Abstract:We report a design strategy for integrative assembly of heteromeric [2]catenanes. The design focuses on the shape and functional group match of two different metalla-rectangles. A series of dipyridyl ligands with different lengths, widths and functional groups were designed and used for assembly experiments. Six heteromeric [2]catenanes were obtained both by direct mixture of two pre-assembled metalla-rectangles and one-pot three-component self-assembly. Multiple analytic methods were employed to characterize the catenanes, including single crystal X-ray diffraction analysis, NMR spectroscopy, mass spectroscopy and elemental analysis.

Abstract:Although the photothermal conversion properties of various self-assembled ensembles based on π-π stacking interactions have been reported, the impact of different stacking modes on photothermal conversion efficiency has rarely been investigated. In this study, three similar flexible pyridine ligands were assembled with the binuclear Cp*Rh building block B1, leading to the formation of three [2]-catenanes in trapezoidal metallacycles with L-B-B-L, L-L-B-B and B-L-L-B stacks. We demonstrated that the π-π stacking modes of the building blocks can be controlled by adjusting the length and flexibility of the linkers. Comparative research studies of the NIR photothermal conversion efficiencies (57.0–83.7%) of the three [2]-catenanes indicated that the conversion efficiencies of the [2]-catenanes depend on the stacking modes of the building blocks and that [2]-catenane with the L-B-B-L stack exhibited the best photothermal conversion ability. This research emphasizes the significance of stacking modes and presents a new approach for creating valuable NIR photothermal conversion materials based on half-sandwich metal structures.

Abstract:Cycloparaphenylenes (CPPs) represent a significant challenge for the synthesis of mechanically interlocked architectures, because they lack heteroatoms, which precludes traditional active and passive template methods. To circumvent this problem and explore the fundamental and functional properties of CPP rotaxanes and catenanes, researches have resorted to unusual non-covalent and even to labor-intensive covalent template approaches. Herein, we report a ring-in-ring non-covalent template strategy that makes use of the surprisingly strong non-covalent inclusion of crown ethers into suitably sized CPPs. By threading a secondary ammonium salt through the crown ether and closing the third ring via Cu-AAC click reaction, we obtained a hetero-[3]catenane comprising [12]CPP, 24-crown-8 and a dibenzylammonium macrocycle. X-ray crystallography shed light on the ring-in-ring pre-organization and the [3]catenane topology was confirmed by NMR and MS-MS studies. Molecular simulations provided insights into the intriguing ring-vs.-ring-vs.-ring dynamics of the [3]catenane, which are highly dependent on the protonation state of the dibenzylammonium site. This ring-in-ring assembly strategy opens new avenues for the synthesis of complex CPP architectures and their use in functional supramolecular systems.

Self-Assembling Calix[4]Arene [2]catenanes. Preorganization, Conformation, Selectivity, and Efficiency

