Abstract:An efficient Y(OTf)(3)-catalyzed generation of azomethine ylides from donor-acceptor aziridines and their [3+2] cycloaddition with imines was developed. The method provides facile access to multisubstituted imidazolidines, which have been extensively used in organic synthesis. Furthermore, a three-component reaction on a gram scale and an asymmetric variation were also developed in this work.

Abstract:A cross 1,3-dipolar cycloaddition of two different ylides between C,N-cyclic azomethine imines with an in situ-generated nonstabilized azomethine ylide from an N-benzyl precursor was realized. The reactions afforded a clean and facile access to diverse fused tricyclic 1,2,4-hexahydrotriazines in high yields (up to 96%). The chemical structures of the typical compounds were confirmed by X-ray single-crystal structure analysis.

Abstract:AbstractIn contrast to the well‐established [3+2] cycloaddition reactions, the catalytic enantioselective [3+n] (n≥3) cycloaddition reaction of activated isocyanides for the preparation of six‐membered or larger ring systems has remained underdeveloped. Herein, we report the first example of highly diastereo‐ and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. By employing silver catalysis, a wide range of biologically important bicyclic 1,2,4‐triazines were obtained in high yields (up to 99 %) with good to excellent stereoselectivities (up to >20 : 1 dr, 99 % ee). In addition, the same catalytic system could be applied to both the late‐stage functionalization of complex bioactive molecules and the kinetic resolution of racemic azomethine imines, further highlighting its versatility and synthetic utility.

Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:An enantioselective [3+2] cycloaddition of donor-acceptor aziridines with N-aryl protected imines was developed with a Ni(ClO4)(2).6H(2)O/N,N '-dioxide catalyst system, providing a broad range of chiral trans-substituted imidazolidine compounds with good yields and excellent enantioselectivities (up to 99 % yield, up to 98 % ee). Control experiments indicated that the products could offer excellent diastereoselectivities with the control of chiral Ni(II)-N,N '-dioxide complex and the interaction of the substrates. The possible catalytic process was proposed to rationalize the stereocontrol.

Abstract:A novel Lewis acid-catalyzed [3+2] intramolecular cross-cycloaddition (IMCC) between aziridine 2,2-diesters and conjugated dienes has been developed. This is the first regiospecific IMCC of intramolecular 1,3-dipolar cycloadditions of azomethine ylides with carbon=carbon double bonds, and supplies a general and efficient strategy for construction of structurally complex and diverse aza-[n.2.1] skeletons. The [3+2]IMCC could be carried out under mild conditions and in gram scale. More importantly, 3-alkyl-substituted aziridines were also successful. The excellent structural diversity, the facile operation and the versatile post-modifications will support the applications of the [3+2]IMCC in natural products synthesis and drugs discovery.

Abstract:A diastereo- and enantioselective formal [3+2] cycloaddition of N-tosylaziridines and aldehydes catalyzed by a Ni(II)-bisoxazoline complex has been accomplished. The 1,3-oxazolidine products are obtained with high diastereoselectivity, good yields (up to 99%) and high ee values (up to 96% ee). The challenging long distant stereocenter control is achieved by a chirality transfer approach.

Abstract:The unprecedented 1,3-dipolar cycloaddition of 1,3,5-triazinanes and aziridines has been described. With readily available starting material, this method offers efficient access to a wide range of functionalized imidazolidine derivatives in moderate to good yields (up to 92%) under mild conditions. Preliminary mechanistic investigations show that the ring opened zwitterionic pathway product dominated over the second-order nucleophilic substitution-like product. This protocol is very promising because the reaction is scalable, and gives versatile transformation of the products into other functionalized imidazolidines.

Abstract:In this work, cis-2,5-disubstitutedoxazolidines were efficiently constructed via a regioselective C–C bond cleavage of N-tosylaziridine 2,2-dicaboxylates and a subsequent [3+2] cycloaddition with aromatic aldehydes in the presence of Zn(OTf)2. The reactions were highly diastereoselective to form oxazolidines in cis configurations. trans-2,5-Disubstituted imidazolidines were also diastereoselectively synthesized in the similar manner using imines as substrates and AgOTf as catalyst. Based on the detailed investigation of the substrate diversity for both reactions, including the electronic effects and the steric effects of the substituted groups on the aziridines, aldehydes, and imines, a stepwise mechanism was postulated for the diastereoselective formation of cis-2,5-disubstitutedoxazolidines and trans-2,5-disubstituted imidazolines.

Abstract:An unprecedented 1,3-dipole cycloaddition between acyclic CF3-ketimines and N-benzyl azomethine ylide has been allowed by tungsten catalysis, furnishing a range of novel imidazolidines bearing a trifluoromethylated tetrasubstituted carbon center. This reaction appears as one of rare examples that challenging acyclic CF3-ketimines have been engaged in 1,3-cycloaddition reactions. The capability for gram-scale synthesis and variant derivatizations of cycloaddition adducts illustrates the synthetic potential of this approach. This protocol provides a facile access to a rapidly enlarging pool of motifs with a trifluoromethylated fully substituted carbon.
