Abstract:One new 1,10-secoeudesmanolide (1), two eudesmanolides (2 and 3), together with nine known compounds (4–12) were isolated from the aerial parts of Inula britannica. The structures of the new compounds were elucidated by detailed spectroscopic analysis, including HRESIMS and 2D-NMR spectroscopic method. In addition, compounds 1–4 were tested for their inhibitory effects against LPS-induced NO production in RAW264.7 macrophages.

Abstract:Phytochemical investigation of the flowers of Inula britannica led to the isolation of nineteen sesquiterpenoids (1-19), including britanlins E-J (2, 3, 5, 8, 13, and 14, respectively) and known sesquiterpenoids (1, 4, 6, 7, 9-12, and 15-19). The structures of these isolates were elucidated by detailed spectroscopic analyses and comparison to the previously reported spectroscopic data. The absolute configurations of compounds 12, 15, 17, and 19 were determined by X-ray single crystal diffraction analyses and chemical transformations.

Abstract:Five new sesquiterpenoids, (4 S , 5 S , 6 S , 7 S , 8 R )-5,6-dihydroxy-1-acetoxy-10(14)-en-britannilactone ( 1 ), (4 S , 5 R , 6 S , 7 S , 8 R )-5,6-dihydroxy-1-acetoxy-10(14)-en-britannilactone ( 2 ), 6-O-propionylbritannilactone ( 3 ), 1 β -hydroxy-3 α -acetoxyeudesma-11(13)-en-12,8 β -olide ( 4 ) and 1 β ,5 β -dihydroxyeudesma-11(13)-en-12,8 β -olide ( 5 ), along with twelve known ones were isolated from the flowers of Pentanema britannicum (L.) D.Gut.Larr. Among them, compounds 1 and 2 were stereoisomers which belong to 1,10-seco-eudesmane sesquiterpenoid with rare double bond between C-10 and C-14. The structures of the isolated compounds were elucidated by various spectroscopic methods, including 1D and 2D NMR experiments.

Abstract:Sesquiterpene dimers are mainly found in the Asteraceae family. However, conflicting reports on the structures of these compounds can be found in the literature. Herein, we describe ten sesquiterpene dimers isolated from the flowers of Inula japonica, including configurational revisions of japonicone H (1-1), japonicone D (2-1), inulanolide A (4-1), japonicone X (5-1), and inulanolide F (5-2) to compounds 1, 2, 4, and 5, respectively. Five new related metabolites (3 and 6-9) are also described. Application of GIAO NMR/DP4+ analyses and ECD/OR calculations enabled us to revise the absolute configurations of an additional 13 sesquiterpene dimers isolated from plants of the genus Inula. Compounds 1, 2, 4, and 6 exhibited inhibition of nitric oxide production in lipopolysaccharide activated RAW264.7 macrophages with IC50 values of 4.07-10.00 μM.

Abstract:Eight new (1-8) and two known (9 and 10) sesquiterpenic acids featuring α-methylene-γ-carboxyl units were isolated from the whole plants of Inula wissmanniana, along with two new germacranolides (11 and 12). Their structures were elucidated based on detailed spectroscopic analysis, including HRESIMS, 1D and 2D NMR, and X-ray crystallography. Notably, the skeleton of 1 was firstly discovered from nature, while that of 2 was discovered for the second time. All the compounds were evaluated for their inhibition against LPS-induced nitric oxide (NO) production in RAW264.7 macrophages. Compound 11 exhibited the strongest activity with the IC50 value of 1.04 μM.

Abstract:Fourteen sesquiterpenoids with an eudesmane C-atom skeleton, including four new ones, (1β,5α,7β,8β,11β)-5-hydroperoxy-1-hydroxyeudesm-4(15)-eno-12,8-lactone (1), (1β,5α,7β,8β)-8-(acetyloxy)-5-hydroperoxy-1-hydroxycostic acid methyl ester (12), and a mixture of (1β,3β,4β,7β,8β)-1,3-dihydroxyeudesma-5,11(13)-dieno-12,8-lactone (7) and (1β,3β,4β,7β,8β,11β)-1,3-dihydroxyeudesm-5-eno-12,8-lactone (8), were isolated from the aerial parts of Inula japonica (Asteraceae). Their structures were determined by extensive spectroscopic methods, and those of 7 and 12 confirmed by means of single-crystal X-ray diffraction analysis.