Abstract:A novel class of preorganized U-shape calix[4]arene clefts, dicationic salts 3a,c,e.2Cl, 3b.2PF(6)(), and 3f.2Br, consisting of one cone calix[4]arene and two bipyridine residues being linked by an aliphatic chain, have been designed and synthesized as precursors for self-assembly of calix[4]arene [2]catenanes by utilizing pi-stacking interactions between the hydroquinone and bipyridinium units. Conformationally flexible 6.2PF(6)() and cone 10.2Cl, whose conformation is fixed by two propyloxy groups on the lower rim, were also prepared in order to explore the effects of conformation and hydrogen bonding of the calix[4]arene moiety on self-assembly. For all reactions, bis-p-phenylene-34-crown ether-10 (11) was employed as the donor component. Alternate cone [2]catenane 13.4Cl is obtained in 8% yield from reaction of ethylene-incorporating 3a.2Cl and 1,4-bis(bromomethyl)benzene (12a). Three cone and one conformationally flexible [2]catenanes were obtained in moderate to good yields from reactions of propylene-incorporating 3b.PF(6)() and 3c.2Cl with 12a, 1,3-bis(bromomethyl)benzene (12b) or 4,4'-(bromomethyl)biphenyl (12c). Both cone and partial cone [2]catenanes were generated in moderate yields from butylene-incorporating 3c.2Cl with four tert-butyl groups on the calix[4]arene moiety and with 12a. In contrast, only cone [2]catenane was obtained from similar tert-butyl-free cleft 3d.2Cl. Cone and conformationally flexible [2]catenanes were obtained in moderate yields, respectively, from the reactions of 3d.2Cl and 3e.2Cl with 12c. No catenanes were isolated from reaction of phenylene-incorporating 3f.2Br or 6.2Cl, whereas reaction of 10.2Cl afforded cone [2]catenane in low yield. It was demonstrated that hydrogen bonding, which may be destroyed after catenation, within the calix[4]arene moiety is crucial for efficient self-assembly of the [2]catenanes. The dynamic (1)H NMR and absorption spectra and luminescent properties of the [2]catenanes were investigated, which reveal that incorporation of calix[4]arene into the tetracationic cyclophane reduces pi-stacking interactions between the donor and acceptor units and catenation has substantial influence on conformational distributions of the calix[4]arene moiety. The results demonstrate the versatility of calix[4]arene derivatives as building blocks in the construction of supramolecular structures.

Abstract:One linear template 13 and one cyclophane template 15, both incorporating two electron rich 1, 4-dialkoxybenzene units and one diamide unit, have been synthesized. By utilizing donor-acceptor interaction and/or intermolecular hydrogen bonding assembling principles, one novel hetero[3]rotaxane 22-40, possessing one neutral and one tetracationic ring components, has been synthesized from 13, through neutral [2]rotaxane 21 as intermediate. With 15 as template, tetracationic [2]catenane 23 .4PF(6) was assembled by using donor-acceptor interaction, but no neutral [2]rotaxane could he obtained under the typical conditions of hydrogen bonding assembling principle. The interlocked supramolecular compounds have been characterized and their spectral properties are investigated.

Abstract:Main observation and conclusionThe construction of interlocked topologies by metal‐cation‐template methods has been extensively explored. However, construction of these species using template‐free chemical self‐assembly methods is hard to rationally achieve due to the inherent unpredictability of self‐assembly processes. In this work, a strategy to construct interlocked [2]catenanes by coordination‐driven template‐free self‐assembly is demonstrated, whereby combining rigid bidentate ligands with large conjugated‐π area units and building blocks with appropriate lengths facilitated construction of interlocked [2]catenane compounds. Two rigid bidentate ligands, L1 and L2, were chosen to self‐assemble with three dinuclear building blocks B1, B2 and B3. Ultimately, the successful construction of three [2]catenanes demonstrated the validity of the strategy. Although [2]catenanes are the simplest of all interlocked topologies, this motif is the basis for constructing more intricate topologies. This successful strategy may therefore lead to construction of other, more intricate topologies in the future.

Abstract:Although the synthesis of mechanically interlocked molecules has been extensively researched, selectively constructing homogeneous linear [4]catenanes remains a formidable challenge. Here, we selectively constructed a homogeneous linear metalla[4]catenane in a one-step process through the coordination-driven self-assembly of a bidentate benzothiadiazole derivative ligand and a binuclear half-sandwich rhodium precursor. The formation of metalla[4]catenanes was facilitated by cooperative interactions between strong sandwich-type π-π stacking and non-classical hydrogen bonds between the components. Moreover, by modulating the aromatic substituents on the binuclear precursor, two homogeneous metalla[2]catenanes were obtained. The molecular structures of these metallacatenanes were unambiguously characterized by single-crystal X-ray diffraction analysis. Additionally, reversible structural transformation between metal-catenanes and the corresponding metallarectangles could be achieved by altering their concentration, as confirmed by mass spectrometry and NMR spectroscopy studies.

