Abstract:Compounds containing aryl structures are important organic synthesis intermediates,which are widely present in a large family of natural products and bioactive molecules.In recent years,direct asymmetric functionalization of benzylic C(sp3)-H bond for the efficient construction of arene and hetero-arene motifs with high stereoselectivity has drawn widespread concern from chemical community.Among the various strategies,small molecule-catalyzed metal-free functionalization of benzylic C(sp3)-H bond represents a more challenging but promising transformations.This review is intended to summarize and discuss the most recent advances in this area.

Abstract:C-H bond sulfonylation of anilines with the insertion of sulfur dioxide under metal-free conditions is described. 2-Sulfonylanilines are generated in moderate to good yields through a three-component reaction of anilines, DABCO(SO2)(2), and aryldiazonium tetrafluoroborates under mild conditions. No metal catalysts or additives are needed for this transformation. This direct C-H functionalization is highly efficient, and broad functional group tolerance is observed. A radical process is believed to be involved. In the reaction process, the arylsulfonyl radical and the tertiary amine radical cation generated in situ from DABCO(SO2)(2), and aryldiazonium tetrafluoroborate are the key intermediates. Additionally, the tertiary amine radical cation acts as the electron carrier through a single electron transfer process.

Abstract:Herein, we disclose the metal-free synthesis of arylamines via the direct amination of phenols using aminating reagents. This reaction procedure uses easy accessible aminating reagents and provides a versatile synthetic route to a broad range of arylamines with various functionalities in good to excellent yield. By using a two-step route of amination and oxidative coupling reaction, we synthesized three naturally occurring carbazole alkaloids: murrayafoline A, mukonine, and clausenine from two commercially available phenols.

Abstract:A highly selective, IBX-promoted reaction has been developed for the oxidative cleavage of inert C(aryl)-N bonds on secondary amides while leaving the C(carbonyl)-N bond unchanged. This metal-free reaction proceeds under mild conditions (HFIP/H2O, 25 degrees C), providing facile access to various useful primary amides, some of which would be otherwise unattainable using conventional aminolysis and hydrolysis approaches.

Abstract:A transition-metal-free deacylative C(sp(3))-C(sp(2)) bond cleavage for the synthetically practical oxidative amination of ketones and aldehydes to nitriles is first described, using cheap and commercially abundant NaNO2 as the oxidant and the nitrogen source. Various nitriles bearing aryl, heteroaryl, alkyl, and alkenyl groups could be smoothly obtained from ketones and aldehydes in high yields, avoiding highly toxic cyanides or transition metals.

Abstract:Regioselective functionalization of C-H bonds has attracted the attention of chemists for recent years. However, transition metal is indispensable in most of C-H activation and often expensive. Here, we describe a metal-free intramolecular heteroarene C(sp2)−H amination from unprotected arylamine. Exposure of 2-(Pyridin-3-yl)aniline to posstium tert-butoxide was discovered to access N-heterocycles without additives, which unlocks new breakthrough. This process is applicable for the synthesis of a wide variety of N-heterocycles including natural products and bioactive compounds. Mechanistic studies have established that these reactions proceed through a SNArH process. Such a unique transformation opens a door toward weak reactivity arylamine regioselective intreamolecular C-H amination with heterocyclic compounds.

Abstract:A general and straightforward method for the regioselective construction of C-3 heteroaryl-containing imidazo[1,2-a]pyridines via cross-dehydrogenative coupling under transition-metal-free conditions has been reported, utilizing N,N-dimethylaniline as the methylenation source and furnishing the C(sp(2))-C(sp(3)) functionalized products in good to excellent yields. Abstract:Catalytic ring-opening of bio-sourced non-strained lactones with aromatic amines can offer a straightforward, 100 % atom-economical, and sustainable pathway towards relevant N-aryl amide scaffolds. Herein, the first general, metal-free, and highly efficient N-aryl amide formation is reported from poorly reactive aromatic amines and non-strained lactones under mild operating conditions using an organic bicyclic guanidine catalyst. This protocol has high application potential as exemplified by the formal syntheses of drug-relevant molecules.
