Abstract:Twenty-four steroid-based natural products, 9,10-secosteroids (1-10), 1,4-dien-3-one steroids (11-19), and 4-en-3-one steroids (20-24), containing varying side-chains, were isolated from the South China Sea gorgonian Muricella sibogae. The structures of one new 9,10-secosteroid, sibogol D (1), and two new 1,4-dien-3-one steroids, sibogols E and F (11 and 12), were elucidated by extensive spectroscopic analyses and by comparison with the literature data. Cytotoxicities for all the isolates were evaluated against four selected tumor cell lines, HL-60, HCT116, K562, and P388. Compounds 3, 9, and 13 exhibited potent cytotoxic activities against the HL-60 cell line, with IC50 values ranging from 1.27 to 10.05 μM, and compound 3 was also cytotoxic against HCT116 with an IC50 value of 5.8 μM. The bioassay results also indicated a potential relationship between structural patterns and activity. The newly presented series of 1,4-dien-3-one and 4-en-3-one types of steroids relating to the unique 9,10-secosteroids in biogenesis were found in this species for the first time, which is of considerable chemotaxonomic significance.

Abstract:Objective To investigate the chemical constituents of gorgonian Muricella sibogae.Methods I-solation and purification were performed by chromatography and HPLC methods.Structural identifica-tions were made by physicochemical and spectroscopic analyses,and comparison with the data of litera-tures.Results Seven compounds including five sterols and two diterpenes,cholesterol(1),24-melth-cholest-5,22-dien-3β-ol(2),Cholest-5,22-dien-3β-ol(3),24-methylenecholest-5,24(28)-diene-3β-ol (4),(Z)-stigmasta-5,24(28)-dien-3β-ol(5),13-deacetoxyl calicophirin B(6)and Ophirin(7),respec-tively,were isolated from the CH_3 OH soluble extract of M.sibogae.Conclusion AIl compounds were obtained from the species M.sibogae for the first time.

Abstract:Two new eunicellin-based diterpenoids, sibogins A (1) and B (2), along with one known analogue (3), and four known 9,10-secosteroids (4-7), were isolated from the gorgonian Muricella sibogae Nutting collected from the South China Sea. The structures of the new compounds were elucidated by comprehensive analysis of spectroscopic data, especially by using 2D NMR spectra. The antifouling activity on barnacle Balanus amphitrite and lethality towards brine shrimp Artemia salina of all the isolated compounds were evaluated.

Abstract:Bioactivity-guided isolation of the rare gorgonian Muricella sibogae (Nutting) yielded the two new eunicellin diterpenes sibogin A and B (1 and 2), the three new 9,10-secosteroids sibogol A-C (6-8), together with the three known eunicellin diterpenes 3-5 and the five known 9,10-secosteroids 9-13. Their structures were established by extensive spectral analysis (1D- and 2D-NMR, IR, and MS). The cytotoxicity of the isolates 1-13 was evaluated in vitro against the selected tumor cell lines P388 and BEL-7402. All the compounds showed only weak activity against P388 cell lines, with an inhibition rate ranging from 10 to 60% at a concentration of 50g/ml, whereas the were inactive against BEL-7402 cell lines.

The in viteo antimicrobial activities of the identified compounds were assessed by an agar diffusion test. Results: Five 5α, 8α-epidioxy sterols were isolated from the Et2O extract (1-5), and their structures were determined as: (22E, 24S)-5α, 8α-epidioxy-24-methyl-cholesta-6, 22-dien-3β-ol (1), (22E,24R)-5α, 8α-epidioxy-24-methyl-cholesta-6, 22-dien-3β-ol (2), (24R)-5α, 8α-epdioxr24-ethyl-cholesta-6-en-3β-ol (3), (22E)-5α, 8α-epidioxy-choles--6, 22-dien-3β-ol (4), and 5α, 8α-epidioxy-choles--6-en-3β-ol (5). These compounds showed different degrees of antimicrobial activities in bioassay in vitro. Conclusion: The five compounds have been identified for the first time from the gorgonian Muriceopsis flavida. Compounds 2 shows a strong anti-algae effect and is worth further studying.

Abstract:Three new guaiazulene alkaloids muriceidines A–C (1–3) and one new bis-sesquiterpene muriceidone A (4), were isolated from the South China Sea gorgonian Muriceides collaris. Muriceidines are the first examples structurally architected by guaiazulene coupling with an inner-salt Δ 1-pipecolic acid via a unique sp2 methine-bridged linkage, and the bis-sesquiterpene was comprised by a guaiazulene and a indene units linked through a unprecedented carbon-carbon σ-bond between the high steric bridgehead carbon C-10 of guaiazulene moiety and C-2′ of indene moiety. The chiral compounds 2–4 were obtained initially as racemates and further separated by chiral HPLC methods. The inner-salt structures of 1–3 and absolute configurations of 2–4 were fully elucidated by calculated 13C NMR, ECD and OR with quantum chemical calculation methods. Compound 1 showed cytotoxicity against K562 cell lines with IC50 value of 8.4 μM and antifouling activity against the larvae of the barnacle Balanus albicostatus with EC50 value of 11.9 μg/mL and potent therapeutic index (LC50/EC50 = 3.66). Also the racemic (±)-3 showed cytotoxicities against both HL-60 and K562 cell lines with IC50 values of 2.2 and 3.7 μM, respectively. A semisynthetic trial was performed to validate the proposed biosynthetic hypotheses.

