Abstract:Two new polyhydroxylated sterols, named verumbsteroids A and B (1 and 2, resp.), along with seven known analog, 3-9, were isolated from the gorgonian Verrucella umbraculum collected from the South China Sea. The structures and relative configurations of the new compounds were elucidated by comprehensive spectroscopic investigations. The absolute configuration of 1 was determined by using modified Mosher method with the acetonide derivative of 1. Compounds 1 and 3-5 were found to show cytotoxicities against five human tumor cell lines (HL-60, K562, HeLa, A-549, and HCT-116) with the IC50 values ranging from 2.76 to 9.62 mu m.

Abstract:One new polyhydroxylated steroid, caerulsteroid A (1), along with nine known analogues (2-10), was isolated from the gorgonian coral Anthogorgia caerulea collected from the South China Sea. The structure of the new compound was elucidated by 1-D and 2-D NMR, and mass spectrometry. The absolute configuration of 1 was assigned on the basis of the absolute configurations of the related congeners 2 and 3, which were determined by application of the modified Mosher's method. The isolated compounds were assayed for their cytotoxicity, antimicrobial activity and lethality towards brine shrimp Artemia salina.

Abstract:Objective To study the chemical components of gorgonian Subergorgia suberosa.Methods The compounds were isolated by chromatography,and their structures were identified on the basis of spectroscopic analysis and comparison with the literature data.Results Eight compounds were isolated and identified as cholest-5,22-diene-3β-ol(1),ergosta-5,24(28)-diene-3β-ol(2),24-melthylcholest-5,22-dine-3β-ol(3),3β-acetoxy-5α-pregnan-20-one(4),pregnan-1,4-diene-3,20-dione(5),6β-hydroxypregnan-4-ene-3,20-dione(6),pregnan-1,4-diene-3-one-20-carboxylic methyl ether(7)and 3β,6α,11-trihydroxy-24-methyl-5α-cholest-9,11-seco-7-en-9-one(8),respectively.Conclusion These eight compounds were obtained fromSubergorgia suberosafor the first time.

Abstract:Two new unusual cholestane derivatives, pentacyclic steroid 16,22-epoxy-20β,23S-dihydroxycholest-1-ene-3-one (1) and 20β,23S-dihydroxycholest-1-ene-3,22-dione (2), along with two new pregnane derivatives, 15β,17α-dihydroxypregna-4,6-diene-3,20-dione (3) and 11α-hydroxypregna-4-ene-3,6,20-trione (4), were isolated from the South China Sea gorgonian coral Subergorgia suberosa. Their structures were established based on the extensive analyses of 2D NMR, IR, and HRMS. Antifouling tests against Balanus amphitrite larvae settlement indicated that 1 and 2 exhibited inhibitory effect with EC50 values of 5.3, and 14.5 μg/mL, respectively.

Abstract:Two new pentahydroxylated steroids, (3β,12β,16β,23E)‐cholesta‐5,23‐diene‐3,12,16,20,25‐pentaol (1) and (3β,12β,16β)‐cholesta‐5,25(26)‐diene‐3,12,16,20,24‐pentaol (2), were isolated from the EtOH extract of the South China Sea gorgonian Subergorgia suberosa. In addition, four known steroids were detected. The structures of these compounds were established by spectroscopic methods and comparison of their data with those of the related known compounds. In addition, the cyctotoxicities of the compounds were evaluated against selected cancer cell lines.

Abstract:OBJECTIVE To isolate and identify the chemical constituents in gorgonian Verrucella sp.METHODS Chromatography and recrystallization were performed for the isolation and purification,the structures were determined by extensive spectroscopic analysis in association with physical and chemical properties.RESULTS Seven compounds were isolated from the CH3COOCH2CH3 extracts of gorgonian Verrucella sp.,their structures were identified as junceellin(1),praelolide(2),cholesterol(3),(E)-N-2-(1,3-dihydroxy octadecan-4-en)-hexadecamide(4),thymine(5),thymidine(6) and batyl alcohol(7).CONCLUSION All the compounds were isolated from the gorgonian for the first time.

Abstract:Four new 9,10-secosteroids, verrucellols A–D (1–4), together with 12 known derivatives (5–16) were isolated from the gorgonian Verrucella umbraculum collected in the South China Sea. The structures of the new compounds were established by spectroscopic analysis and comparison with reported data. These compounds exhibited significant suppressive effects on CD4+ T lymphocyte cell differentiation in an in vitro bioassay. Abstract:Six sterols named Cholesterol (Ⅰ), Cholesta 5 ene 3β,7β diol (Ⅱ), Cholesta 5,22(E)-dien 3β,7α diol (Ⅲ), 24 methylene-cholesta-5-en 3β, 7α diol (Ⅳ), 24-methyl-cholesta 5,22(E) 3β, 7α diol (Ⅴ), Cholesta-5-ene 3β, 7α diol (Ⅵ), were isolated from the soft coral Dendronephthya gigantea collected from the South China Sea. Their structures were identified by EIMS, 1 H NMR and 13 C NMR analysis and the spectral data were compared with those reported in literatures. The compound Ⅰ～Ⅵ are isolated from the soft coral for the first time, and the NMR data of compounds Ⅱto Ⅵwere concluded fully by extensive 2D NMR spectrum.

Abstract:Objective To investigate the chemical constituents of gorgonian Muricella abnormalis.Methods Column chromatography on silica gel and Sephadex LH-20,and semi-preparative HPLC were used to isolate and purify the compounds.The structures of compounds were identified by spectroscopic analysis including NMR and MS,and by comparison with the data of literature.The bioactivities of the isolated compounds were evaluated with brine shrimp lethality and zebrafish toxicity models.Results Six compounds were isolated from gorgonian M.abnormalis and their structures were identified as 1-formyl-4-methyl-7-isopropylazulene (1),3-guaiazulenecarbaldehyde(2),ketolactone(3),cholesterol(4),chimyl alcohol(5),and n-octodecyl alcohol(6).Conclusion Compound 2 was a new natural compound,and the other compounds were obtained from the species M.abnormalis for the first time.Compound 2 exhibited high brine shrimp lethality and strong zebrafish embryo toxicity.

Abstract:Chemical investigation of the ethanol extract of soft coral Sinularia sp. collected from the South China Sea led to the isolation of three new polyoxygenated sterols, (3S,23R,24S)-ergost-5-ene-3 beta,23 alpha,25-triol (1), (24S)-ergostane-6-acetate-3 beta,5 alpha,6 beta,25-tetraol (2), (24S)-ergostane-6-acetate-3 beta,6 beta,12 beta,25-tetraol (3) together with three known ones (4-6). The structures, including relative configurations of the new compounds (1-3), were elucidated by detailed analysis of spectroscopic data (IR, UV, NMR, MS) and by comparison with related reported compounds. The absolute configuration of 1 was further determined by modified Mosher's method. Compound 5 exhibited moderate cytotoxicity against K562 cell line with an IC50 value of 3.18 mu M, but also displayed strong lethality toward the brine shrimp Artemia salina with a LC50 value of 0.96 mu M.

Abstract:Two polyhydroxylated steroids have been isolated from the South China Sea soft coral Sinularia microclavata, and their structures were established as 24-methylenecholestane-1 alpha,3 beta,5 alpha,6 beta-tetrol and 1 alpha,3 beta,5 alpha-trihydroxy-24-methylenecholestan-6-one from spectral evidence and from comparison with two reference compounds, numersterol A and 24-methylenecholestane-1 alpha,3 beta,5 alpha,6 beta, 25-pentol, which were isolated from the soft corals, Simularia numerosa and Sarcophyton glaucum, respectively.
