Abstract:Herein, we demonstrate a facile, straightforward and green method for synthesis of highly red-emitting fluorescent bimetallic Au/Ag nanoclusters (NCs) employing lipoic acid as both a reducing agent and a protecting agent. The as-prepared Au/Ag NCs exhibited bright red emission with a strong peak centered at 630 nm (quantum yield measured as 6.4%), and showed excellent stability and remarkable temperature-dependent fluorescent property. The fluorescence responses were reversible and linear to the environment temperature from 20 to 65 C with a resolution as high as 1.34 degrees C, which allows its potential application as a fluorescent nanothermometer. Moreover, Au/Ag NCs were also explored for selective and sensitive determination of Fe3+ based on the specific quenching of the fluorescence by Fe3+, and explored for fluorescence imaging Fe3+ in living HeLa cells thanks to their very low cytotoxicity. One-pot Green Synthesis of Highly Fluorescent Glutathione-Stabilized Copper Nanoclusters for Fe3+ Sensing

Abstract:Herein, we reported a simple one-pot green method for preparation of fluorescent copper nanoclusters (Cu NCs) with a high quantum yield up to 8.6%, using glutathione as the stabilizing agent. The as-prepared Cu NCs had good water solubility, excellent stability and ultrafine size through the characterization of UV-vis absorption spectroscopy, fluorescence spectroscopy, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared (FTIR) spectroscopy. Experimental results showed that the fluorescence of Cu NCs was linearly quenched by the Fe3+ concentrations in the range of 1-100 mu M, with the detection limit of 0.3 mu M. Abstract:An easily prepared fluorescent Ag nanoclusters (Ag NCs) probe for the sensitive and selective detection of Hg(2+) and Cu(2+) ions was developed here. The Ag NCs were synthesized by using polymethacrylic acid sodium salt as a template via a convenient hydrothermal process. The as-prepared fluorescent Ag NCs were monodispersed, uniform and less than 2 nm in diameter, and can be quenched in the presence of mercury (Hg(2+)) or copper (Cu(2+)) ions. Excellent linear relationships existed between the quenching degree of the Ag NCs and the concentrations of Hg(2+) or Cu(2+) ions in the range of 10 nM to 20 μM or 10 nM to 30 μM, respectively. By using ethylenediaminetetraacetate (EDTA) as the masking agent of Cu(2+), Hg(2+) was exclusively detected in coexistence with Cu(2+) with high sensitivity (LOD = 10 nM), which also provided a reusable detection method for Cu(2+). Furthermore, the different quenching phenomena caused by the two metals ions such as changes in visible colour, shifts of UV absorbance peaks and changes in size of Ag NCs make it easy to distinguish between them. Therefore the easily synthesized fluorescent Ag NCs may have great potential as Hg(2+) and Cu(2+) ions sensors.
Sensitive detection and intracellular imaging of free copper ions based on DNA-templated silver nanoclusters aggregation-inducing fluorescence enhancement effect

Abstract:Free copper ions (Cu+ and Cu2+) have critical toxicity to cells, although copper is an essential element for human body. Hence, sensitive monitoring is crucial to avoid over intake of Cu+/Cu2+. We herein designed a ssDNA sequence (A31) for synthetizing A31-templated silver nanoclusters (AgNCs), and demonstrated that Cu+/Cu2+ can induce the aggregation of A31-templated AgNCs and thus greatly enhanced the fluorescence emission of A31-templated AgNCs. Based on Cu+/Cu2+-induced fluorescence enhancement effect of A31-templated AgNCs, a label-free and signal-on fluorescent sensing platform was developed for the specific and sensitive detection of Cu+/Cu2+ in biological samples and intracellular imaging of Cu+/Cu2+ in cells. The signal-on fluorescent sensing platform could be used to rapidly detect Cu+ and Cu2+ with a detection limit of 0.1 µM within 30 min., and to perform the intracellular imaging of Cu+ and Cu2+ in cells with good cell permeability and biocompatibility. By using the signal-on fluorescent sensing platform, we have successfully detected Cu+ and Cu2+ in cells fluids and human serum with a recovery of 90-104% and a RSD (n = 5) < 5%, and performed the imaging of Cu+/Cu2+ in Hela cells. The developed fluorescent sensing platform has obvious analytical and imaging advantages such as signal-on, simple operation, short analysis time, both Cu+ and Cu2+ detection, similar or higher sensitivity, good cell permeability and biocompatibility, which promising a reliable approach for the rapid and on-site detection or imaging of free copper ions in biological samples in clinical diagnosis.

