Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:An efficient methodology for the synthesis of dibenzo[a,c]carbazoles via annulation of 2,3-diphenyl-1H-indoles in EtOH under UV light irradiation (lambda = 365 nm) along with hydrogen evolution is described. This method exhibits the advantages of mild reaction conditions, no requirement of any oxidants and catalysts, and release of hydrogen as the only byproduct. Notably, the mechanism investigation confirms that the trans-4b,8a-dihydro-9H-dibenzo[a,c]carbazole intermediate could convert into cis-4b,8a-dihydro-9H-dibenzo[a,c]carbazole, which relies on the nitrogen atom of the indole ring. This is followed by intramolecular dehydrogenation which yields the dibenzo[a,c]carbazoles.

Abstract:Tunable catalytic annulation reactions of βalkynyl ketones with indoles have been developed, enabling multiple chemical bond‐forming events to selectively access skeletally diverse indole‐containing heterocycles with generally good yields. Silver‐catalyzed intermolecular benzannulation reaction of β‐alkynyl ketones with indoles afforded tetracyclic benzo[b]carbazoles whereas isochromen‐1‐yl‐substituted indoles could be obtained using the same silver catalysis by lowering the reaction temperature (0 oC or rt). Interestingly, using Sc(OTf)3 and AgOTf as a combined catalytic system led to the formation of C3‐naphthylated indoles via intramolecular benzannulation reaction

Abstract:Polysubstituted carbazoles were efficiently synthesized through direct benzannulation reaction between 1,4‐dicarbonyl compounds and indoles with catalytic amount of inexpensive zirconium(IV) chloride. This transformation proved to be regioselective and furnishes 1,4‐disubstituted and 1,2,4‐trisubstituted carbazoles with yields ranging from 26% to 91% and broad substrate scope. Moreover, this protocol benefits from using readily accessible starting materials without the need of their pre‐functionalization. The synthetic utility of the products was exemplified by functionalization of an iodocarbazole by means of Suzuki‐Miyaura reactions and by the synthesis of a 3‐deaza‐derivative of the natural product canthin‐6‐one.

Abstract:This paper explores an original synthesis of novel heterocyclic compounds derived from photoinduced intramolecular coupling reactions, involving the formation of C‐N and C‐C bonds. Specifically, two classes of compounds were targeted: 4H‐benzo[e]pyrazolo[1,5‐b][1,2,4]thiadiazines and 5‐amino‐6‐thia‐4,5a‐diazaacephenanthrylenes. The synthetic strategy involved nucleophilic substitution reactions to prepare key sulfonamide starting substrates, followed by photoinduced cyclization (via the Substitution Radical Nucleophilic Unimolecular, SRN1) under mild conditions using visible light. The scope of the reaction was explored with different substituent groups on the starting materials, which demonstrated moderate to very good product yields, with 13 examples and yields of up to 78%. Moreover, this strategy represents the first synthetic approach to obtain 5‐amino‐6‐thia‐4,5a‐diazaphenanthrylenes.

Abstract:An approach to hydrocarbazoles bearing an all‐carbon quaternary center at C4a position was developed via a Brønsted acid‐initiated Diels‐Alder cycloaddition/retro‐aza‐Michael addition cascade process from azepino[4,5‐b]indoles and commercially available dienophiles. The method provided a range of hydrocarbazoles in 63–99% yields. The practicality of this transformation was demonstrated by a scale‐up experiment and various transformations to several hydrocarbazole derivatives and the tetracyclic indoline scaffold. Moreover, hetero‐Diels‐Alder cycloadditions of azepino[4,5‐b]indoles were explored with 4‐phenyl‐3H‐1,2,4‐triazole‐3,5(4H)‐dione (PTAD) and 82–99% yields were obtained.

Abstract:A tandem Cp*Rh(III)-catalyzed C-H activation/Bronsted acid-catalyzed intramolecular cyclization allows a facile synthesis of carbazoles from readily available indoles. The reaction proceeds under rather mild reaction conditions with the generation of water and N-2 as the only byproducts. Broad substrate scope, excellent functional group tolerance, and high yields were observed. The benzannulation of pyroles for the synthesis of indoles is also feasible using the same protocol.

Abstract:A simple and mild approach to access polycyclic 8H-indolo[3,2,1-de]phenanthridin-8-ones through a visible-light induced free radical cyclization of (2-bromophenyl)(9H-carbazol-9-yl)methanones using benzothiadiazole-containing photocatalyst is reported. This transition metal free method can be utilized for the synthesis of 7H-thieno[2′,3′:4,5]pyrido[3,2,1-jk]carbazol-7-one, 14H-isoquinolino[4,3,2-kl]phenoxazin-14-one, 14H-isoquinolino[4,3,2-kl]phenothiazin-14-one, and 10,11,12,13-tetrahydro-8H-indolo[3,2,1-de]phenanthridin-8-one. Moreover, some products were found to emit strong blue or green fluorescence.
