Abstract:AbstractIn contrast to the well‐established [3+2] cycloaddition reactions, the catalytic enantioselective [3+n] (n≥3) cycloaddition reaction of activated isocyanides for the preparation of six‐membered or larger ring systems has remained underdeveloped. Herein, we report the first example of highly diastereo‐ and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. By employing silver catalysis, a wide range of biologically important bicyclic 1,2,4‐triazines were obtained in high yields (up to 99 %) with good to excellent stereoselectivities (up to >20 : 1 dr, 99 % ee). In addition, the same catalytic system could be applied to both the late‐stage functionalization of complex bioactive molecules and the kinetic resolution of racemic azomethine imines, further highlighting its versatility and synthetic utility.

Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:An atom-economic route to ring-fused 1,2,4-hexahydrotriazines relying upon a new synthetic application of vinylaziridines in a rhodium-catalyzed intermolecular [3+3] cycloaddition of C,N-cyclic azomethine imines has been reported. Highly efficient chirality transfer in the present transformation was also observed, providing a new method for the synthesis of sp(3) -carbon-rich 1,2,4-hexahydrotriazines in an enantioselective manner.

Abstract:A novel α,β-regioselective [3+2] cycloaddition reaction of arylallene with C,N-cyclic azomethine imine is reported. The axial-to-central chirality transfer phenomenon has been disclosed with chiral allenes in the reaction. The wide substrate scope, including different functional groups and natural products, reveals the generality of the methodology. Both experiments and density functional theory calculations have been used to elucidate a plausible mechanism.

Abstract:A copper-catalyzed efficient, operationally simple, general method for straightforward syntheses of 1,2,4-triazinane derivatives employing EMCCs (ethynyl methylene cyclic carbamates) as precursors reacting with azomethine Imines was firstly reported. A wide variety of 1,2,4-triazinane derivatives were obtained in acceptable to good yields under mild conditions. This protocol features broad substrate scope, high functional group tolerance and easy operation, therefore enabling late-stage functionalization. In addition, a scale-up experiment further highlighted the synthetic utility.

Abstract:A new synthetic application of vinyl aziridines as N-containing three-atom components in a rhodium-catalyzed [4+3] cycloaddition reaction is described. The reaction proceeds well with various silyl dienol ethers and vinyl aziridines, and enables the efficient synthesis of highly functionalized azepines in an enantioselective manner with net inversion of absolute configuration. The salient features of the transformation include the use of readily available substrates, high selectivity, and mild reaction conditions, as well as the versatile functionalization of the products.

Abstract:A diastereo- and enantioselective formal [3+2] cycloaddition of N-tosylaziridines and aldehydes catalyzed by a Ni(II)-bisoxazoline complex has been accomplished. The 1,3-oxazolidine products are obtained with high diastereoselectivity, good yields (up to 99%) and high ee values (up to 96% ee). The challenging long distant stereocenter control is achieved by a chirality transfer approach.

Abstract:Various nitrogen-substituted iso(thio)cyanates engage in [3 + 2]-cycloaddition reactions to form azomethine imines containing triazolone, triazole-thione, and pyrazole-thione cores. First, iminoisothiocyanates are shown to undergo aminothiocarbonylation reactions with strained alkenes, and a comparison with recently reported reactions of iminoisocyanates highlights their reduced reactivity. In contrast, amino(thio)carbonylation reactions of imines with iminoisocyanates and iminoisothiocyanates proved more efficient, providing access to triazolone and triazole-thione cores. The dipole products can be converted to valuable heterocyclic cores through simple derivatization reactions.

Abstract:By utilizing an underexplored reaction mode of C,N-cyclic azomethine imines, a catalyst-free [1+2+3] cycloaddition/N-N bond cleavage sequential reaction for accessing spiroindolines with syn-stereoselectivity was developed. On the basis of experimental results and DFT calculations, peroxide and ethereal solvent were identified to trigger the hydrogen abstraction of the unstable [1+2+3] cycloaddition adducts, followed by homolytic cleavage of the N-N bond and hydrogen absorption.

Abstract:An unprecedented copper(II)-catalyzed enantioselective 1,3-dipolar [3+4] cycloaddition of azomethine imines with in situ formed azoalkenes has been realized. This strategy provides a facile access to biologically important 1,2,4,5-tetrazepine derivatives in high yield with exclusive regioselectivity and high stereoselectivity. Moreover, enantioenriched azomethine imines could be obtained via an efficient kinetic resolution using the same approach.

