Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:A highly efficient palladium-catalyzed cascade reaction of propargyl carboxylates bearing a furanyl group with organoborons was developed. This methodology offers rapid access to polycyclic Diels-Alder cycloadducts in good to high yields. The thus-formed oxygen-bridged products were further converted into anthracene derivatives in a chemoselective manner under mild conditions.

Abstract:Synthesis of the cis-fused Δ5,6-hexahydroisoindol-1-one core of cytochalasins B2-B5, K, Z8, Z9, Z12-Z15, and Z17 has been established starting from an intramolecular Diels-Alder reaction of the amide-tethered (8 E)-1,3,8-nonatriene. The trans-fused 5/6-bicyclic adduct was then subjected to highly stereoselective C9-β-hydroxylation and epimerization of the C7-α-OH group.

Abstract:Abstract A novel tandem reaction by combination of Diels–Alder [4+2] cycloaddition and [3+2] IMCC (intramolecular cross-cycloaddition) of dienylcyclopropane 1,1-diesters has been successfully developed. In this reaction, three new rings and four new stereocenters were generated in one-pot. This supplies a strategy for rapid construction of 6,6- and 6,7-fused carbocyclic skeletons which are common cores in many biologically important natural products.

Abstract:The Diels-Alder (D-A) reaction is one of the most powerful reactions in organic synthesis. The intermolecular D-A reaction attracts much less attentions than its intramolecular counterpart in natural product synthesis, possibly due to the issues of reactivity and selectivity. In the past decade, the intermolecular D-A reaction has been increasingly utilized in the total synthesis of structurally complex natural products. In this article, we present a few examples for the elegant applications of the intermolecular D-A reaction that are inspired by biosynthetic hypotheses of the target natural products. These examples demonstrate that D-A reaction is not only useful for preparing building blocks but also powerful for coupling structurally complicated and sterically demanding segments in a highly chemo- and stereo-controlled fashion, which may inspire further developments of intermolecular D-A reaction from both strategy and methodology perspectives.
