Abstract:Three new stilbenoids, namely two rare plant-derived phenanthrenes denominated Cajananthrenes A and B ( 1 , 2 ) and one bibenzyl named Cajanbenzyl ( 3 ), together with a diphenyl ether derivative designated Cajanether ( 4 ), as well as five other known compounds ( 5 - 9 ) were isolated from the ethanolic extract of the leaves of Cajanus cajan . Their structures were determined through extensive spectroscopic analysis including UV, IR, NMR (1D and 2D) and HRESIMS as well. A plausible biogenesis pathway was proposed for the biosynthesis of compounds 1 - 3 . Compounds 1 and 2 displayed moderate anti-inflammatory activity as evident from the inhibitory effect on NO production in LPS-stimulated RAW 264.7 macrophages with IC 50 values of 73.6 and 44.6 μM respectively.

Abstract:Four compounds, including one new lignan cavaol H (1), one new coumarinolignoid 5′-hydroxycleomiscosin B (4) and two known lignans (2–3) were isolated from the 95% ethanol extract of the twigs of Eurycorymbus cavaleriei. Their structures were established on the basis of various spectroscopic analyses including 1D-(1H, 13C, and DEPT) and 2D-NMR (COSY, HMQC, and HMBC). The absolute stereochemistry of the two known lignans was firstly reported in this article by 1H NMR studies on Mosher's ester derivatives. In the present study, quinone reductase induction activities of compounds 1–4 were assayed, compound 4 showed moderate quinone reductase induction with concentration to double the enzyme activity (CD) of 10.5 ± 0.8 μg/mL. Then we established LC-MS-MS analysis of glutathione (GSH) incubation with compound 4 to explain whether compound 4 induced quinone reductase through alkylating of the sulfhydryl groups of Keap1. Reconstructed selected ion chromatogram (SIC) of m/z 706 after compound 4 incubation with GSH was different from that with Tris–HCl buffer solution, which meant the quinone reductase induction activity of compound 4 attributed to alkylating the sulfhydryl groups of Keap1.

Abstract:Four novel stilbene dimers (1-4), together with their biosynthetically related stilbene monomers (5 and 6), were isolated from the leaves of Cajanus cajan. Their structures with absolute configurations were determined by comprehensive analysis of spectroscopic data and electronic circular dichroism calculations. Compounds 1 and 2 are two novel dimeric stilbenes with an unusual coupling pattern that resulted in a rare configurationally stable Csp2-Csp3 chiral axis with both point and axial chirality in their molecules. Due to their unique inherent structural features, both of them naturally occur as equilibrating mixtures of unequally populated atropo-diastereomers and their respective enantiomers. Compounds 3 and 4 are two pairs of novel dimeric stilbene atropisomers featuring a rotationally hindered central biaryl axis. Notably, 3 contains a rare arylbenzoquinone core and 4 is a symmetric dimer with a C2 symmetry axis. The hypothetical biosynthetic pathway of 1-4 was also proposed herein. All the new compounds exhibited significant protein tyrosine phosphatase-1B (PTP1B) inhibition effects. In addition, the preliminary mode of action for the most potent compound 3 was investigated by molecular docking and binding free energy calculation.

Abstract:Four new stilbenes ( 1 – 4 ) and one new flavonoid ( 5 ), named cajanines D–H, together with three known stilbenes ( 6 – 8 ) were isolated from the leaves of Cajanus cajan (L.) Millsp. (pigeon pea). The structures of these compounds were elucidated unambiguously on the basis of IR, 1D, and 2D NMR, as well as HRESIMS data. Structurally, stilbenes 1 – 4 bore an isopentyl side chain, and further hydroxylation of compounds 1 – 3 generated a greater variety of structural forms. Compound 5 was a flavonoid owning an isopentyl side chain. Besides, antibacterial activity of the isolated compounds against Staphylococcus aureus , Bacillus cereus , and Escherichia coli was studied in vitro. Compounds 1 – 8 were endowed with profound antibacterial activity. Among them, the MIC values of compounds 4 , 6 , and 7 against S. aureus were 1.56, 0.78, and 0.78 μg/mL, respectively, among which 6 and 7 had better antibacterial activity than the positive control Vancomycin with the MIC values of 1.56 μg/mL. Additionally, the anti-SARS-CoV-2 main protease activity of all the isolated compounds was evaluated, and it was worth mentioning that the IC 50 values of compounds 5 – 7 were 8.27, 4.65, and 8.30 μM, respectively, being comparable to the positive control Ebselen. Our findings may provide valuable guidance for the application of stilbenes as lead compounds in the future for the development of drugs with antibacterial or anti-COVID-19 activity.

Abstract:Three new compounds, including two new diterpenoids, named epianhydrocinnzeylanol (1) and cinnacasiol H (2), and one hydroxylasiodiplodin, (3R,4S,6R)-4,6-dihydroxy-de-O-methyllasiodiplodin (3), together with five known diterpenoids (4–8) and two known phenolic glycosides (9–10) were isolated from the barks of Cinnamomum cassia. Their structures were elucidated by extensive spectroscopic analysis and comparison of the chemical shift values with those of related known compounds. The anti-inflammatory activities of the isolates were evaluated on nitric oxide production in lipopolysaccharide-induced BV-2 microglial cells and the compounds showed weak inhibition activities.

