA in μL volume in array and fix them on the carrier of biochip.The character of biochip is determined by detection of hybridization signal.For the detection of hybridization signal of fluorescence marked DNA,the following methods can be used:laser fluorescence scan microscopy,laser scanning confocal microscopy,fluoresce microscope with CCD camera,fiber biosensor,etc.For the biotin Marked DNA,chemical illminance,storage screen of light excited phosphorescent materials,light scattering etc.can be used.DNA chip has great foreground in biomedical applications.

Abstract:The gene chip technigue is a high and new technigue developed in 1990's.It can efficiently and rapidly detect a great many of genetic information at one time,which resolved many problems encountered in conventional methods,and has been successfully applied to almost every aspect of biological research,such as gene expression analysis,mutation detection,polymorphism analysis,gene sequencing and drug screening,etc.As a high throughput technique for gene analysis,gene chip technigue has a tremendous potential in microbe detection and identification.In this paper,the general concept and technigue flow of gene chip technigue were introduced briefly,and its current applications in microbe detection and identification were reviewed.

The essence of biochip technology is a parallel analysis for biology message.It is based on the principle of nucleic molecule hybridization or protein immunochemistry.The occurrence of hybridization or immunoreaction is detected by fluo-labelled technology.After analyzed by special computer program, the needed message can be obtained quickly.This paper summarizes the new advances in biochip technology ,and talk about challenges and trends of this technology.
