A New Simple Automated Synthesis of the Central Benzodiazepine Receptor Imaging Radiotracer [11C]flumazenil

Abstract:1468 Objectives The radiotracer, [11C]Flumazenil, a highly selective benzodiazepine antagonist, has been the most widely used for PET imaging of the central benzodiazepine receptor. For the routine use, a fully automated synthesis method utilizing ethanol solution instead of acetonitrile for HPLC purification has been developed. Methods The module was completed by modifying the current module (PET-CS-1) for the [11C]choline. All reagents were loaded in the module before [11C]CH3OTf was bubbled into the reaction vial with 0.5 mg precursor desmethylflumazenil in 0.3 ml KOH in DMF solution at 0 °C. Upon maximal trapping of radioactivity, the final reaction mixture was heated to room temperature for 5 minute. The reaction mixture was then diluted with 1.0 ml of HPLC mobile phase (30% ethanol solution), followed by loaded into the prep-HPLC column. The flow rate is 3ml/min. With UV and radio-detector analysis the required fraction (5 ml) is then collected into a product vial. Saline (25 ml) is added to give the final product solution containing 10% v/v ethanol via a 0.22μm vented filter. Results With this method, the radiochemical yields of [11C]Flumazenil was obtained as 120±10mCi (n=20) from 1500±50 mCi [11C]COx. The overall synthesis time need only 18 min and radiochemical purity was higher than 98%. The average specific activity at end of synthesis was 3500mCi/mole. Conclusions A new simple automated synthesis of [11C]Flumazenil without acetonitrile was successfully developed. [11C]Flumazenil was prepared with high radiochemical yields and purity for routine use. In contrast to the previous method, there was no requirement to perform solid phase extraction (SPE) to remove toxic solvents acetonitrile. Instead the [11C]Flumazenil fraction was diverted directly into the final product vial, simplifying the synthesis procedure and eliminating the residual acetonitrile
