Abstract:A mild and operationally simple protocol for the direct C-H functionalization of the 1-position of tetrahydrocarbazoles is reported. The diastereoselective oxidative coupling process is mediated by tert-butyl hypochlorite, and allows the successful generation of 1-functionalized tetrahydrocarbazoles with good to excellent yields and good functional group tolerance.

Abstract:The C-O bond cleavage for activation of alcohols is synthetically useful and practically challenging. This work describes carbazate as a new type of electrochemically activated alkylating agent derived from ubiquitous alcohols for direct functionalization of heteroarenes under mild electrolytic conditions. The simple undivided cell at low oxidative potentials with carbon/platinum electrode set-ups offers excellent substrate tolerance, affording a variety of primary, secondary and tertiary alkyl-decorated heterocycles in good chemical yields. Furthermore, the mechanism for this electrochemical deoxyalkylation reaction has been investigated.

Abstract:Herein we report an additive-free protocol for the facile synthesis of α,α-dichloroketones and α-chlorohydrins from various aryl terminal, diaryl internal, and aliphatic terminal alkynes and alkenes, respectively. The commercially available tert -butyl hypochlorite ( t BuOCl) was employed as a suitable chlorinating reagent, being accompanied by the less harmful t BuOH as the by-product. In addition, the oxygen atoms in the products came from water rather than molecular oxygen, based on the 18 O-labelling experiments. Meanwhile, the diastereoselectivity of the Z - and the corresponding E -alkenes has been compared and rationalized. Using a group of control experiments, the possible mechanisms have been proposed as the initial electrophilic chlorination of unsaturated C–C bonds in a Markovnikov-addition manner in general followed by a nucleophilic addition with water. This work simplified the oxychlorination method with a mild chlorine source and a green oxygen source under ambient conditions.

Abstract:A transition-metal- and harsh-oxidant-free strategy for diastereoselective homocoupling of benzylic α-boryl carbanions has been developed. Central to this methodology is the ability of the halogen transfer reagent to seamlessly integrate halogenation and substitution within a compatible process. Additionally, this methodology is also applicable to the homocoupling of diarylmethanes and alkylheteroarenes. Substrates bearing oxidatively sensitive functional groups were well-tolerated. Preliminary studies suggest that the hydrogen bond between two boryl groups contributes to the high diastereoselectivities.

Abstract:C2-formyl indoles are important building blocks for the organic synthesis of alkaloids and small molecules with broad biological activities. Generally, its preparation involves both direct formylation of indoles at the activated C2-position with as unstable carbonyl sources and oxidative cleavage of tedious prepared tetrahydro- β -carbolines (TH β Cs) in the harsh conditions. In continuation of our research on the oxidative functionalization of indoles, in this letter, we report that straightforward effectively oxidation of easily available 2-methylindoles bearing C3-heteroatom functional group proceeds regioselectively C2-formyl indoles in good and excellent yields employing an oxidative combination of NCS/DMSO. This methodology presents a reasonably broad substrate scope and excellent functional group tolerance, thus enabling the preparation of highly versatile building blocks susceptible to further functionalization. An additional feature of this approach includes high efficiency, ease of operation, facile purification, and short reaction time, making it a promising strategy for organic synthesis and drug discovery.

The combination of tert-butoxide bases with 2-halothiophene halogen oxidants enables the first general protocol for generating and using benzyl halides through a deprotonative pathway. In contrast to existing radical-based pathways for C–H functionalization, this process is guided by C–H acidity trends. This gives rise to new synthetic capabilities, including the ability to functionalize diverse methyl(hetero)arenes, tolerance of oxidizable and nucleophilic functional groups, precision regioselectivity for polyalkylarenes and use of a double C–H etherification process to controllably oxidize methylarenes to benzaldehydes.

Abstract:A palladium-catalyzed carbohalogenation of olefins with alkynyl oxime ethers has been described, which provides efficient and practical access to various chlorine-containing isoxazoles. This method exhibits excellent regioselectivity, good functional group compatibility, and mild reaction conditions. The mechanistic studies suggest that the reaction proceeds via a stabilized π-benzyl palladium intermediate, which is essential for the formation of C(sp3)-Cl bonds.
