Abstract:Twelve new 9,10-secosteroids designated as subergorgiaols A-L (1-12), along with four known analogues (13-16), were isolated from the gorgonian Subergorgia rubra collected from the South China Sea. Their planar structures and the relative configurations were elucidated by comprehensive spectroscopic methods including NOESY spectra. The absolute configuration of 1 was established by a dimolybdenum tetraacetate [Mo2(AcO)4] induced circular dichroism (ICD) procedure and the modified Mosher's method. Compounds 1-12 represent the first series of 9,10-secosteroids characterized with a hydroxy group at C-8, which are 8-OH derivatives of astrogorgiadiols/calicoferols. Compound 4 exhibited cytotoxicity against the cervical carcinoma cell line (CaSki) with an IC50 value of 2.4 μM, and 6 showed toxicity toward brine shrimp Artemia salina with an LC50 value of 2.0 μM.

Abstract:Five new 9,11-secosteroids 1, 2, and 4-6, and seven known analogs, 3 and 7-12, with the same steroid skeleton, (5aH)-3 beta, 6 alpha, 11-trihydroxy-9,11-secocholest-7-en-9-one, were isolated from the South China Sea gorgonian Subergorgia suberosa. Among them, 2/3 and 4/5 are C(24)-epimeric mixtures, and 6/7 is an (E)/(Z) mixture of (C(24) C(28)). Their structures and relative configurations were elucidated by using comprehensive spectroscopic methods including NOESY spectra. The absolute configuration of the steroidal nucleus was established by the modified Mosher method applied to 10 and on the basis of a common biogenesis for all of these compounds. All isolated compounds, 1-12, and five synthetic acetylated derivatives, 12a-12e, were evaluated for their cytotoxicities in vitro. Compounds 4/5, 11, 12, and 12b-12d showed cytotoxic activities against K562 cell line with the IC50 values ranging from 1.09 to 8.12 mu m.

Abstract:Five new suberosanone-purine hybrids, namely subergorgines A–E (1–5), were isolated from the South China Sea gorgonian Subergorgia suberosa. Their structures were elucidated on the basis of extensive spectroscopic data and the absolute configurations were clarified by the theoretical ECD calculation. Compounds 1–5 were rare purine alkaloids merged with the same suberosanone moiety via different C (6)–N bridges. Cytotoxic activities of the isolates were tested. Compound 4 was found to be the most active against the HL-60 cancer cell line with an IC50 value of 14.3 μM. A plausible biosynthetic pathway for suberosanone-purine hybrids was also discussed.

Abstract:A repeated silica gel column chromatography followed by HPLC purification on the methanol extract of marine soft coral Sinularia dissecta, resulted in the isolation of fifteen polyhydroxylated steroids (1–15), involving six new C-18 functionalized steroids, 3β-acetoxy-1α,11α-dihydroxygorgost-5-en-18-oic acid (1), gorgost-5-en-1α,3β,11α,18-tetrol (2), 18-acetoxy-1α,3β,11α-trihydroxygorgost-5-ene (3), 24(S)-3β-acetoxy-1α, 11α-dihydroxyergost-5-en-18-oic acid (4), 24(S)-ergost-5-en-1α,3β,11α,18-tetrol (5), and dissectolide (6). The structures of the new compounds were determined on the basis of extensive spectroscopic data (IR, MS, 1H and 13C NMR, HMQC, HMBC, and NOESY) analysis. The structure of the new compound was elucidated by 1-D and 2-D NMR, and mass spectrometry. The absolute configuration of 1 was assigned on the basis of the absolute configurations of the related congeners 2 and 3, which were determined by application of the modified Mosher's method. The isolated compounds were assayed for their cytotoxicity, antimicrobial activity and lethality towards brine shrimp Artemia salina.

