Abstract:Abstract A simple and highly efficient approach has been developed for a multicomponent one-pot reaction of aldehydes or ketones with amines and diethyl or triethyl phosphite to give the corresponding α-aminophosphonates. In the presence of quaternary ammonium salts, which are environmental friendly, inexpensive, and recyclable, α-aminophosphonates were obtained in excellent yields at room temperature within short times.

Abstract:A simple, green and highly efficient approach for the one-pot three-component synthesis of beta-phosphonomalonates has been developed. In the presence of the quaternary ammonium salt catalysts, the beta-phosphonomalonates were obtained in excellent yields within short times via tandem Knoevenagel-phospha-Michael reaction. The reaction of aldehydes/ketones, active methylene compounds, and diethyl phosphite performed at room temperature under solvent-free conditions. No column purification is required and the products can be purified by simple crystallization. The catalysts can be easily recovered and reused several times without significant activity loss.

Abstract:Some acyclic acidic ionic liquids were prepared and used as the catalysts for the one-pot, three-component synthesis of -aminophosphonates from aldehydes, amines, and trimethylphosphite at room temperature in water. The products could simply be separated from the system of catalyst/water, and the catalysts could be reused at least six times without noticeably decreasing the catalytic activity.

Abstract:The various alpha-aminophosphonates have been prepared from tertiary aromatic and aliphatic amines with P(O)H compounds via a tertbutyl hydroperoxide mediated cross-dehydrogenative coupling reaction, eliminating the need for transition-metal catalysts. The oxidation of tertiary amine by tert-butyl hydroperoxide generates an iminium cation intermediate. A further addition of P(O)H compound to iminium cation gives the desired product.

Abstract:α-Aminophosphonic acids are phosphorus analogues of α-amino acids. Compounds of this type find numerous applications in medicine and crop protection due to their unique biological activities. A crucial factor in these activities is the configuration of the stereoisomers. Only a few methods of stereoselective transformation of α-amino acids into their phosphorus analogues are known so far and all of them are based on asymmetric induction, thus involving the use of a chiral substrate. In contrast, we have focused our efforts on the development of an effective method for this type of transformation using a racemic mixture of starting N-protected α-amino acids and a chiral catalyst. Herein, a simple and efficient stereoselective organocatalytic α-amidoalkylation of dimethyl phosphite by 1-(N-acylamino)alkyltriphenylphosphonium salts to enantiomerically enriched α-aminophosphonates is reported. Using 5 mol% of chiral quinine- or hydroquinine-derived quaternary ammonium salts provides final products in very good yields up to 98% and with up to 92% ee. The starting phosphonium salts were easily obtained from α-amino acid derivatives by decarboxylative methoxylation and subsequent substitution with triphenylphosphonium tetrafluoroborate. The appropriate self-disproportionation of enantiomers (SDE) test for selected α-aminophosphonate derivatives via achiral flash chromatography was performed to confirm the reliability of the enantioselectivity results that were obtained.

Abstract:An efficient enantioselective synthesis of cyclic α-aminophosphonates via multicomponent reactions of 2-alkynylbenzaldehydes, amines, and dimethylphosphonate has been developed with the use of a chiral silver spirocyclic phosphate as the catalyst. This protocol provides straightforward access to a series of chiral C1-phosphonylated 1,2-dihydroisoquinoline derivatives with high yields (up to 99%) and high enantioselectivities (up to 94% ee) for a broad substrate scope. Abstract:An efficient and practical method for the synthesis of alpha,alpha-difluoro-gamma-aminophosphonates through photocatalyzed intermolecular aminodifluoromethylphosphonation of alkenes has been developed. In this reaction, difluoromethylphosphonate is used as an important fluorinated reagent. Furthermore, the mild reaction conditions, simple operation, and broad substrate scope make this protocol very practical and attractive. The derivatization reaction in the synthesis of difluoromethylphosphonated chiral binaphthylamine ligands and alpha,alpha-difluoro-gamma-aminophosphoric acid highlight the applicability of this method.
