Abstract:Six novel dihydrobenzofuran and benzofuran neolignans compounds(1～6) were synthesized by knoevenagel reactiton,esterfication,biomimetic oxidative coupling,DDQ dehydrogenation,reduction and hydrolysis with 4-hydroxybenzaldehyde and malonic acid as starting material.The Ag2O-Catalyzed biomimetic oxidative coupling of methyl 4-hydroxy cinnamate into its dimmer(1) and DDQ dehydregenation reaction were the key steps of the synthetic route.DDQ dehydrogenation reaction made the transformation from dihydrobenzofuran to benzofuran ring structure succesfully.This offered a new method to synthesize benzofuran neolignans compounds.

Abstract:Benzofurans compound 2-(3',4'-dihydroxyphenyl)-3-methoxy carbonyl-5-methoxy carbonyl vinyl-7-hydroxy-2,3-dihydrobenzofuran (1)and benzodioxanes compound 2-(3',4'-dihydroxyphenyl)-3-me-thoxy carbonyl-6-methoxy carbonyl vinyl-2,3-dihydro-1,4-benzodioxane (2)were synthesized from caffeic acid through esterification and biomimetic oxidative coupling reactions.Moreover,a series of benzofuran-neolignan compounds 3~7 and benzodioxaneneolignan compounds 8~10 were synthesized from compounds 1 and 2 respectively through acetylation,DDQ oxydehydrogenation,Pd/C catalytic hydrogenation,lithium aluminium hydride reduction and deacetylation in alkaline condition.All of these synthesized compounds were confirmed with MS,IR,1 H NMR and 13 C NMR spectra.Among them,5~7,9 and 10 are new com-pounds.8 is the natural product isoamericanol A.The biological activities of benzofuranneolignan com-pounds 1 ,3~5 against five human cancer cell lines were evaluated in the standard MTT method,and the results have shown that compounds1,3,4 and 5 exhibit good inhibitory effect on leukemia cells (HL-60), lung carcinoma cell (A-549),breast cancer cell (MCF-7),colon cancer cell (SW-480),and hepatoma car-cinoma cell (SMMC-7 7 2 1 ).

Abstract:The natural benzodihydrofuran neolignans licarin A (1) was synthesized from isoeugenol through biomimetic oxidative coupling. Then licarin A (1) was transformed into another benzodihydrofuran compound 2-(3'-methoxy-4'-hydroxy)phenyl-3-methyl-5-formylvinyl-7-methoxy-2,3-benzodihydrofuran (2) through 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) oxidative reaction. Based on Mannich reaction of 1 or 2 with viarious secondary amines and formaldehyde in acidic alcohol system under microwave-assistanced, eleven new benzodihydrofuran neolignan Mannich base derivatives 3 similar to 13 were synthesized. All the synthetic compounds were tested for antiproliferation activity against human cervical carcinoma Hela cells by the standard CCK-8 method. The results showed that most of the target compounds exhibited moderate to potent antiproliferation activity against Heala cells. Among them, compound 2 (IC(50)0.438 mu mol/L) and compound 9 (IC(50)8.358 mu mol/L) displayed the strongest antiproliferation activity against Hela cells.

Abstract:Eleven 8-5' neolignan compounds 3a similar to 9b have been synthesized via reactions of dihydrobenzofuran compounds I and 2 with dry K2CO3 or 10% NaOH (aq.) under distinct conditions. 1a and 1b were synthesized from methyl 4-hydroxy cinnamate and methyl ferulate respectively by Ag2O-catalyzed biomimetic oxidative coupling reaction while 2a and 2b were obtained through methylation of la and 1b, respectively. Synthesis of compounds 3a similar to 9b not only achieved transformation successfully from dihydrobenzofuran neolignans to 8-5' neolignans, but also offered a new method for the synthesis of stilbenoids. The effect of 8-electron-withdrawing substituents such as COOCH3 group made transformation smoothly from dihydrobenzofuren neolignans to 8-5' neolignan compounds. All of these synthesized compounds have been confirmed by MS, IR, H-1 NMR and C-13 NMR techniques.

Abstract:Although various methods for the preparation of dibenzofurans are available, an efficient, general synthesis of dibenzofurans from benzofurans remains an unsolved problem. Herein, we provide the first benzannulation of readily prepared 2‐nitrobenzofurans with alkylidene malononitriles. This methodology enables facile assembly of a wide variety of highly functionalized dibenzofurans in moderate to excellent yields under metal‐free conditions. Gram‐scale synthesis and further elaborations of the product were succesfully performed, demonstrating synthetic utility of this method.

Abstract:5-Arylbenzofuran neolignans, a newfound class of natural products, were reported to possess several kinds of pharmacological activities. To solve the lack of natural sources and promote the research of 5-arylbenzofuran neolignans in all fields, an available semi-synthesis methodology of 5-arylbenzofuran neolignans was developed, and a detailed structural modification was conducted. In the meantime, a one-pot process of Waker-type cyclization and Wacker-type oxidation was developed. To explore the potential of 5-arylbenzofuran neolignans as bioactive substances, 5-arylbenzofuran neolignans and their derivatives were evaluated for their cytotoxicity. As a result, a preliminary structure-activity relationship was obtained. Most derivatives revealed low cytotoxic effects suggesting that they were relatively safer than the natural 5-arylbenzofuran neolignan. Several derivatives showed high cytotoxicities which were found to be closely associated with apoptosis-inducing. The selectivity assay for cytotoxicity showed tumor cells were more sensitive to the promising compounds than normal cells.

Abstract:A series of 10 related benzofuran neolignan derivatives were obtained by utilising a biomimetic reaction sequence involving oxidative dimerisation of methyl ferulate, followed by derivatisation reactions. The structures of the new compounds were confirmed by 1H NMR, elemental analysis, MS and IR. All compounds were evaluated for their cytotoxic potential against five human cancer cell lines (HL-60, SMMC-7721, A-549, SK-BR-3 and PAN-1) by the standard MTT method. The results showed that most of them exhibit potent cytotoxicity.

Abstract:Three neolignans, (+/-)-demethylnitidanin(1), herpetol(2) and salvinal(3) were synthesized from methyl 4,5-dihydroxy-3-methoxy cinnamate or methyl 4-hydroxy-3-methoxy cinnamate with Ag2O-catalyzed biomimetic oxidative coupling as the key step of the synthetic route. Five novel benzofuran neolignan-4'-O-beta-D-glycosides(4-8) were achieved by the glycosylation reactions of benzofuran neolignans 2, 3 and 15. The structures of all the compounds synthesized were determined by MS, H-1 NMR and IR spectra. (+/-)-Demethylnitidanin(1) and herpetol(2) were the first synthesized and the synthesis of salvinal(3) was efficiently improved by the new synthetic routes.

Abstract:Abstract Six new benzofuran neolignans (1–6) were isolated from the EtOAc-soluble fraction of the aerial part of Aristolochia fordiana, together with twelve known analogues (7–18). The structures of the isolated compounds were elucidated by spectroscopic methods. All isolates were evaluated for their inhibitory effects on nitric oxide production in lipopolysaccaride-activated RAW264.7 macrophages and for their cytotoxic activities on three human cancer cell lines. Compound 2 showed significant nitric oxide production inhibitory activity with an IC50 value of 10.00 µM, while compound 16 exhibited cytotoxic activity with an IC50 value of 11.9 µM against MG-63 and compound 18 of 9.15 µM against HepG2 cell lines, respectively.
