Abstract:An efficient copper-mediated ortho-C(sp2)-H thiolation of aromatic amides directed by a novel directing group [4-chloro-2-(1H-pyrazol-1-yl)phenyl]amine has been developed without the need of other additives or oxidants, allowing for an increased usefulness. With the high compatibility of sterically demanding substrates, this reaction is scalable and can tolerate a wide scope of functional groups to provide alkyl and aryl thioethers in good to excellent yields (up to 93%). Furthermore, the protocol has been successfully implemented for the selenylation as well.

Abstract:A highly efficient and practical procedure for palladium-catalyzed direct thioetherification of quinolone derivatives with diaryl disulfides through decarboxylative C-S coupling has been established. The reaction could proceed smoothly under air in the presence of Pd(OAc)2 and Ag2 CO3 in DMSO. This protocol provides an appealing alternative to existing approaches to construct aryl sulfides of quinolone derivatives, which may be used as key intermediates in the synthesis of drug candidates.

Abstract:An iron‐catalyzed direct C3‐H radical trifluoromethylthiolation of quinoxalin‐2(1H)‐ones has been developed. This protocol use earth‐abundant FeSO4·7H2O as the catalyst, and AgSCF3 as the trifluoromethylthiolating reagent to provide various SCF3‐containing quinoxalin‐2(1H)‐one derivatives in 26‐88% yields. Preliminary mechanistic investigations indicate that the reaction proceeds through a radical pathway.

Here, we report a straightforward strategy to prepare electrophilic S-methyl-d3 arylsulfonothioates in one-step without column purification. These reagents exhibit good radical reactivity toward silver-catalyzed vicinal thiosulfonylation of alkenes or 1,6-enynes on water. As a result, simultaneous incorporation of both SCD3 and ArSO2 units into unsaturated carbon-carbon bonds with 100% atom economy has been established. Moreover, the ATRA adducts with >99% D incorporation can serve as nucleophilic d3-methylthiolating synthons via retro-Michael addition under mild basic conditions, providing a good alternative in trideuteromethylthiolating alkyl iodides to access corresponding trideuteromethyl sulfides with high efficiency.

Abstract:A two-component activation system that combines metal catalysis (AgOTf) and the employment of catalytic amount of organocatalyst (PPh3) has been successfully employed in the three-component reaction of 2-alkynylbenzaldehyde, amine, and α,β-unsaturated ketone. This reaction proceeds smoothly in THF under mild conditions leading to the functionalized 1,2-dihydroisoquinolines in moderate to good yields.