Abstract:Ten new 9,11-secosteroids, subergorgols A-J (1-10), including two pairs of epimers (3/4, and 6/7), along with three known analogues (11-13) were isolated from the South China Sea gorgonian Subergorgia suberosa. The structures of these compounds were elucidated on the basis of extensive spectroscopic analyses and comparison with the literature data. The cytotoxicities in vitro against three selected human tumor cell lines for all the compounds were evaluated. Compound 9 showed significant cytotoxicities toward both K562 and MDA-MB-231 cell lines with IC50 values of 5.5 and 6.2 mu M, respectively, and compound 10 also showed inhibitory activity against K562 cell line with an IC50 value of 6.5 mu M. Abstract:A new 19-oxygenated steroid, 25-acetate-nebrosteroid K (1), and five known steroids (2–6), along with one known 19-hydroxy steroidal glycoside (7) were obtained from the gorgonian Dichotella gemmacea collected from the South China Sea. The structure and relative configuration of 1 were elucidated by using comprehensive spectroscopic data including NOESY spectra. Compound 1 represents the first example of 19-oxygenated steroid with a 19-oic acid methyl ester group isolated from gorgonians. All of the isolated compounds were evaluated for lethal activity to brine shrimp Artemia salina and cytotoxicity against A549 and HL-60 cell lines. Among them, 1 showed strong lethality towards A. salina.

Abstract:Four new steroids, acanthovagasteroids A-D (1-4), along with two known analogues (5, 6), were isolated from the gorgonian Acanthogorgia vagae Aurivillius from the South China Sea. The structures of the new compounds, characterized with 19-hydroxy groups, were identified on the basis of extensive spectral analysis as well as by comparison with literature data. This is the first report of 19-hydroxy steroids from a gorgonian and suggests a possible biosynthetic relationship between 19-nor and 19-hydroxy steroids.

Abstract:Seven new polyoxygenated steroids (1–7) were isolated together with seven known analogues (8–14) from the South China Sea soft coral, Sarcophyton sp. The structures of the new compounds were identified on the basis of extensive spectroscopic analysis and comparison with reported data. All the steroids are characterized with 3β,5α,6β-hydroxy moiety, displaying carbon skeletons of cholestane, ergostane, gorgostane and 23,24-dimethyl cholestane. In the in vitro bioassay, metabolites exhibited different levels of antimicrobial activity against bacterial species Escherichia coli and Bacillus megaterium, and fungal species Microbotryum violaceum and Septoria tritici. No inhibition was detected towards microalga Chlorella fusca. Preliminary structure-activity analysis suggests that the 11α-acetoxy group may increase both antibacterial and antifungal activities. The terminal-double bond and the cyclopropane moiety at the side chain may also contribute to the bioactivity.

Abstract:Fourteen new polyoxygenated steroids (6, 9, 14-18, 20-23, 25-27) having carbon skeletons of cholestane, ergostane, and 24-norcholestane, were isolated together with thirteen known analogues (1-5, 7, 8, 10-13, 19, 24) from the South China Sea gorgonian Menella kanisa. The structures of the new compounds were elucidated by detailed analysis of spectroscopic data and comparisons with reported data. This is the first report of chemical investigation on the title gorgonian. Compounds 12 and 13 were reported for the first time from natural sources. These compounds exhibited different levels of growth inhibition activity against A549 and MG-63 cell lines in bioassay in vitro. Preliminary structure-activity analysis revealed an important role of side chain in the activity. A substitution of a 5α-hydroxy or an oxidation of 6β-hydroxy to a ketone carbonyl group may decrease the activity whereas the contribution of the 1-ketone group remains uncertain.

Abstract:Two new polyhydroxylated sterols, named verumbsteroids A and B (1 and 2, resp.), along with seven known analog, 3-9, were isolated from the gorgonian Verrucella umbraculum collected from the South China Sea. The structures and relative configurations of the new compounds were elucidated by comprehensive spectroscopic investigations. The absolute configuration of 1 was determined by using modified Mosher method with the acetonide derivative of 1. Compounds 1 and 3-5 were found to show cytotoxicities against five human tumor cell lines (HL-60, K562, HeLa, A-549, and HCT-116) with the IC50 values ranging from 2.76 to 9.62 mu m.

Abstract:Three guaiane-type sesquiterpenoids (1−3) and five steroids with 3β-hydroxy-5-en-7-one moiety (4−8) were obtained from gorgonian Echinogorgia sassapo reticulata collected from the South China Sea. Among them, new compound 1 was identified as (1S,5S,8S)-8-methoxy-menelloide B by spectroscopic methods including IR, HRESIMS, 1D and 2D NMR, and ECD spectra. All compounds except for 3 were obtained for the first time from the genus Echinogorgia. The chemotaxonomic significance of these compounds was summarized.