Abstract:One fascinating and challenging synthetic target in the field of mechanically interlocked molecules is the family of linear [4]catenanes, which are topologically identical to the logo of automobile maker Audi. Herein, we report an "all-in-one" synthetic strategy for the synthesis of linear metalla[n]catenanes (n = 2–4) by the coordination-driven self-assembly of Cp*Rh-based (Cp* = η5-pentamethylcyclopentadienyl) organometallic rectangle π-donors and tetracationic organic cyclophane π-acceptors. We selected the pyrenyl group as the π-donor unit, leading to homogeneous metalla[2]catenanes and cyclic metalla[3]catenanes via π-stacking interactions. By taking advantage of the strong electrostatic interactions between π-donor units and π-acceptor units, a heterogeneous metalla[2]catenanes and linear metalla[3]catenanes, respectively, could be obtained by the simple stirring of homogeneous metalla[2]catenanes with a suitable tetracationic cyclophane. On this basis, this "all-in-one" synthetic strategy was further used to realize a quantitative one-step synthesis of a linear metalla[4]catenanes via the self-assembly of cyclic metalla[3]catenanes and tetracationic cyclophanes. Abstract:Although catenated cages have been widely constructed due to their unique and elegant topological structures, cyclic catenanes formed by the connection of multiple catenane units have been rarely reported. Herein, based on the orthogonal metal-coordination-driven self-assembly, we prepare a series of heterometallic [2]catenanes and cyclic bis[2]catenanes, whose structures are clearly evidenced by single-crystal X-ray analysis. Owing to the multiple positively charged nature, as well as the potential synergistic effect of the Cu(I) and Pt(II) metal ions, the cyclic bis[2]catenanes display broad-spectrum antibacterial activity. This work not only provides an efficient strategy for the construction of heterometallic [2]catenanes and cyclic bis[2]catenanes but also explores their applications as superior antibacterial agents, which will promote the construction of advanced supramolecular structures for biomedical applications.

Abstract:A stepwise self-assembly protocol has been used to synthesize [2]- and [3]catenanes. Firstly, binuclear Cp*Rh/Ir-directed (Cp*=pentamethylcyclopentadienyl) pseudorotaxanes were prepared through self-assembly, driven by donor-acceptor interactions between electron-deficient naphthalenediimide (NDI) units and an electron-rich crown ether. Subsequently, the pre-organized pseudorotaxanes were applied as tectons for self-assembly of [2]- and [3]catenanes by combination with very simple linkers. The structures of the catenanes were confirmed by NMR spectroscopy, ESI mass spectrometry, single-crystal X-ray diffraction analysis, and elemental analysis.
π-π Interaction-facilitated formation of interwoven trimeric cage-catenanes with topological chirality

Abstract:Catenanes as interlocked molecules with a nonplanar graph have gained increasing attention for their unique features such as topological chirality. To date, the majority of research in this field has been focusing on catenanes comprising monocyclic rings. Due to the lack of rational synthetic strategy, catenanes of cage-like monomers are hardly accessible. Here we report on the construction of an interwoven trimeric catenane that is composed of achiral organic cages, which exhibits topological chirality. Our rational design begins with a pure mathematical analysis, revealing that the formation probability of the interwoven trimeric catenane surpasses that of its chain-like analogue by 20%; while driven by efficient template effect provided by strong {\pi}-{\pi} stacking of aromatic panels, the interwoven structure emerges as the dominant species, almost ruling out the formation of the chain-like isomer. Its topological chirality is unambiguously unravelled by chiral-HPLC, CD spectroscopy and X-ray diffraction. Our probability analysis-aided rational design strategy would pave a new venue for the efficient synthesis of topologically sophisticated structures in one pot.

