Abstract:A simple and efficient methodology for the arylation of benzoxazoles with aldehydes using iodine as the mediator has been developed. The reaction proceeded smoothly with a range of substrates to give the corresponding arylated products in moderate to good yields.

Abstract:The first aryl iodide catalyzed intramolecular C-H amination of phenylurea has been disclosed for high-efficiency synthesis of benzimidazolone derivatives in excellent yields (up to 97%) by an operationally simple one-step organocatalytic oxidative process. Fluorinated protic alcohols can efficiently accelerate the conversion of this transformation. The straightforward method has good functional group tolerance and can be performed with an inexpensive and readily accessible catalyst with high proficiency.

Abstract:An iodine-promoted, metal-, base-, and solvent-free cross-coupling reaction was developed for the synthesis of various useful secondary amides via an aryl N-addition reaction of aryl groups to cyano groups. This aryl transfer reaction proceeds with arylhydrazine hydrochlorides serving as the aryl donors. A labelling experiment shows that the N atom in the product comes from the cyano group of the nitriles, which are low in cost. A plausible radical-driven mechanism is also proposed.

Abstract:Iodine-mediated intermolecular C-N and C-I bond formation of alkenes is described. A series of alkenes could be selectively converted into the corresponding aminoiodination products, which are versatile building blocks in organic synthesis and medicinal chemistry. Wide substrate scope, broad functional group tolerance and easy scale-up operation make this protocol very practical and attractive in synthetic chemistry.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:The direct arylation of aryl iodides with 2-phenylpyridines and related substrates was carried out smoothly in the presence of 5 mol% RuCl3 using benzoyl peroxide as a promoter to generate biarylated products in high yields. The method is simple, efficient, and regioselective, and employs only commercially available reagents.

Abstract:A novel zinc-catalyzed hypervalent iodine mediated cascade transformation of anilines and aldehydes to benzimidazoles was developed. A series of 1,2-disubstituted benzimidazoles were achieved with aromatic, heteroaromatic, and aliphatic aldehydes. In this procedure, initial condensation of anilines with aldehydes afforded N-arylimine intermediates which underwent C-H/C-H amination/annulation to generate the desired products in moderate to excellent yields. The reaction features an extremely simple reaction system, and good functional group toleration with water as the only byproduct.

Abstract:An I(III)-catalyzed oxidative cyclization reaction using Selectfluor as the oxidant was developed that converts ortho-substituted anilines to benzimidazoles is reported. The mild reaction requires as little as 0.5 mol % of iodobenzene tolerating electron-poor groups on the aniline portion. The size of the ring can be varied to access the pyrrole, azepine, and azocines scaffolds. Preliminary mechanistic investigations suggest that benzimidazole formation occurs via cationic reactive intermediates, and an intramolecular kinetic isotope effect of 1.98±0.01 was measured.

Abstract:We report the preparation of 1-(1-alkenyl)benzotriazoles from alkenyl (phenyl)iodonium salts and benzotriazole. The reaction offers a simple and convenient route for the stereoselective synthesis of 1-(1-alkenyl)benzotriazoles. In addition, present method is particularly efficient for preparing 1-(1-alkenyl)benzotriazoles with amino-, acyl- and alkoxycarbonyl substituents, which were difficult to prepare by conventional methods.

Abstract:An iodine-catalyzed aerobic-oxidative C-H functionalization of o-alkylazoarenes to afford 2H-indazoles has been developed. CuI was found to be an effective additive to accelerate the regeneration of iodine in the catalytic cycle. This catalytic system is suitable for both electron-rich and electron-deficient azoarenes and tolerates a variety of functional groups with high yields. A gram-scale reaction was successfully conducted, proving the scalability of this reaction.

The amino- and alkoxy-BX derivatives can be readily synthesized by reacting the respective amines or alcohols with chlorobenziodoxole in the presence of a suitable base. Unlike previously known nitrogen- and oxygen-bound iodane compounds, which have primarily been employed as electrophilic group transfer agents or oxidants, the present amino- and alkoxy-BX reagents manifest themselves as nucleophilic amino and alkoxy transfer agents toward arynes. This reactivity leads to the aryne insertion into the N–I(III) or O–I(III) bond to afford ortho-amino- and ortho-alkoxy-arylbenziodoxoles, iodane compounds nontrivial to procure by existing methods. The BX group in these insertion products exhibits excellent leaving group ability, enabling diverse downstream transformations.

Abstract:Cu- or Fe-based catalyst systems have been reported to selectively catalyze the N,N-diarylation or N-monoarylation of benzoxazoles ring-opening with aryl iodides in the absence of additional added ligand in polyethylene glycol under an inert atmosphere. Two types of coupling products (triphenylamines and diphenylamines) have been examined and the reaction routes can be simply controlled by changing the metal salts (Cu or Fe) as catalyst. A range of substrates have been investigated for the diverse reactions, and the corresponding arylation products were achieved in good to high yields. This selective, low-cost, and environmentally friendly protocol displays great potential for replacing existing methodologies as well as extending the synthetic applications of benzoxazoles.

Abstract:We report an efficient palladium-catalyzed approach to the synthesis of benzoxazole derivatives via sequential heteroarylation/acylation reaction of iodobenzenes. Three readily available starting materials, iodobenzenes, anhydrides, and benzoxazoles, were smoothly coupled to form new C-C bonds at the ortho and ipso positions of the iodobenzenes to afford 2-heteroaryl-3-acylbenzene derivatives in good yields.

Abstract:Abstract Oxazoles and aryl sulfides are chemical entities that are found in many natural products and biologically and pharmaceutically active molecules. It is therefore highly desirable to develop an efficient and practical approach to the synthesis of arylsulfanyl-substituted oxazolines. We developed a simple and efficient method for boron-catalyzed sequential arylsulfanylation and oxygenation of N-allylamides. The protocol uses readily available 1-(arylsulfanyl)pyrrolidine-2,5-diones as the arylsulfanylation reagents and inexpensive boron trifluoride etherate as the catalyst; no ligands or additives are required, and it is not necessary to purge the reaction vessel of air. The method therefore provides an efficient and practical strategy for the synthesis of arylsulfanyl-substituted heterocycles.
