Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:Natural products are a source of many novel compounds with biological activity for the discovery of new pesticides and pharmaceuticals. Quinoxaline is a fused N-heterocycle in many natural products and synthetic compounds, and seven novel quinoxaline derivatives were designed and synthesized via three steps. Pesticidal activities of title quinoxaline derivatives were bioassayed. Most of these compounds had herbicidal, fungicidal, and insecticidal activities. The compounds 2-(6-methoxy-2-oxo-3-phenylquinoxalin-1(2H)-yl)acetonitrile (3f) and 1-allyl-6-methoxy-3-phenylquinoxalin-2(1H)-one (3g) were the most active herbicides and fungicides. Mode-of-action studies indicated that 3f is a protoprophyrinogen oxidase-inhibiting herbicide. Compound 3f also possessed broad-spectrum fungicidal activity against the plant pathogen Colletotrichum species. Some of these compounds also had insecticidal activity. Molecular docking and DFT analysis can potentially be used to design more active compounds.

Abstract:Three quinoline carbonothioate compounds were synthesized using o-fluoroaniline and ethyl 2-methylacetoacetate as starting materials via multi-steps. The structures of three compounds were confirmed using H-1 NMR and HRMS. The structure of O-ethyl S-(8-fluoro-2,3-dimethylquinolin-4-yl) carbonothioate (3a) was further determined through X-ray single-crystal diffraction. Single-crystal X-ray diffraction analysis indicated that compound 3a crystallizes in the monoclinic space group C2/c, with unit cell parameters: a = 14.3284(6) & Aring;, b = 19.2857(8) & Aring;, c = 10.3990(5) & Aring;, beta = 108.1220(10)degrees, V = 2731.0(2) & Aring;(3). In addition, the physicochemical properties of compound 3a were calculated and discussed using molecular electrostatic potential, frontier molecular orbital and Hirshfeld surface analysis. The antifungal activity of the three compounds was evaluated at 50 mu g/mL, with compound 3b exhibiting good activity (75.8%) against R. solani.

Abstract:A series of novel indole[1,2-c]-1,2,4-benzotriazine derivatives were obtained by a modified Sandmeyer reaction in the presence of tert-butylnitrite (t-BuONO). As compared with hymexazol, a commercially available agricultural fungicide, at the concentration of 50 μg/mL, two indole[1,2-c]-1,2,4-benzotriazines, 5h and 5k, exhibited the more promising and pronounced antifungal activities in vitro against five phytopathogenic fungi. It clearly demonstrated that introduction of appropriate substituents on the indolyl ring of indole[1,2-c]-1,2,4-benzotriazine (5a) would lead to the more potent derivatives.

Abstract:Different substituted phenylhydrazone groups were linked to the quinoxaline scaffold to provide 26 compounds (6a-6z). Their structures were confirmed by (1)H and (13)C NMR, MS, elemental analysis, and X-ray single-crystal diffraction. The antifungal activities of these compounds against Rhizoctonia solani were evaluated in vitro. Compound 6p is the most promising one among all the tested compounds with an EC50 of 0.16 μg·mL(-1), more potent than the coassayed positive control fungicide carbendazim (EC50: 1.42 μg·mL(-1)). In addition, these compounds were subjected to antioxidant assay by employing diphenylpicrylhydrazyl (DPPH) and mice microsome lipid peroxidation (LPO) methods. Most of these compounds are potent antioxidants. The strongest compounds are 6e (EC50: 7.60 μg·mL(-1), DPPH) and 6a (EC50: 0.96 μg·mL(-1), LPO), comparative to or more potent than the positive control Trolox [EC50: 5.90 μg·mL(-1) (DPPH) and 18.23 μg·mL(-1) (LPO)]. The structure and activity relationships were also discussed.

Abstract:A series of new quinoline ester derivatives containing diphenyl ether motif were designed and synthesized using 4-chlorophenol, 1‐chloro-2-nitrobenzene, as raw materials via four steps. The structures were confirmed by 1 H NMR and HRMS. The compound 8-(4-chlorophenoxy)-2,3-dimethylquinolin-4-(4‐chloro) benzoate (4j) exhibited moderate fungicidal activity (68.2% inhibition) against the plant pathogens Sclerotinia sclerotiorum at 50 μg/mL.

