Abstract:The synthesis of 1,4,5,8-tetramethoxynaphthalene-2-carboxaldehyde,an intermediate of shikonin was investigated.1,4,5,8-tetramethoxynaphthalene was prepared from the oxidation of 1,5-dinitronaphthalene and the reaction product naphthazarin was directly methylated without been purified.The title compound was obtained by the formylation of 1,4,5,8-tetramethoxynaphthalene.The overall yield was 38.5% based on 1,5-dinitronaphthalene.

Abstract:A convenient and concise synthesis of 1,4,5,8-tetramethoxynaphthalene can be achieved by methylation of 2,3-dihydronaphthazarin with dimethylsulfate in the presence of phase transfer catalytic in 90.3% yield, then formylation of 1,4,5,8-tetramethoxynaphthalene can afford 2-formyl-1,4,5,8-tetramethoxynaphthalene in 96.2% yield.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:A novel total synthesis of (+/-)shikonin 1 was presented. The key intermediate 6 was achieved by the formylation of 1,8:4,5-bis(methylenedioxy)naphthalene 5 with N-methylformanilide in good yield. It was first reported that the addition of prenyllithium to 2-formyl-1,8:4,5bismethylenedioxy naphthalene 6 gave 2-(1-hydroxy-4-methylpentyl) 1,8:4,5-bis(methylenedioxy) naphthalene 8 in 45% yield and 2-(1-hydroxy-2,2-dimethyl-3-butenyl)-1,8:4,5-bis(methylenedioxy)naphthalene 9 in 48% yield. After the electrooxide deprotection of 8, (+/-)shikonin 1 was prepared in high yield.

Abstract:2,3-Dihydronaphazarin was prepared from 1,4-dimethoxybenzene and dichloromaleic anhydride. Reaction with bromochloromethane gave 1,8:4,5-bis(methylenedioxy)naphthalene which with NBS, gave 2-bromo-1,8:4,5-bis-(methylenedioxy)naphthalene. Reaction of the Grignard derivative with DMF afforded 2-formyl-1,8:4,5-bis(methylenedioxy)naphthalene. This method had several advantages compared with the reported synthesis, including fewer steps, milder condition and higher yields.

Abstract:A convenient synthesis of 5, 8-O-dimethylshikonin and the 6-isomer of 5,8-O-dimethylshikonin was developed in 81.8% total yield from shikonin. (R)-4-Methyl-1-(1,4,5,8-tetramethoxynaphthalen-2-yl)pent-3-en-1-ol was prepared in a one-pot two-step approach and then oxidised with ceric ammonium nitrate. This is simpler and the overall yield is higher than previous methods. The reaction conditions are milder and more environmentally friendly to environment, avoiding the use of the toxic and expensive reagents methyl iodide and chloromethyl ether and are more applicable to large-scale preparation. This is important because shikonin and its closely related derivatives have attracted much attention in view of their antitumor activities.

Abstract:Based on the Friedel–Crafts acylation reaction, a one-step synthesis of 2-acetyl-1,4,5,8-tetramethoxynaphthalene and its selective demethylation product was carried out under various reaction conditions. The product and yield of the reaction were highly catalyst and solvent dependent. The developed method is characterized by short process time, low cost, mild conditions, simple work-out, high yield, and low chemical waste.
