Abstract:Aldehydes and indoles generally undergo nucleophilic addition reactions to generate alcohols rather than ketones. Here, we report a method for synthesis of 3-acyl indoles from aldehydes by a combination of photocatalysis and palladium catalysis. This mild, robust method is compatible with a wide array of functional groups and has a broad substrate scope. Our findings may inspire the development of new dehydrogenative cross-coupling reactions.

Abstract:New functionalizations of indoles via palladium-catalyzed reaction of indoles and 2-acetoxymethyl-substituted electron-deficient alkenes are reported. It was found that for N-protected indoles the reaction proceeded smoothly in the presence of 5 mol % of Pd(acac)2 and 10 mol % of PPh3 at 80 degrees C in HOAc, while for N-unprotected indoles, the reaction was carried out by using 5 mol % of Pd(dba)2 or 2.5 mol % of Pd2(dba)3.CHCl3 with 10 mol % of 2,2'-bipyridine as the catalyst in toluene. This strategy allows the selective installation of electron-deficient olefin functionality at the 3-position of indoles, which might be difficult to obtain by other methods and can be further elaborated.

Abstract:The chosen one! A novel Pd-catalyzed direct and selective C3-acetoxylation of indole derivatives has been accomplished (see scheme). This selective CH activation reaction was implemented without the assistance of directing groups and took place under mild conditions. The kinetic study revealed that the reaction was zero-order with respect to the oxidant and first-order with respect to the indole derivatives.

Abstract:An intermolecular annulation reaction of 2‐aryl‐1,3‐indandions with alkynes was reported using Pd(OAc)2 to access spirobi[indene]‐1,3‐diones. Under palladium catalysis, the substrates form a homocoupling dimer intermediate through a catalytic dehydrogenative cross‐coupling process. The palladium(II) species could come from dimer or 2‐aryl‐1,3‐indandion. Notably, this pathway is not typically observed in enol‐directed formal sp3 C‐H functionalization/oxidative annulation palladium chemistry. This transformation provides a route to access a class of functionalized spiro carbocyclic indenes.

Abstract:A novel approach for the synthesis of the C1 and C3-dialkoxy indoles has been developed for using 2-(phenylethynyl)aniline with formaldehyde and alcohols under the catalysis of acid and palladium acetate. This multicomponent reaction that provides a straightforward alkoxymethylation process to construct substituted indoles is expected to be an important method for the synthesis of indole derivatives.
