Abstract:The most prevalent type of liquid chromatography is ion exchange chromatography. In recent years, the acceptance of ion exchange chromatography has been increased because of its application in agricultural, pharmaceutical, environmental and biotechnology industries. Amino acids are principal components of proteins and their separation from protein hydrolysate or from a mixture of amino acids, is always in focus and ion exchange chromatography finds its application in separation of amino acids. Separation of amino acids by ion exchange chromatography has widespread applications because it has high capacity and it is easy to design. This chapter covers modern applications and basic principles of Ion exchange chromatography in the separation of amino acids.

Abstract:A membrane dialysis method was developed to separate amino acid mixtures. A bipolar membrane electrodialysis process was developed to separate amino acids based on calculated cation and anion concentrations in typical amino acid mixtures. Three-chambered electrodialysis experiments verified the prediction. For amino acids with very large isoelectric point differences, such as glutamic acid and arginine, or with moderate isoelectric point differences, such as glutamic acid and glycine or arginine and glycine, the separation process yields very pure amino acids with more than 96% purity. The current density ranges from 5-10 mA·cm-2. For proline and serine which have a small isoelectric point difference, the separation is difficult to achieve and the current density is below 1 mA·cm-2. Buffering solution is not needed in this process and the separation efficiency can be raised.

Abstract:Based on the steady-state condition of ionization equilibrium of amino acid and charge conservation of the solution distributed between the two sides of membrane, the relationship between the total retention of amino acid and retentions of molecule (zwitterion) of amino acid, and the retention of hydrogen ion and the ionization degree of solute was derived, as well as expression of various ion rejection in the amino acid solution. A phenomenological model for membrane separation of amino acid was established. The calculating curves basically tallied with the experimental curves by using the model, and the mathematical model established could provide a reference to the theoretical investigation of the multiple organic acids retention rules with membrane.

Abstract:A new technique for separation of glutamine from its fermentation broth was developed by using one-step anion exchange, based on the difference of the ionization constants of glutamic acid (Glu) and glutamine (Gln). When the solution of crude glutamine crystal from the fermentation was separated by single anion exchange column, the highest recovery of Gln was 83%, and the removal ratio of Glu reached 90%. These results show that this new process is effective in separation of glutamine from the fermentation broth and reduction of the cost.
Separation of Amino Acids Using Multistage Electrodialysis with Reflux

Abstract:The feasibility of multistage electrodialysis with reflux to separate amino acids with the isoelectric points near to each other,such as serine(Ser) and proline(Pro),was studied.The effects of reflux stages,current intensity and the flow rate of reflux amino acids on the separation were investigated.The results show:(1) increasing reflux stages is beneficial to the separation;(2) with 4 reflux stages,the purity of the obtained Pro product and Ser product is 94.9% and 90.9%,respectively;(3) the purity of Ser product can be improved by increasing of the flow rate of reflux amino acids,while Pro product's purity will be reduced by it;(4) increasing current intensity can enhance the purity of Pro product,whereas reduce Ser product's purity.

Separation and Purification Technology

Abstract:Two-phase electrophoresis, coupling traditional solvent extraction with electrophoresis, is a novel separation technique. Being bio-compatible, aqueous two-phase electrophoresis provides a successful method for separating mixtures of biomolecules. In this work, the separation of amino acids by aqueous two-phase electrophoresis was examined with dextran-polyethylene glycol-water as a working system. The influences of separation time, field strength, initial solute concentration and two-phase contact area were studied in intermittent separation equipment. The experiment results show that aqueous two-phase electrophoresis is an effective separation technique for amino acids. With time, field strength, and contact area increasing, mass fluxes across the interface are increased quickly.

Therefore, scientists have proposed the new concept of proteomics, in which protein separation technology plays a momentous role. It has been diffusely used in the food industry, agricultural biological research, drug development, disease mechanism, plant stress mechanism, and marine environment research. In this paper, combined with the recent research situation, the progress of protein separation technology was reviewed from the aspects of extraction, precipitation, membrane separation, chromatography, electrophoresis, molecular imprinting, microfluidic chip and so on.</p>

Abstract:Based on the 001×7 cation-exchange resin,the separation of L-lysine and L-histidine by different concentration of aqua-ammonia under the same conditions such as(the eluation velocity,surface height,etc) L-lysine and L-histicine was eluted on the cation-exchange resin and then use capillary electrophoresis method to measure the content of the two amino-acid in the eluation;As a result,the aqua-ammonia's optimum concentration of elute is 0.4% to separate the two amino-acid through cation-exchange resin is found in this paper.

