Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:Herein, a novel strategy for the synthesis of various heterocyclic fused 2-aminoquinolines via palladium-catalyzed tandem cyclization of o-alkynylanilines with isocyanides has been developed. This process includes trans-oxy/aminopalladation, isocyanide insertion, elimination and 1,3-hydrogen migration. Besides high atom and step economy, this method shows good functional group compatibility with excellent chemo- and regioselectivities under mild reaction conditions.

Abstract:Quinazoline‐2,4(1H,3H)‐diones are core structural subunits frequently found in many biologically important compounds. The reaction of 2‐aminobenzonitrile and CO2, which was frequently studied, only provided N3‐unsubstituted quinazoline‐2,4(1H,3H)‐dione compounds. Herein we report palladium‐catalyzed cyclization reactions of o‐haloanilines, CO2 and isocyanides to prepare N3‐substituted quinazoline‐2,4(1H,3H)‐diones. Electron‐rich o‐bromoanilines participated in the cyclization reaction using Cs2CO3 at high temperature, and electron‐deficient o‐bromoaniline or o‐iodoaniline substrates conducted the reaction using CsF as base to deliver corresponding quinazoline‐2,4(1H,3H)‐dione products in good yields.

Abstract:A facile and direct intramolecular oxidative C(aryl)-H amidation reaction was developed for the synthesis of quinolino[3,4-b]quinoxalin-6(5H)-ones in moderate to excellent yields starting from readily available materials by tethering the adjacent N-methoxyamide and aryl portions in the presence of phenyliodine(III) bis(trifluoroacetate) at room temperature. This metal-free approach is a valuable addition to the traditional methods already available for the preparation of these molecules.

Abstract:A simple and expedient approach for the synthesis of functionalized 2-aminoquinolines via palladium-catalyzed annulation of N-acyl- o-alkynylanilines with isocyanides has been developed with high atom economy, in which an unconventional 6- endo-dig cyclization process is observed. Further investigations of the mechanism demonstrated that an intramolecular acyl migration of the N-protecting groups was involved in this transformation.

Abstract:An efficient strategy for synthesis of isoquinolines via Pd(II)-catalyzed cyclization reaction of oximes with vinyl azides or homocoupling of oximes is reported. Oximes could serve as a directing group and an internal oxidant in the transformation. This reaction features good functional group tolerance and provides a useful protocol for the synthesis of different kinds of isoquinolines under mild conditions. Some control experiments and (15)N isotope labeling experiments were conducted for the mechanistic research.

Abstract:Herein, we report a direct method for palladium-catalyzed coordination-induced oxidative remote C-H aryl etherification of 8-amidoquinolines with p-benzoquinone monoacetal. The method provides access to C5-aryl etherified quinolines and shows site-selectivity different from that of typical palladium-catalyzed C(sp2)-H activation reactions. The p-benzoquinone monoacetals act both as oxidants and as aryl etherification reagents. By using proper substrates, C5-aminated quinolines and C4-etherifed 1-naphthylamine could also be formed.