Abstract:A creatively designed and constructed a multifunctional ratiometric fluorescence probe is reported by assembling glutathione (GSH)-protected gold nanoclusters (AuNCs) with fluorescein-doped mesoporous silica nanoparticle (FS) for the detection of Cu 2+ and Ag + ions, which could eliminate most interferences by self-calibration. Under the excitation at 450 nm, the fluorescence connected with AuNCs can rapidly respond by quenching or enhancement, respectively, for Cu 2+ and Ag + ions, while the fluorescein isothiocyante (FITC) fluorescence served as reference with negligible change. The fluorescence intensity ratio showed good linear relationships with Cu 2+ and Ag + concentrations in the range 0.5–10 μM and 0.1–8 μM, respectively. The detection limits were as low as 140 nM and 60 nM for Cu 2+ and Ag + ions, respectively. The color change induced by fluorescent intensity ratio variation could also be employed for visual discrimination. The AuNC-embedded FS (FS-Au) nanoprobe was successfully used for Cu 2+ and Ag + ion determination in drinking water and intracellular Cu 2+ imaging, which exhibits promising prospects in cost-effective and rapid determination of both Cu 2+ and Ag + with good sensitivity and selectivity.
Preparation of Orange-Red Fluorescent Gold Nanoclusters Using Denatured Casein As a Reductant and Stabilizing Agent, and Their Application to Imaging of HeLa Cells and for the Quantitation of Mercury(ii)

Abstract:We describe a method for the preparation of water-soluble gold nanoclusters (Au-NCs) from chloroauric acid using denatured-casein as both a reducing and stabilizing agent. The resulting Au-NCs were characterized by photoluminescence, UV–vis absorption, and X-ray photoelectron spectroscopies, and by transmission electron microscopy. The Au-NCs have an average diameter of 1.7 ± 0.2 nm and exhibit orange-red fluorescence emission peaking at 600 nm (with a Stokes’ shift as large as 237 nm), a quantum yield of 4.3 %, and good stability over the physiologically relevant range of pH values and ionic strength. Cytotoxicity studies showed the Au-NCs to display negligible effects in terms of altering cell proliferation or triggering apoptosis. Fluorescence imaging of HeLa cancer cells was accomplished by loading such cells with the Au-NCs. The fluorescence of the Au-NCs is found to be strongly quenched by Hg(II) ions, and thus the Au-NCs can be used for detecting and, possibly, imaging of Hg(II). An assay was worked out for the determination of Hg(II), and its limit of detection is 1.83 nM, which is 5.5 times lower than the maximum allowed concentration of Hg(II) in drinking water as defined by the US EPA.

Abstract:Detection of Cu(II) is very important in disease diagnose, biological system detection and environmental monitoring. Previously, we found that the product TPE-CS prepared by attaching the chromophores of tetraphenylethylene (TPE) to the chitosan (CS) chains showed excellent fluorescent properties. In this study, we tried to use TPE-CS for detecting Cu(II) because of the stable complexation of CS with heavy metals and the luminosity mechanism of the Restriction of Intramolecular Rotations (RIR) for aggregation-induced emission (AIE)-active materials. The fluorescence intensity changed when TPE-CS was contacted with different metal ions, to be specific, no change for Na + , slightly increase for Hg 2+ , Pb 2+ , Zn 2+ , Cd 2+ , Fe 2+ , Fe 3+ due to the RIR caused by the complexation between CS and metal ions. However, for Cu 2+ , an obvious fluorescence decrease was observed because of the Photoinduced-Electron-Transfer (PET). Moreover, we found that the quenched FL intensity of TPE-CS was proportional to the concentration of Cu(II) in the range of 5 μmol/L to 100 μmol/L, which provided a new way to quantitatively detect Cu(II). Besides, TPE-CS has excellent water-solubility as well as absorbability (the percentage of removal, R = 84%), which is an excellent detection probe and remover for Cu(II).