However,traditional extraction as well as separation and purification techniques are tedious,inefficient,and the effective components are easily lost. In this work,the recent progresses on the extraction techniques such as ultrasound-assisted extraction,supercritical fluid extraction,accelerated solvent extraction and microwave-assisted extraction,and some separation and purification techniques including macroporous adsorption resins,preparative high-performance liquid chromatography and high-speed counter-current chromatography,were reviewed. Abstract:The polymeric reversed micelles were formed with polyoxyalkylene block copolymers as surfactant and 1-octanol as cosolvent in the polyoxyalkylene block copolymers/1-octanol/p-xylene system. The polyoxyalkylene block copolymers consist of hydrophilic poly(ethylene oxide) (PEO) and hydrophobic poly(propylene oxide) (PPO). Two types of copolymers, PEO–PPO–PEO-type and PPO–PEO–PPO-type triblock copolymers were used. Our present interests are focused on the formation of the reversed polymeric micelles and subsequently separation of amino acids by the polymeric reversed micelle solutions. Extraction equilibrium partition of amino acids between polymeric reversed micelle solutions and water was investigated to study the extraction behaviours. The copolymer type and aqueous phase pH affected the extraction process. A model for solute solubilisation by the polymeric reversed micelles is proposed.

Abstract:Background: Bioactive compounds from various natural sources have been attracting more and more attention, owing to their broad diversity of functionalities and availabilities. However, many of the bioactive compounds often exist at an extremely low concentration in a mixture so that massive harvesting is needed to obtain sufficient amounts for their practical usage. Thus, effective fractionation or separation technologies are essential for the screening and production of the bioactive compound products. The applicatons of conventional processes such as extraction, distillation and lyophilisation, etc. may be tedious, have high energy consumption or cause denature or degradation of the bioactive compounds. Membrane separation processes operate at ambient temperature, without the need for heating and therefore with less energy consumption. The "cold" separation technology also prevents the possible degradation of the bioactive compounds. The separation process is mainly physical and both fractions (permeate and retentate) of the membrane processes may be recovered. Thus, using membrane separation technology is a promising approach to concentrate and separate bioactive compounds. Methods: A comprehensive survey of membrane operations used for the separation of bioactive compounds is conducted. The available and established membrane separation processes are introduced and reviewed. Results: The most frequently used membrane processes are the pressure driven ones, including microfiltration (MF), ultrafiltration (UF) and nanofiltration (NF). They are applied either individually as a single sieve or in combination as an integrated membrane array to meet the different requirements in the separation of bioactive compounds. Other new membrane processes with multiple functions have also been developed and employed for the separation or fractionation of bioactive compounds. The hybrid electrodialysis (ED)-UF membrane process, for example has been used to provide a solution for the separation of biomolecules with similar molecular weights but different surface electrical properties. In contrast, the affinity membrane technology is shown to have the advantages of increasing the separation efficiency at low operational pressures through selectively adsorbing bioactive compounds during the filtration process. Conclusion: Individual membranes or membrane arrays are effectively used to separate bioactive compounds or achieve multiple fractionation of them with different molecule weights or sizes. Pressure driven membrane processes are highly efficient and widely used. Membrane fouling, especially irreversible organic and biological fouling, is the inevitable problem. Multifunctional membranes and affinity membranes provide the possibility of effectively separating bioactive compounds that are similar in sizes but different in other physical and chemical properties. Surface modification methods are of great potential to increase membrane separation efficiency as well as reduce the problem of membrane fouling. Developing membranes and optimizing the operational parameters specifically for the applications of separation of various bioactive compounds should be taken as an important part of ongoing or future membrane research in this field.

Abstract:Abstract Fatty acids (FAs) are a very important group of raw materials for chemical industry, and the technology of separating or purifying the FAs from the reaction product mixture has always been the hotspot of research. Membrane processes for separation of FAs are being increasingly reported. Compared with conventional FAs separation methods, membrane separation has the advantages of low energy consumption, system compactness, high separation efficiency, easy scale-up, high available surface area per unit volume and low working temperatures, thereby attracting considerable attention of many researchers. In this regards, this paper critically reviewed the developments of methods for FAs separation and purification, and the future prospects of coupling membrane technology with hydrolysis for enhanced production of FAs.

Abstract:Bioactive peptide (BP) is short sequence of amino acids, which can play different biological activities depending on its structural characteristics and amino acid sequence. As health promoters, bioactive peptides (BPs) have attracted more and more attention in recent years. Separation and purification methods of BPs such as pressure-driven and electrically-driven membrane processes were reviewed in this paper. Especially, cell configuration and application of electrodialysis with filtration membrane (EDFM), a new emerging electro-driven membrane technology, in separating anionic and cationic BPs were introduced in detail. It is difficult to separate and purify BPs due to their excellent water solubility as well as uncertain composition and structure. Thus, further development in efficient and economical technologies for the separation and purification of BPs is urgent to improve human health.

Abstract:Ion exclusion method was used to purify sodium chloride-containing β-alanine.Different types of strong-acid cation exchange resins and operation conditions were studied.The suitable parameters were:50～100 mesh exchange resin of model 001×8,feeding volume 15%～20% BV(bed volume),flow rate 1/120～1/60 BV/min,concentration of feeding solution:β-alanine 4%～8%,NaCl 7%～14%.