Abstract:Objective To study the chemical constituents of gorgonian Muriceides Collaris.Methods Colum chromatography on Sephadex LH-20,silica gel and ODS and semi-preparative HPLC were applied for the isolation and purification of the compounds,and their structures were determined by spectroscopic analysis including NMR and MS,and by comparison with the data from literature.Results From the methanol extracts of Muriceides Collaris,ten compounds were isolated and identified as guaiazulene(1),sesquiterpene ketolactone(2),guaianediol(3),1,1'-biguaiazulene(4)、1,2'-biguaiazulene(5),phydroxyaceto phenone(6),deoxyadenosine(7),deoxyuridine(8),deoxythymidine(9)and uracil(10),respectively.Conclusion Compounds 4~6 were isolated from this coral for the first time,of which compound 5 was a new natural product.Compound 4 exhibited cytotoxicity against HeLa and HL-60cell lines at the concentration of 50(mol/L with the proliferation inhibition ratio of 16.29%and21.92%,respectively.

Abstract:A systematic re-study on gorgonian Muricella sibogae from South China Sea yielded 10 eunicellin-based diterpenoids including two new ones, sibogins C and D (1 and 2). Their structures were elucidated by extensive spectroscopic analyses (1D and 2D NMR, IR and MS) and by comparison with data reported in literatures. All the isolates were tested for cytotoxic and antifouling activities. Compounds 3 and 5 showed significant antifouling activity against the green mussel Perna viridis, and especially 3 was suggested as a potent low-toxic antifouling agent with a large LC50/EC50 value of 18.6. This was the first report on the antifouling activity of the eunicellin-type diterpenoids against the green mussel.

Abstract:Five new 9,11-secosteroids 1, 2, and 4-6, and seven known analogs, 3 and 7-12, with the same steroid skeleton, (5aH)-3 beta, 6 alpha, 11-trihydroxy-9,11-secocholest-7-en-9-one, were isolated from the South China Sea gorgonian Subergorgia suberosa. Among them, 2/3 and 4/5 are C(24)-epimeric mixtures, and 6/7 is an (E)/(Z) mixture of (C(24) C(28)). Their structures and relative configurations were elucidated by using comprehensive spectroscopic methods including NOESY spectra. The absolute configuration of the steroidal nucleus was established by the modified Mosher method applied to 10 and on the basis of a common biogenesis for all of these compounds. All isolated compounds, 1-12, and five synthetic acetylated derivatives, 12a-12e, were evaluated for their cytotoxicities in vitro. Compounds 4/5, 11, 12, and 12b-12d showed cytotoxic activities against K562 cell line with the IC50 values ranging from 1.09 to 8.12 mu m.

Abstract:Objective To study the chemical constituents of the South China Sea gorgonian Muriceopsis flavida.Methods The compounds were isolated and purified by repeated column chromatographies including normal-phase silica gel,Sephadex LH-20 and semi-preparative RP-HPLC.The structures were elucidated by the physicochemical evidences and spectra data.The in vitro bio-activities of the compounds were assessed by an agar diffusion test.Results Two sesquiterpenes were isolated and indentified as menverins C(1) and D(2).The two compounds showed different levels of antimicrobial activity in bioassay in vitro.In particular,compounds 1 and 2 displayed considerable growth inhibition toward bacterium Bacillus migaterium and the alga Chlorella fusca,respectively.Conclusions This was the first report of menverins C(1) and D(2) from the genus Muriceopsis.The antimicrobial activity of both compounds suggested a possible ecological role of these metabolites in the chemical defensive strategy of the gorgonian.

Abstract:Two new pentahydroxylated steroids, (3β,12β,16β,23E)‐cholesta‐5,23‐diene‐3,12,16,20,25‐pentaol (1) and (3β,12β,16β)‐cholesta‐5,25(26)‐diene‐3,12,16,20,24‐pentaol (2), were isolated from the EtOH extract of the South China Sea gorgonian Subergorgia suberosa. In addition, four known steroids were detected. The structures of these compounds were established by spectroscopic methods and comparison of their data with those of the related known compounds. In addition, the cyctotoxicities of the compounds were evaluated against selected cancer cell lines.

The structures of the new compounds were elucidated by detailed spectroscopic analysis in combination with comparison of reported data. All the compounds are reported for the first time from the animal. In the bioassay in vitro, these compounds exhibited different levels of growth inhibition activity against A549 and MG63 cell lines. In particular, compound 18 displayed a considerable activity, being similar as that of positive control adriamycin. An annexin V analysis indicated that compounds 7 and 18 can significantly induce apoptosis in A549 cell, and compound 7 is more potent in the induction of apoptosis. Preliminary structure–activity analysis suggests that the acetylation on 3-OH and appearance of Δ7 may decrease the activity while substitution of 1-OH and the nature of side chain may also play an important role in the activity. Methylation of 5-OH contributed a little to the activity.