Abstract:By taking advantage of vinyl aziridines as a heteroatom-containing five-atom component in rhodium-catalyzed intramolecular formal hetero-[5 + 2] cycloaddition reactions with alkynes, a highly efficient method for the synthesis of fused azepine derivatives at 30 °C was developed. The reaction has broad substrate scope and tolerates a wide range of functional groups. The chirality of vinyl aziridine-alkyne substrates can be completely transferred to the cycloadducts, representing an atom-economic and enantiospecific protocol for the construction of fused 2,5-dihydroazepines for the first time.

Abstract:A highly efficient and stereoselective synthesis of enantioenriched imidazolidines by rhodium-catalyzed intermolecular [3 + 2] cycloaddition reaction of chiral vinyl aziridines and oxime ethers has been successfully developed. Notably, both aldoximes and ketoximes are suitable substrates to afford the corresponding chiral imidazolidines in high yields with good stereoselectivity. This transformation represents an unprecedented example that utilizes ketimine derivatives as an aza-[2C]-component in cycloadditions of vinyl aziridines.

Abstract:Reported herein is an example of highly regio‐, diastereo‐ and enantioselective Cu(I)‐catalyzed intermolecular [3+2] cycloaddition reaction of α ‐substituted iminoesters with α ‐trifluoromethyl α,β ‐unsaturated esters. This novel strategy provided a facile access to pyrrolidines with two skipped (aza)quaternary stereocenters including a CF 3 all‐carbon quaternary stereocenter. A broad substrate scope was observed and high yields (up to 94%) with excellent diastereoselectivity (up to >20 : 1 d.r .) and enantioselectivity (up to 98% ee ) were obtained.

Abstract:A novel pattern of the cleavage and reorganization of CN bond in the multicomponent reaction (MCR) of terminal alkynes or haloalkynes, carbodiimides, and benzynes is achieved for the first time to construct efficiently 2-aminoaryl alkynyl imines. The selective formation and ring-opening of the azetine intermediate with the high ring strain is essential for this reaction. Further transformation of 2-aminoaryl alkynyl imines via the Cu-catalyzed cycloisomerization is explored to provide steroselectively the bi-, tri-, and tetracyclic fused pyrrolines.

Abstract:The first asymmetric decarboxylative [4 + 3] annulation of propargylic carbamates with C, N-cyclic azomethine imines has been developed successfully by a copper- N-heterocyclic carbine system. This strategy led to a series of optically active isoquinoline-fused triazepine derivatives in good yields and with excellent enantio- and diastereoselectivities. Remarkably, Cu-allenylidene intermediates play a crucial role in this transformation.

Abstract:The first rhodium-catalyzed intermolecular [3+2] cycloaddition reaction of vinyl aziridines and allenes for the synthesis of enantioenriched functionalized pyrrolidines was realized. [3+2] cycloaddition with the proximal C=C bond of N-allenamides gave 3-methylene-pyrrolidines in high regio- and diastereoselectivity, whereas, 2-methylene-pyrrolidines were obtained as the major products by the cycloadditions of vinyl aziridines with the distal C=C bond of allenes. Use of readily available starting materials, a broad substrate scope, high selectivity, mild reaction conditions, as well as versatile functionalization of the cycloadducts make this approach very practical and attractive.

Abstract:The cycloaddition reaction involving bicyclo[1.1.0]butanes (BCBs) offers a versatile and efficient synthetic platform for producing C(sp3)-rich rigid bridged ring scaffolds, which act as phenyl bioisosteres. However, there is a scarcity of catalytic asymmetric cycloadditions of BCBs to fulfill the need for enantioenriched saturated bicycles in drug design and development. In this study, an efficient synthesis of valuable azabicyclo[2.1.1]hexanes (aza-BCHs) by an enantioselective zinc-catalyzed (3+2) cycloadditions of BCBs with imines is reported. The reaction proceeds effectively with a novel type of BCB that incorporates a 2-acyl imidazole group and a diverse array of alkynyl- and aryl-substituted imines. The target aza-BCHs, which consist of α-chiral amine fragments and two quaternary carbon centers, are efficiently synthesized with up to 94% yield and 96.5:3.5 er under mild conditions. Experimental and computational studies reveal that the reaction follows a concerted nucleophilic ring-opening mechanism of BCBs with imines. This mechanism is distinct from previous studies on Lewis acid-catalyzed cycloadditions of BCBs.