Abstract:Alendronate sodium (ALDS) is an important and regularly used medicine in clinical therapy of osteoporosis. In this work, a new analytical method is developed for the detection of ALDS based on a fluorescent-platform of copper(II)-mediated silver nanoclusters (AgNCs). Specifically, Cu2+ can selectively bind with DNA-stabilized AgNCs and thus greatly quench their red fluorescence. In the presence of ALDS, their fluorescence can largely recover again owing to the stronger coordination of ALDS with Cu2+. On the basis of such a mechanism, fluorescence "on-off-on" mode of the nanoprobe is applied to detect ALDS in a wide linear range of 800 pM-200 mu M, with a detection limit of 100 pM. Compared with previously reported methods, the present approach has significant advantages regard to simplicity, high sensitivity, and a wide detectable concentration range. Abstract:Copper plays a key role in the human body; meanwhile, excess Cu2+ ions can result in various diseases. Nanoclusters (NCs) are often used to measure Cu2+ ions, but there are two difficulties. On the one hand, a single probe of NCs is easily affected by environmental factors. On the other hand, it is difficult to mask the interference of Pb2+ ions and Cd2+ ions in the process of detecting Cu2+ ions. As a new type of quantum dots (QDs), tungsten disulfide quantum dots (WS2-QDs) have some advantages of simple synthesis and stable luminescence properties. Stable WS2-QDs with blue fluorescence are used as a reference probe, while gold silver nanoclusters (AuAgNCs) with red fluorescence are used as a response probe. A ratiometric fluorescent sensor was constructed by mixing the two styles of fluorescent probes, which is abbreviated as NCs/QDs. This nano-sensor can be used to detect the concentration of Cu2+ ions, in which the fluorescence of QDs does not change significantly, while the fluorescence of NCs can be quenched by Cu2+ ions. The concentration of Cu2+ ions can be determined as low as 0.12 μM with a linear range from 0.3 to 3 μM. The common interference caused by Pb2+ and Cd2+ ions can be eliminated by the phosphate buffer solution (PBS). This sensor was used to detect the concentration of Cu2+ in river water with satisfactory results.

Abstract:In this study, fluorescent copper nanoclusters (Cu NCs) have been efficiently synthesized in a novel strategy via sonochemical method, using N-acetyl-L-cysteine (NAC) as reducing and stabilizing reagent. The preparation of Cu NCs could complete within 15 min due to the ultrasonic radiation treatment. The obtained NAC-Cu NCs exhibited red fluorescence at λem 630 nm. Silver ions could significantly induce fluorescence quenching of NAC-Cu NCs. Subsequently, a detection method for Ag+ with high response sensitivity and selectivity was constructed. The proposed method displayed a good linear relationship in the Ag+ concentration range of 1.64×10-10-1.16×10-8 M. The limit of detection (LOD) was as low as 7.76×10-11 M (signal/noise = 3). In addition, the practical application was further evaluated with environmental water samples, and the tested results were compared with those obtained by the inductively coupled plasma mass spectrometry (ICP-MS) method. The satisfactory recoveries (98.50%-101.6%) showed that the fluorescent detection method had potential application in environmental monitoring field.

Abstract:The concept that HLA‐linked immune response genes may influence survival of patients with malignant disease has been investigated by performing HLA typing on a series of 150 patients with acute myelogenous leukemia. Patients with A1 and B8 and/or A2 and B12 survive longer than patients without either of these pairs of antigens in association. However, after correction of the statistical probability for the number of A and B locus combinations theoretically possible, these differences are not statistically significant. Clarification of this must await independent analysis of other series and more direct approaches to the study of immune response genes in man.

