Abstract:Copper(I) catalysed azide-alkyne cycloaddition reactions (CuAAC) occur smoothly in liquid ammonia (LNH3) at room temperature to give exclusively 1,4-substituted 1,2,3-triazoles with excellent yields (up to 99%). The CuAAC reactions in liquid ammonia require relatively small amounts of copper(I) catalyst (0.5 mole%) compared with that in conventional solvents. The product can be obtained conveniently by simply evaporation of ammonia, indicating its potential application in industry. The rate of the CuAAC reaction in liquid ammonia shows a second order dependence on the copper(I) concentration and the reaction occurs only with terminal alkynes. Deuterium exchange experiments with phenyl acetylene-d(1) show that the acidity of the alkyne is increased at least 1000-fold with catalytic amounts of copper(I) in liquid ammonia. The mechanism of the CuAAC reaction in liquid ammonia is discussed.

Abstract:Although copper-catalyzed amination of activated C(sp3)-H bonds through radical relay has been developed, amination of unactivated C(sp3)-H bonds is rare. Herein, copper-catalyzed intramolecular amination of remote unactivated C(sp3)-H bonds is reported. The reaction is conducted in a mild and effective manner with moderate to good yields, demonstrating broad tolerance toward various functional groups and exhibiting complete regio- and chemoselectivities. This innovation supplies novel synthetic pathways for the construction of saturated nitrogenated heterocycles.

Abstract:Anilines are ubiquitous in bio‐active compounds and their synthesis can be achieved via metal‐catalyzed cross‐coupling reactions involving aryl halides. We describe an unusual, yet simple, CuII‐catalyzed system for the amination of aryl chlorides in pure aqueous ammonia with 2.5 mol% catalyst loading under non‐inert conditions. Different from previous systems, the reaction proceeds even without an additional organic solvent. Copper(II) sulfate in combination with 4,7‐dimethoxy‐1,10‐phenanthroline enabled the amination of several aryl chlorides containing electron‐neutral, ‐donating and ‐withdrawing groups to the corresponding anilines with good to excellent yields. The upscaling potential of the procedure has been shown by the synthesis at 50 mmol scale. The reaction proceeds as one of the rare cases of a CuII‐assisted coupling, in contrast to the typical CuI―CuIII intermediates postulated for most Ullmann‐type coupling reactions. The copper(II) center allows for a nucleophilic substitution pathway, enabled by the deprotonation of coordinated ammonia.

Abstract:The amination of aryl halides in liquid ammonia (LNH3) is catalyzed by a copper(I) salt/ascorbate system to yield primary aromatic amines in good to excellent yields. The low concentrations of catalyst required and the ease of product isolation suggest that this process has potential industrial applications. Commonly used ligands for analogous metal-catalyzed reactions are not effective. The rate of amination of iodobenzene in liquid ammonia is first order in copper(I) catalyst concentration. The small Hammett rho = 0.49 for the amination of 4-substituted iodobenzenes in liquid ammonia at 25 degrees C indicates that the C-I bond is not significantly broken in the transition state structure and that there is a small generation of negative charge in the aryl ring, which is compatible with the oxidative addition of the copper ion being rate limiting.

Abstract:A binuclear Cu(II) complex is prepared by reacting copper(II) chloride with diethylenetriamine and ammonium oxalate. This compound displays high activity catalytic performance in the CuAAC reaction for the synthesis of triazoles (yields: 71‐99%) in aquatic medium, and A3‐coupling reaction for the producing of propargylamines (yields: 77‐96%) in solvent‐free conditions. Further investigations show that this catalyst could reuse five times without a significant reduction in its activity for the both CuAAC and A3‐coupling reactions. A binuclear copper (II) complex [Cu2(dien)2(ox)(ClO4)] (dien = diethylenetriamine and ox = oxalate) was readily synthesized and obtained in the perchlorate salt form. The structure of the compound has been determined using X‐ray crystallography, UV–Vis spectroscopy, and infrared techniques. This new catalyst has effectively facilitated the production of 1,2,3‐triazole derivatives by "Click reaction" in an aqueous environment and propargylamine derivatives by "A3‐coupling reaction" under solvent‐free conditions, resulting in yields ranging from good to excellent. Additionally, subsequent investigations have revealed that this catalytic system can be recovered and reused for up to five cycles in both cycloaddition and coupling reactions.

Abstract:In this article, a combination of Cu(OAc)2·H2O/2-aminophenol was developed as a highly efficient and controlling catalytic system for sulfonyl azids involved CuAAC. By using this catalytic system, sulfonyl azids reacted with normal alkynes or propynoates to selectively give the ring products or the chain products, respectively, in excellent yields within minutes. HOAc in situ produced in the reaction has been proved to be a super protonation reagent, by which the unstable intermediate 5-cuprated 1,2,3-triazole was protonated efficiently to yield ring-product 1-sulfonyl 1,2,3-trizoles. The control experiments also proved that 2-aminophenol played dual roles as both ligand and reductant, which led to the cheap and chemically stable Cu(OAc)2·H2O being an efficient copper source for our purpose.