Abstract:Objective Based on the structure-activity relationships on the reported antifungal agents bearing quinoline or thiophene moieties ,novel compounds bearing both quinoline and thiophene were designed ,synthesized ,and evaluated for in vitro antifungal activity against Candida albicans .Methods With 5-cyanothiophene-2-carbaldehyde or 5-cyanothiophene-3-car-baldehyde as starting materials ,13 compounds were synthesized via reductive amination ,reduction of cyano group and amida-tion of quinoline-or isoquinoline-carboxylic acid .Their chemical structures were characterized by 1 H NMR and MS . In vitro antifungal screening against Candida albicans SC5314 was performed with the microbroth dilution method .Results All the compounds exhibited potent antifungal activities against Candida albicans .Among them ,compound 6k showed the highest an-tifungal activity with MIC80 value of 0 .5 μg/ml ,which is same potent as fluconazole .Conclusion The designed compounds bearing both quinoline and thiophene exhibited potent antifungal activities ,and deserve further research .

Abstract:A series of novel 1,3-thiazoline derivatives containing a 5- or 6-position halogenated indole moiety were synthesized via a one-pot and three-step reaction. Their structures were confirmed by spectroscopic methods. The fungicidal activities of all the title compounds were evaluated. Most of them exhibited moderate to high activity against eight plant pathogens at the concentration of 50 ug/mL. Generally the fungicidal activity of fluorinated indolyl thiazolines was higher than that of chlorinated indolyl thiazoline derivatives.Supplemental materials are available for this article. Abstract:A series of new fluorinated quinoline analogs were synthesized using Tebufloquin as the lead compound, 2-fluoroaniline, ethyl 2-methylacetoacetate, and substituted benzoic acid as raw materials. Their structures were confirmed by 1H NMR, 13C NMR, and HRMS. The compound 8-fluoro-2,3-dimethylquinolin-4-yl 4-(tert-butyl)benzoate (2b) was further determined by X-ray single-crystal diffraction. The antifungal activity was tested at 50 μg/mL, and the bioassay results showed that these quinoline derivatives had good antifungal activity. Among them, compounds 2b, 2e, 2f, 2k, and 2n exhibited good activity (>80%) against S. sclerotiorum, and compound 2g displayed good activity (80.8%) against R. solani.

Abstract:A series of novel quinazolin-4(3H)-one derivatives 3a similar to 3e and 4a possessing 3,4-benzo[c] coumarin and amino side chain were designed and synthesized by the condensation reaction of 2-aminobenzamide and 3,4-benzo[ c] coumarin aldehyde prepared by Heck coupling reaction, followed by S(N)2 substitution reaction with haloalkane. The compounds were evaluated for their anti proliferation activities against four tumor cells and antimicrobial activities. The results showed that 3e showed moderate anti Hela activity with the IC50 value of 22.63 and 23.35 mu mol/L for 4a against MCF-7. Most tested compounds exhibited significant anti Escherichia coli activities, and the inhibition rate was above 89% at the concentration of 50 mu g/mL. The inhibition rates of 2-phenyl-4-(2-(piperidin-1-yl) ethoxy) quinazoline (1b) against Fusarium oxysporum and Rhizoctonia solani and 3d against Verticillium dahliae are 100%.

Abstract:A series of novel indole Schiff base derivatives (2a–2t) containing a 1,3,4-thiadiazole scaffold modified with a thioether group were synthesized, and their structures were confirmed using FT-IR, 1H NMR, 13C NMR, and HR-MS. In addition, the antifungal activity of synthesized indole derivatives was investigated against Fusarium graminearum (F. graminearum), Fusarium oxysporum (F. oxysporum), Fusariummoniliforme (F.moniliforme), Curvularia lunata (C. lunata), and Phytophthora parasitica var. nicotiana (P. p. var. nicotianae) using the mycelium growth rate method. Among the synthesized indole derivatives, compound 2j showed the highest inhibition rates of 100%, 95.7%, 89%, and 76.5% at a concentration of 500 μg/mL against F. graminearum, F. oxysporum, F.moniliforme, and P. p. var. nicotianae, respectively. Similarly, compounds 2j and 2q exhibited higher inhibition rates of 81.9% and 83.7% at a concentration of 500 μg/mL against C. lunata. In addition, compound 2j has been recognized as a potential compound for further investigation in the field of fungicides.
