Abstract:Self-assembled catalyst: A new kind of chiral supramolecular organocatalyst, self-assembled from chiral amines and poly(alkene glycol)s, was developed. The resulting self-assemblies were found to be highly efficient in the asymmetric catalysis of the unusual Diels–Alder reaction between cyclohexenones and nitrodienes, nitroenynes or nitroolefins, giving excellent chemo-, regio- and enantioselectivities (see scheme). Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:A regio-, diastereo- and enantioselective 1,3-dipolar cycloaddition reaction between nitrones and electron-deficient alkenes has been developed employing C 1 -symmetrical Ti(OTos) 2 - and TiCl 2 -TADDOLate as catalysts. In the presence of powdered 4 Å molecular sieves and 100 mol% of Ti(OTos) 2 -TADDOLate as the catalyst, the nitrones smoothly react with alkenes at room temperature to give isoxazolidines in high conversion, giving the trans-diastereomer of the isoxazolidines with de's up to >90% and ee up to 58%. Furthermore, C 1 -symmetrical TiCl 2 -TADDOLate has a better chiral induction, giving the trans-diastereomer of the isoxazolidines with high enantioselectivity up to 70% ee. The influence of catalyst amount, temperature and solvents on the reaction course has been investigated

Abstract:The Diels-Alder reaction stands as one of the most pivotal transformations in organic chemistry. Its efficiency, marked by the formation of two carbon-carbon bonds and up to four new stereocenters in a single step, underscores its versatility and indispensability in synthesizing natural products and pharmaceuticals. The most significant stereoselectivity feature is the "endo rule". While this rule underpins the predictability of the stereochemical outcomes, it also underscores the challenges in achieving the opposite diastereoselectivity, making the exo-Diels-Alder reactions often considered outliers. This review delves into recent examples of exo-Diels-Alder reactions, shedding light on the factors inverting the intrinsic tendency. We explore the roles of steric, electrostatic, and orbital interactions, as well as thermodynamic equilibriums in influencing exo/endo selectivity. Furthermore, we illustrate strategies to manipulate these factors, employing approaches such as bulky substituents, s-cis conformations, transient structural constraints, and innovative control physics. Through these analyses, our aim is to provide a comprehensive understanding of how to predict and design exo-Diels-Alder reactions, paving the way for new diastereoselective catalyst systems and expanding the chemical scope of Diels-Alder reactions.

Abstract:Dearomative Diels‐Alder cycloadditions between nitroarenes and 2‐trimethylsilyloxycyclohexadiene are carried out under high pressure at room temperature in the absence of any chemical promoter. Reactions are performed with different arenes, including the highly aromatic naphthalenes and quinolines. They lead to 3D‐scaffolds with exquisite exo‐diastereoselectivity. The exo approach is characterized by lower distortion of the substrates in a late TS and by more favorable orbital interactions presumably between the nitro group and the dienic part, explaining the stereoselectivity

Abstract:Catalytic asymmetric Diels−Alder reaction is one of the most powerful reactions in organic chemistry. It is still a challenge to achieve high and general exo-selectivity. A novel bispyrrolidine diboronate compound has been derived from the reaction of two molecules of oxazaborolidine with one molecule of water. Upon activation, it effectively catalyzes (0.4-5 mol % loading) the Diels–Alder reaction between a variety of mono- and di-carbonyl activated dienophiles and dienes with better than 20:1 regioselectivity, up to >99:1 enantioselectivity, and better than 20:1 exo/endo selectivity. Mechanistic studies show a remarkable nonlinearity in enantioselectivity, and a second-order kinetics with respect to the catalyst concentration. A model is proposed to rationalize the above observations. The reaction is expected to find wide applications in organic synthesis.

Abstract:AbstractThe enantioselective aza‐Diels–Alder reaction is efficient for constructing chiral tetrahydropyridines, but the catalytic asymmetric aza‐Diels–Alder reaction of ketimines with unactivated dienes is still a challenging topic. Herein, guided by computational screening, a highly enantioselective aza‐Diels–Alder reaction of 2‐aryl‐3H‐indol‐3‐ones with unactivated dienes was realized by using a B(C6F5)3/chiral phosphoric acid catalyst system under mild conditions. The reaction has a broad scope with respect to both aza‐Diels–Alder reaction partners and hence offers rapid access to an array of tetrahydropyridine derivatives with pretty outcomes (up to 99 % yield, >20:1 dr and 98:2 er). The reaction is very efficient: lowering catalyst loadings for the model reaction to 0.1 mol %, enantioselectivity is still maintained. The synthetic utility was confirmed by transformations of the products. DFT calculations provide convincing evidence for the interpretation of stereoselection.

Abstract:The cinnamates having an ortho-formyl group can potentially form vinylogous iminium ion species under the catalysis of chiral amines, which facilitates the Diels–Alder cycloaddition reaction with the concurrently generated trienamines between dienals and amine catalysts in a regioselectivity umpolung manner.
