Abstract:In this article, we present an efficient, one-pot regioselective approach towards the synthesis of 2-aryl/hetaryl-4-methyl-5-acylthiazoles obtained during the reaction of α-bromo-1,3-diketones generated in situ by triturating unsymmetrical 1,3-diketones with N-bromosuccinimide, with various thioamides under solvent-free conditions. This environmentally benign protocol showed large functional group tolerance, resulted in the exclusive formation of a single isomer, out of the two possible regioisomers in admirable yields. The structure of the isomeric thiazole was assigned unambiguously on the basis of multinuclear NMR [(1H-13C) HMBC, (1H-13C) HMQC, (1H-15N) HMBC] and X-ray crystallographic studies. Abstract:This study reports on the thiourea dioxide catalyzed transfer hydrogenation of diverse C=N–containing heterocyclic compounds with Hantzsch ester as the hydrogen source. With this cost effective and readily available catalyst, a wide range of 2-substituted quinolines, 3-substituted-2H-1,4-benzoxazines and 3-substituted-2H-1,4-benzothiazines were efficiently reduced to the corresponding tetrahydroquinolines, dihydro-2H-benzoxazines and dihydro-2H-benzothiazines under mild conditions.

Abstract:With water as the sole solvent, a green and efficient method has been developed for the synthesis of various α,α-dihaloketones via ultrasound assisted p-tolylthiourea catalyzed tandem reaction of alkynes with trihaloisocyanuric acids. This synthetic route could effectively avoid the use of toxic organic solvents and transition metal catalysts, and the products could be obtained in a very short time at room temperature with good to excellent yields.

Abstract:A facile catalyst-free sulfonylation of phenoxazine with sulfonyl hydrazides was efficiently developed in H 2 O/HFIP without any catalyst. Abstract:A simple and general method for anti-selective hydrosulfonylation of unactivated alkynes with sulfonyl chlorides in the presence of a catalytic amount of phenanthroline-based Lewis base and (Me3Si)3SiH as the hydrogen atom donor has been developed. The protocol proceeds efficiently under mild and metal-free conditions, delivering a diverse set of (Z)-vinyl sulfones with high stereoselectivity. Additionally, the method displays excellent functional-group compatibility and can be applied to the late-stage modifications of complex drugs and their derivatives. Experimental and density functional theory studies unveiled that the key to the transformation’s success is the employment of a phenanthroline-based Lewis base, which interacts with sulfonyl chlorides to form a halogen-bond complex that accelerates the cleavage of the S–Cl bonds of sulfonyl chlorides under the irradiation of visible light.

Abstract:Although sulfoxide is an important structural motif found in biological molecules and prevalent feedstocks in synthetic chemistry, a general and sustainable access route to ( Z )- α,β -unsaturated sulfoxides remains highly challenging due to the thermodynamic instability of Z -type geometric and the notorious sulfur nucleophiles poisoning of metal catalyst. Here we report a new protocol based on homogeneous octamolybdate-catalyzed anti -hydrothiolation and after Z -retentive sulfoxidation methodology, allowing access to ( Z )- α,β -unsaturated sulfoxides from commercially available alkynes, thiols and H 2 O 2 . Control experiment investigations indicate that the molecular molybdenum is a thiyl radical initiator, Z -type geometric mediator and sulfoxidation catalyst. Notably, this one-pot reaction features broad substrate scopes with high stereoselectivity (Z/E up to > 99:1) and mild reaction conditions, thus providing a promising approach for the preparation of Z -vinyl sulfoxide compounds.

Abstract:A solvent-controlled divergent thiocyanation of carbazoles and di/triphenylamines has been developed. Using either CH 3 CN/H 2 O (3:1) or DMSO as the solvent, a facile and efficient protocol for the solvent-controlled divergent synthesis of mono- and di-thiocyanation of carbazoles and di/triphenylamines was achieved. Abstract:Mono substituted thioureas are valuable chemical building blocks for the synthesis of pharmaceutically relevant molecules in both academia and the industry. We report here a catalyst-free preparation of mono substituted thioureas under very mild reaction condition. Condensation reactions of amines with carbon disulfide in ammonia solution gave mono substituted thioureas in good to excellent yield under catalyst-free condition.

Abstract:Chiral tetrahydroquinoxalines and dihydroquinoxalinones represent the core structure of many bioactive molecules. Herein, a simple and efficient Rh-thiourea-catalyzed asymmetric hydrogenation for enantiopure tetrahydroquinoxalines and dihydroquinoxalinones was developed under 1 MPa H pressure at room temperature. The reaction was magnified to the gram scale furnishing the desired products with undamaged yield and enantioselectivity. Application of this methodology was also conducted successfully under continuous flow conditions. In addition, H NMR experiments revealed that the introduction of a strong Brønsted acid, HCl, not only activated the substrate but also established anion binding between the substrate and the ligand. More importantly, the chloride ion facilitated heterolytic cleavage of dihydrogen to regenerate the active dihydride species and HCl, which was computed to be the rate-determining step. Further deuterium labeling experiments and density functional theory (DFT) calculations demonstrated that this reaction underwent a plausible outer-sphere mechanism in this new catalytic transformation.

Abstract:A simple method to synthesize diverse sulfonyl chlorides through NaClO2-mediated oxidative chlorosulfonation of S-alkyl isothiourea salts is presented. The approach features safe operation, environmental friendliness, convenient purification procedures, and delivers high yields of up to 96%. The procedure is also applicable to substrates such as thiols, disulfides, thioacetates, and xanthates. It is a versatile and convenient method for the synthesis of various sulfonyl chlorides from different thiol precursors and derivatives.