Abstract:Cu2+ has increasingly become a great threat to the natural environment and human health due to its abundant content and wide application in various industries. DL-Mercaptosuccinic acid and ZnS-modified Fe3O4 nanocomposites were designed, synthesized, and applied in the determination of Cu2+. The prepared nanocomposites were characterized by scanning electron microscopy (SEM), transmission electron microscopes (TEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffractometer (XRD), X-ray photoelectron spectroscopy (XPS), vibrating sample magnetometer (VSM), and thermogravimetric analyzer (TG). The magnetic fluorescent nanoprobe exhibited highly selective and sensitive fluorescence-quenching characteristics with Cu2+ ions. The fluorescence detection linear range was 0–400 μM, with the detection limit being 0.489 μM. In addition, the magnetic fluorescent nanoprobe exhibited a high adsorption and removal rate for Cu2+. It had been successfully applied to detect Cu2+ in real water samples with a satisfactory recovery rate. The magnetic fluorescent nanoprobe could simultaneously realize the functions of enrichment, quantitative detection, and separation, reduce the pollution of copper ions and probes, and establish an environment-friendly detection method. Consequently, the magnetic fluorescent nanoprobe offered a new pathway for the removal and detection of not only Cu2+ but also other heavy metal ions in water.

Facile preparation of bimetallic Au-Cu nanoclusters as fluorescent nanoprobes for sensitive detection of Cr3+ and S2O82- ions

Abstract:Metallic nanoclusters (NCs) have attracted special attention from researchers due to their interesting optical properties. In this experiment, we proposed a facile one-step method for the synthesis of bimetallic gold-copper nanoclusters (AuCuNCs). The prepared AuCuNCs were characterized by fluorescence spectroscopy (FL), UV-vis absorption spectrum, transmission electron microscopy (TEM), etc. The emission peak of the prepared AuCuNCs was located at 455 nm and showed blue luminescence under the excitation of 365 nm UV light. Furthermore, after the addition of Cr3+ and S2O82- ions, the FL emission intensity of AuCuNCs was significantly reduced at 455 nm and there was a color change of diminished blue luminescence under UV lamp. The AuCuNCs exhibited excellent linearity and sensitivity for the detection of Cr3+ and S2O82- ions. The limits of detection (LOD) for the Cr3+ and S2O82- ions were calculated to be 1.5 and 0.037 μM, respectively. Finally, the recoveries of Cr3+ and S2O82- ions in Runxi Lake and tap water were measured by standard addition recovery test and were 96.66 ∼ 116.29 %, 95.75 ∼ 119.4 %.

Abstract:Copper nanoclusters (CuNCs) have become promising nanomaterials due to their high electronic conductivity and low cost. This study reports the fast synthesis of CuNCs through the use of hydrogen peroxide (H2O2) additive and their application for the fluorescent detection of Hg2+ in water samples. Cu2+ was rapidly reduced into Cu-0 in the presence of H2O2 and bovine serum albumin (BSA), further leading to the production of CuNCs. Under optimized conditions, the synthesis needs 1 h to achieve a high conversion rate of Cu2+ (more than 99%). In the resulting CuNCs, no Cu2+ can be found by the sodium sulfide test. Research on resonance light scattering, synchronous fluorescence and circular dichroism spectroscopy reveals that H2O2 may play two important roles in the synthesis of CuNCs. One role as a ligand is to combine with BSA-Cu complex to form BSA-Cu-H2O2 complex, which diminishes the reduction potential of Cu2+ and leads to the fast reduction of Cu2+ into Cu-0. Another role as an oxidizing agent is to partly destroy disulfide bonds in BSA, which increases the degree of exposure of free amino groups. This results in an enhanced reduction ability of BSA towards Cu2+. Interestingly, the as-prepared CuNCs display better fluorescence intensity and optical stability when compared with the CuNCs prepared by the conventional method. The nanosensor based on the CuNCs was developed for the rapid, reliable, sensitive and selective sensing of Hg2+ with a detection limit of 4.7 x 10(-12) M (S/N = 3) and a dynamic range of 1 x 10(-5)-1 x 10(-11) M. It has been successfully used for the detection of Hg2+ in water samples.
Green-Emissive Copper Nanocluster with Aggregation-Enhanced Emission for Selective Detection of Al3.

