Abstract:Protonated 1,8-diazabicyclo[5,4,0]undec-7-ene as catalyst for cascade addition/cyclization of 2-alkynylaniline and carbon disulfide has been described. This process provides a convenient route for synthesis of a variety of benzo[d][1,3]thiazine-2(4H)-thiones in high yields with high regio- and stereoselectivity at room temperature without metal.

Abstract:A rhodium-catalyzed cascade reaction of benzoic acids with 1,4,2-dioxazol-5-ones was studied. The carboxyl group enabled a double C-H amidation followed by further intramolecular cyclization to afford 2,5-substituted benzoxazinones, which exhibited aggregation-induced emission (AIE) properties with a promising excited-state intramolecular proton-transfer (ESIPT) phenomenon.

Abstract:We describe here an organocatalytic asymmetric cascade formal [3 + 3] cycloaddition of benzothiazoles with 2-nitroallylic acetates and nitroenynes. This dearomative methodology provided a facile and efficient strategy for the construction of a broad range of valuable benzothiazolopyridines bearing two adjacent stereogenic centers in moderate to good yields with good to excellent stereocontrol.

Abstract:Metal‐free [2 + 2 + 1] three‐component cascade annulations for the one‐pot synthesis of multiply substituted 3‐cyano‐thiophenes from 3‐oxo‐nitriles, enones and elemental sulfur is described. This DBU‐promoted protocol features good functional group tolerance, and provides high value 3‐cyano‐thiophenes in moderate to good yields under simple reaction conditions. This approach also enables late‐stage modifications of structurally complex bioactive molecules and drugs, which exhibits potential applications in medicinal chemistry.

Abstract:We have developed an efficient method for the synthesis of benzimidazoles via cascade reactions of o-haloacetoanilide derivatives with amidine hydrochlorides. The protocol uses 10 mol % CuBr as the catalyst, Cs2CO3 as the base, and DMSO as the solvent, and no ligand is required. The procedure proceeds via the sequential coupling of o-haloacetoanilide derivatives with amidines, hydrolysis of the intermediates (amides), and intramolecular cyclization with the loss of NH3 to give 2-substituted 1H-benzimidazoles.

Abstract:A highly enantioselective cascade carbonylation/annulation of benzyl bromides, CO, and vinyl benzoxazinanones under mild conditions has been established by Pd/chiral Lewis base relay catalysis, providing an efficient method to assemble chiral quinolinones from readily available starting materials in good yields with excellent diastereo- and enantioselectivities. The palladium catalyst plays two roles in this reaction, enabling both the carbonylation process and the generation of the zwitterionic π-allyl palladium intermediate.