Abstract:Eighteen stilbenes (1–18), including six previously undescribed ones (1–6), with diverse modification patterns were isolated from the leaves of edible and medicinal plant Cajanus cajan. Among the new isolates, compounds 1–3 were initially obtained as three racemic mixtures, which were further resolved into three pairs of optically pure enantiomers, respectively, by chiral HPLC. Besides, compounds 8, 10, 11, and 18 were obtained from C. cajan for the first time. The chemical structures and absolute configurations of the new stilbenes were elucidated unambiguously on the basis of extensive spectroscopic analyses, single crystal X-ray crystallographic study, and quantum chemical electronic circular dichroism (ECD) calculations. In addition, the in vitro anti-inflammatory activities of all isolated stilbenes were evaluated. Compounds 2, 9, 10, 11, and 14 exerted moderate suppression of nitric oxide (NO) secretion in lipopolysaccharide (LPS)-induced RAW264.7 cells without exhibiting substantial cytotoxicity.

Their structures were elucidated on the basis of HR-ESI-MS and NMR analyses, and their absolute configurations were determined by ECD comparison. Cephaliverin A (1), herpetotriol (5) and hedyotol A (6) exhibited moderate antitumor activity against HepG2 and A549 cell lines.

Abstract:Two new cassane diterpenoids, sucupiranin MN ( 1 ) and sucupiranin ML ( 2 ), together with two known compounds sucutinirane C ( 3 ) and deacetylsucutinirane C ( 4 ) were isolated from the seed kernels of Caesalpinia sinensis . Their structures were elucidated by means of analysis of comprehensive spectroscopic data, especially HRESIMS and 1D/2D NMR spectroscopy. Compounds 1–4 are typical furan-type cassane derivatives with an aromatized C ring. Biological evaluation revealed that compounds 1–4 at the concentration of 10 μM could inhibit the overproduction of NO in LPS-stimulated RAW 264.7 macrophages.

Abstract:Eight previously undescribed diterpenoids, caesamins A-H (1-8), were separated and identified from the seeds of Caesalpinia minax Hance. Their structures were characterized by extensive spectroscopic data and X-ray crystallographic analysis. Structurally, caesamin A (1) is the first cassane-type diterpenoid with a C23 carbon skeleton containing an unusual isopropyl. Caesamin F (6) represents the first example of cleistanthane diterpenoid from the genus Caesalpinia. Caesamins B (2) and F (6) exhibited inhibitory activity against LPS-induced nitric oxide production in RAW 264.7 macrophages with IC50 values of 45.67 ± 0.92 and 42.99 ± 0.24 μM, comparable to positive control 43.69 ± 2.62 μM of NG-Monomethyl-L-arginine. Furthermore, the chemotaxonomic significance of the isolates was discussed.

Abstract:Objective: To isolate and identify the undescribed compounds from the fruits of Cinnamomum migao and evaluate its nitric oxide inhibition potential.Methods: The chromatographic techniques of silica gel, Sephadex, and HPLC were used for isolation and purification of the compounds, while HR-ESI-MS, 1D NMR, 2D NMR, ECD, and X-ray diffraction tech-niques were used to characterize and confirm the isolated compounds. Moreover, the anti-inflammatory activity of the isolated compounds was carried out to check inhibitory potential against the production of nitric oxide with RAW264.7 cells stimulated by LPS.Results: Camganoid A (1), a novel sesquiterpene possessing an unprecedented skeleton, and camganoid B (2), containing a unique eight-membered sesquiterpene moiety with a new carbon skeleton, were iso-lated and identified from the fruits of C. migao. The absolute configurations of 1 and 2 were confirmed by single crystal X-ray diffraction and electronic circular dichroism (ECD) calculations. Among these com-pounds, compound 1 exhibited potent inhibitory activity against the production of nitric oxide with IC50 value of 4.59 lmol/L in RAW264.7 cells stimulated by LPS.Conclusion: The isolation of two new skeletons from the fruits part of C. migao possessed unique skele-tons which have not been reported before.(c) 2022 Tianjin Press of Chinese Herbal Medicines. Abstract:Fifteen previously undescribed coumarin derivatives, murratins A-M, including two pairs of coumarin enantiomers with a cyclopropane unit, two benzocoumarins, a dimeric coumarin, and eight C-8-substituted coumarins, together with six known analogues were isolated from the extract of the leaves and twigs of Murraya exotica L., a medicinal plant named "Jiulixiang" in Chinese. Their structures were elucidated on the basis of comprehensive analysis of 1D and 2D NMR and HRMS spectroscopic data, and the absolute configurations were assigned via comparison of the specific rotations, the ECD exciton coupling method, comparison of experimental and calculated ECD data, and the ECD data of in situ formed transition metal complexes. All the isolates were evaluated for their inhibitory effects on lipopolysaccharide-induced nitric oxide production in RAW 264.7 cells, and five coumarin derivatives showed moderate inhibitory activities. The possible mechanism for NO inhibition of undescribed bioactive compounds was deduced to interact with iNOS protein via molecular docking. The above results shed some light on a better understanding of the traditional anti-inflammation effect of M. exotica and coumarins are disclosed to be its potential anti-inflammatory constituents.

Abstract:A new coumarin ( 1 ) and a new flavonoid ( 2 ) were isolated from the air-dried flower buds of Ochrocarpus longifolius , together with ten known compounds ( 3 – 12 ). The structures of two new compounds were established by 1D and 2D NMR and MS data. In addition, the new compound 2 showed significant proliferation inhibitory activity on Eca-109 and MGC-803 cells. The results of this study may enrich the diversity of compounds from O. longifolius and provide a basis for further research on its natural products and pharmacological activities.

Abstract:Two new flavanocoumarins, 1 and 2, together with phyllocoumarin (3) and epiphyllocoumarin (4), were isolated from the leaves of Litsea coreana LEVL. in Anhui Province, China. The structures of 1-4 were elucidated by spectroscopic methods. Biological tests revealed that 1-4 exhibited moderate anti-inflammatory activities through inhibition of TNF-α and IL-1 production in primary mouse peritoneal macrophages activated with lipopolysaccharides (LPS).