Abstract:Selective detection of Al3+ is of great significance both for the benefit of human health and environmental safety considerations. In this work, a sensitive and selective fluorescence assay for Al3+ was proposed based on the green-emissive Cu nanoclusters (Cu NCs). Different from the commonly reported works, the green emissive Cu NCs showed dual emission bands at 450 and 510 nm, attributed to the reaction product between polyvinyl pyrrolidone and ascorbic acid and the Cu core, respectively. Al3+ could induce the aggregation of Cu NCs by forming covalent bonds, which results in the enhancement of photoluminescence intensity. This enhancement phenomenon is rather selective to Al3+, which endows the detection in real samples. These results provide new insights for the fluorescence mechanisms of metal NCs, which also provided a functional luminescent material for various applications, such as chemical sensing, bioimaging and photoelectric devices.

Abstract:Glutathione-Au nanoclusters (GSH-AuNCs) are known as typical luminophores with aggregation-induced emission (AIE) features, which can be triggered by low temperature, low pH values, and metal ions. However, these strict "triggers" restrict the application of this AIE probe under mild and neutral conditions. In this work, fluorescent amino-modified silicon nanoparticles (SiNPs) and GSH-AuNCs can self-assemble into well-designed nanospheres through electrostatic interactions under physiological conditions. The hybrid probe of SiNPs@GSH-AuNCs exhibits dual emissions located at 450 nm from SiNPs and 570 nm originating from the aggregation enhanced emission (AEE) of GSH-AuNCs. Interestingly, the addition of Zn2+ or Cd2+ can connect the nanospheres to form large aggregates, so a unique second increase of AEE from GSH-AuNCs appears, while the emission of SiNPs remains constant to act as an internal reference. The fluorescence ratios (I570/I450) of SiNPs@GSH-AuNCs are positively correlated with Zn2+ or Cd2+ with the linear range from 1.5 μM to 500 μM. Most importantly, besides the advantages of dual emissions, high fluorescence brightness, and good stability, the SiNPs@GSH-AuNCs also exhibit excellent biocompatibility and low cytotoxicity, and have been successfully applied in zinc imaging in live cells. Furthermore, the proposed method is also used to measure Zn2+ and Cd2+ in real samples with satisfactory recoveries.
Regulation of Metal Ion Selectivity of Fluorescent Gold Nanoclusters by Metallophilic Interactions.

Abstract:Although numerous sensors have been successfully fabricated for the detection of various heavy metal ions by employing fluorescent gold nanoclusters (AuNCs) as nanoprobes, serious cross-interference often occurs when these ions coexist in samples, which results in glaring errors in quantification. In this study, glutathione-protected AuNCs (GSH-AuNCs) were synthesized and found to respond to both Cu2+ and Hg2+ via fluorescence suppression. Intriguingly, addition of Ag+ to GSH-AuNCs could completely inhibit the quenching effect of Hg2+ while not affecting the Cu2+-mediated quenching process. Ag+ can combine with Au+ on the surface of AuNCs to form a strong Ag+-Au+ metallic bond, which disrupts the interaction between Hg2+ and Au+ and thus eliminates the corresponding quenching effect. Based on this phenomenon, a simple sensing approach for highly selective and sensitive detection of Cu2+ in aqueous solution was developed using the GSH-AuNC/Ag+ complex as a fluorescent turn-off nanoprobe. The proposed method exhibited good linearity in the concentration range 0.02-10 mu M with a limit of detection of 12 nM. This assay was demonstrated to be suitable for determination of Cu2+ in real water samples even in the presence of Hg2+, showing great promise as a tool for assessment of environmental security and drinking water quality.
