A Simple Automated Synthesis of [11c]carfentanil Utilizing Spe Method Without Hplc Separation for the Routine Use

Abstract:1960 Objectives [11C]carfentanil, a highly potent mu-opioid receptor agonist, has become routinely used in human studies of opioid dependence etc. For the routine use, a fully automated module adopting a solid phase extraction (SPE) purification method instead of HPLC for [11C]carfentanil synthesis has been developed. Methods The module was completed by modifying the current module (PET-CS-1) for the [11C]choline. All reagents were loaded before [11C]CH3OTf was bubbled into the reaction vial with 0.5 mg salt of the precursor carboxylate in 0.5 ml DMSO. Upon maximal trapping of radioactivity, the reaction was quenched. The reaction mixture was then diluted with 10 ml of 0.15 N ammonium hydroxide aqueous solution, followed by passing through the disposable C2 cartridge. The impurities were successfully removed by rinsing the cartridge with 10 ml water. Since the [11C]carfentanil was trapped in C2 cartridge, it was easily eluted ethanol. The [11C]carfentanil product was compared to an authentic sample of unlabelled carfentanil with HPLC . Results With this module, the radiochemical yields of [11C]carfentanil was obtained as 61.0±5.0mCi (n=12) from 410±30 mCi [11C]CO2. The overall synthesis time need only 15 min and radiochemical purity was higher than 97%. The average specific activity at end of synthesis was 3400mCi/mmole. Conclusions A fully automated module for [11C]carfentanil synthesis was successfully developed through improving the current module. Consequently, [11C]carfentanil was prepared with high radiochemical yields and purity for routine use. In contrast to the previous method, less precursor (0.5 mg) was used and, more importantly, no strong base (tetrabutylammonium hydroxide) was necessary in this method.