Abstract:Abstract Despite extensive research and several stunning breakthroughs in the synthesis of interlocked molecular species, [3]catenanes, Borromean rings and ring-in-ring complexes are exceedingly rare and their targeted synthesis remains a formidable challenge. Herein, a series of Cp*Rh-based homogeneous and heterogeneous interlocked structures have been prepared by coordination-driven self-assembly, not only including metalla[2]catenanes and molecular Borromean rings, but also linear metalla[3]catenanes and ring-in-ring complexes. The interlocked structures are all based on bithiophenyl groups. The bithiophenyl groups effectively enhance the strength of the inter-ring interactions and play a crucial role in the formation of these interlocked structures. By taking advantage of the strong interaction between π-donor (D) and π-acceptor (A) groups, the electron-deficient methylviologen cation was introduced into a cationic metallarectangle based on bithiophenyl groups. Taking inspiration from these results, a cationic metallarectangle based on A units was threaded into a metallarectangle based on D units, leading to a heterogeneous D–A ring-in-ring structure.

Abstract:A multicatenane is described in which two belts consisting of four annelated rings attached to the wide rims of two calix[4]arenes are interwoven in such a way that each ring of one belt penetrates two adjacent rings of the other belt and vice versa. The key step of the synthesis of this [8]catenane is the exclusive formation of preorganized heterodimers between a multimacrocyclic tetraurea calix[4]arene and an “open-chain” tetraurea calix[4]arene containing eight ω-alkenyl groups. When a tetraurea calix[4]arene containing four alkenyl groups is used, a bis-[3]catenane is formed analogously.

Abstract:Abstract The study of cross-catenated metallacages, which are complex self-assembly systems arising from multiple supramolecular interactions and hierarchical assembly processes, is currently lacking but could provide facile insights into achieving more precise control over low-symmetry/high-complexity hierarchical assembly systems. Here, we report a cross-catenane formed between two position-isomeric Pt(II) metallacages in the solid state. These two metallacages formed [2]catenanes in solution, whereas a 1:1 mixture selectively formed a cross-catenane in crystals. Varied temperature nuclear magnetic resonance experiments and time-of-flight mass spectra are employed to characterize the cross-catenation in solutions, and the dynamic library of [2]catenanes are shown. Additionally, we searched for the global-minimum structures of three [2]catenanes and re-optimized the low-lying structures using density functional theory calculations. Our results suggest that the binding energy of cross-catenanes is significantly larger than that of self-catenanes within the dynamic library, and the selectivity in crystallization of cross-catenanes is thermodynamic. This study presents a cross-catenated assembly from different metallacages, which may provide a facile insight for the development of low-symmetry/high-complexity self-assemble systems.

Abstract:A variety of novel calix[4]arene-incorporating crown ethers with or without intramolecular hydrogen bonding have been prepared by two efficient methods and utilized as donor rings to assemble calix[4]arene [2]catenanes based on n-stacking interaction between hydroquinone and bipyridinium units. Treatment of calix[4]arene crown ethers 4, 10a, or 10b, whose cone conformation was fixed by intramolecular hydrogen bonding within the calix[4]arene moiety, with dicationic salt 15.2PF(6) and dibromide 16 afforded the corresponding [2]catenanes 17a.4PF(6), 17b.4PF(6), and 17c.4PF(6) in 20%, 53%, and 55% yields, respectively, whereas from the reactions of 15.2PF(6) and dibromide 16 in the presence of conformationally flexible 11 or 12 with a cone conformation kept by two propyl groups, [2]catenanes 18.4PF(6) and 19.4PF(6) were obtained in 12% and 6% yields. [2]Catenanes 21a.4C, 21b.4Cl, and 21c.4Cl, incorporating calix[4]arene in both the donor and acceptor rings, were also successfully assembled from 10a or 10b, 16, and dicationic salts 20a.2PF(6) or 20b.2PF(6). The dynamic H-1 NMR and absorption spectra of the [2]catenanes have been investigated, which revealed a strongest donor-acceptor interaction in 17a.4PF(6) and that the cone [2]catenanes 17a.4Cl can isomerize to the partial cone isomer at high temperature. The difference of the dynamic properties of these catenanes was discussed. The results demonstrate that catenation is one new general method to change the conformational distributions of calix[4]arenes.
