Abstract:A convenient and efficient synthetic method of the important intermediate naphthazarin is presented starting from 1, 4, 5, 8-tetramethoxynaphthalene in an overall yield of 87%. Compared with the reported synthesis, this method has several advantages. Firstly, the reaction conditions are milder; secondly, the workup of each step is simpler and the yield is considerably higher; thirdly, all the reactions involved in the method are more suitable for large-scale preparations.

Abstract:A convenient and efficient synthesis of the important naphthazarin derivative 2-acetyl-5,8-dimethoxy-1,4-naphtho-quinone is introduced in this paper. The key steps of the synthetic strategy involve the selective demethylation of 2-acetyl-1,4,5,8-tetramethoxynaphthalene with anhydrous aluminum chloride and subsequent oxidation of the demethylated product by cerium ammonium (IV) nitrate. Compared with the reported routes, the overall yield is much higher and the work-up of each step is simpler. Moreover, the reaction conditions are milder and all the reactions involved in the method are more suitable for large-scale preparations.

Abstract:2,3-Dihydronaphazarin was prepared from 1,4-dimethoxybenzene and dichloromaleic anhydride. Reaction with bromochloromethane gave 1,8:4,5-bis(methylenedioxy)naphthalene which with NBS, gave 2-bromo-1,8:4,5-bis-(methylenedioxy)naphthalene. Reaction of the Grignard derivative with DMF afforded 2-formyl-1,8:4,5-bis(methylenedioxy)naphthalene. This method had several advantages compared with the reported synthesis, including fewer steps, milder condition and higher yields.

Abstract:A convenient synthesis of 2,6-dihydroxynaphthalene from 6-bromo-2-naphthol has been achieved with high overall yield (52%) and good purity (95.7%) based on the conversion of 6-(methoxymethoxy)-2-naphthaldehyde to 6-(methoxymethoxy)-2-naphthol formate by a Baeyer–Villiger oxidation-rearrangement. Compared with the reported methods, the reaction conditions are milder and the work-up of each step is much simpler. Moreover, 6-bromo-2-naphthol as the starting material for the synthesis is readily available.

Abstract:The synthesis of 1,4,5,8-tetramethoxynaphthalene-2-carboxaldehyde,an intermediate of shikonin was investigated.1,4,5,8-tetramethoxynaphthalene was prepared from the oxidation of 1,5-dinitronaphthalene and the reaction product naphthazarin was directly methylated without been purified.The title compound was obtained by the formylation of 1,4,5,8-tetramethoxynaphthalene.The overall yield was 38.5% based on 1,5-dinitronaphthalene.