B. Clarke. Their structures were established based on spectroscopic methods including HRESIMS, 1D and 2D NMR, and CD techniques. All compounds were evaluated for their cytotoxic activities against HepG2, HeLa, PC-3, and MGC-803 cell lines by CCK-8 assay. Some of the isolates, especiallly pseudoguaianolides and guaianolides, exhibited significant cytotoxicities against these four examined cell lines.

Abstract:OBJECTIVE:To investigate the sesquiterpene lactones of the aerial parts of Inula helianthus-aquatica.METHOD:Compounds were isolated and purified by silica gel, Sephadex LH-20 and preparative HPLC. On the basis of physicochemical properties and spectroscopic data, their structures were identified.RESULT:Seven sesquiterpene lactones and four other compounds were obtained and identified as 2-desoxy-4-epi-pulchellin (1), 6-acetoxy-4-hydroxy-1, 10H-pseudoguaia-11 (13)-en-12,8-olide (2), 4-acetoxy-6-hydroxy-1, 10H-pseudoguaia-11(13)-en-12,8-olide (3), 8-epi-inuviscolide (4), 2,3,11,13-tetrahydroaromaticin (5), 11,13-dihydro-ergolide (6), 4-epipulchellin-2-O-acetate (7), 7-epiloliolide (8), loliolide (9), beta-sitosterol (10) and daucosterol (11).CONCLUSION:All the compounds were isolated from this plant for the first time.

Abstract:A structurally novel 8,9-seco-eudesma-9,4:8,12-dilactone sesquiterpenoid, chlorajapodiolide (1), a new eudesmane sesquiterpenoid, chlorajapotriol (2), a new lindenane-type sesquiterpenoid dimer, chlorajaponilide I (3), and four known lindenane-type sesquiterpenoid dimers were isolated from the whole plant of Chloranthus japonicus. Their structures were established through extensive analysis of NMR spectroscopic data in combination with mass spectrometry. All compounds were evaluated for cytotoxicities against four human cancer cell lines A549, HCT116, MGC803, and HepG2.

Abstract:Three new sesquiterpenes (1-3), together with four known sesquiterpene lactones, were isolated from the flowers of Inula britannica var. chinensis. Structures were established on the basis of high-field 1D and 2D NMR methods supported by HRMS. All sesquiterpene lactones were tested for cytotoxicity as well as apoptotic ratio in human COLO 205, HT 29, HL-60, and AGS cancer cells. Compounds 3 and 4, two alpha-methylene gamma-lactone-bearing sesquiterpenes, were modestly active in these assays.

The first and third compound showed cytotoxic activity against KB and P-388 cell lines.

Abstract:Four new pseudoguaianolides ( 1 – 4 ), one new guaianolide ( 5 ), together with ten known compounds ( 6 – 15 ) were isolated from the aerial parts of Inula hupehensis . Their structures were elucidated mainly on the basis of 1D and 2D spectroscopic methods and circular dichroism analysis. In addition, compounds 1 – 10 and 13 were tested for their inhibitory effects against LPS-induced NO production in RAW264.7 macrophages. Compounds 2 , 6 , 8 and 9 exhibited significant inhibitory activities with IC 50 values in the range of 0.6–6.6 μM.

Expert Opinion on Investigational Drugs

Abstract:INTRODUCTION:The plants of the genus Inula (Asteraceae) are widely distributed throughout Europe, Africa and Asia, and many of these plants have long been used in folk medicine. This genus is a rich source of sesquiterpenoids, which exhibit a wide range of biological activities. Recently, a series of bioactive sesquiterpenoid dimers, with unusual carbon skeletons, have been reported and these have gathered considerable interest.AREAS COVERED:This article systematically reviews sesquiterpenoids isolated from the genus Inula that have appeared in literature up to August 2013, critically highlighting their anti-tumoral activities and relevant mechanistic insights. The authors also discuss the initial structure-activity relationships for the cytotoxic and anti-tumoral activities of the Inula sesquiterpenoids. Finally, the authors discuss the challenges and potential applications of these sesquiterpenoids in the future.EXPERT OPINION:Cytotoxic and anti-tumor activities of Inula sesquiterpenoids have been extensively studied since the 1970s. One promising compound, Japonicone A, a dimeric sesquiterpene lactone from traditional herb Inula japonica, has displayed potent in vitro and in vivo anti-tumor activity against Burkitt's lymphoma. Additionally, acetylbritannilactone is thought to be capable of suppressing the abnormal vascular smooth muscle cell proliferation, with the induction of apoptosis in vivo and in vitro. In this regard, it may be worthwhile further investigating acetylbritannilactone in patients with vascular restenosis. Furthermore, given the anti-inflammatory property of britanin, clinical studies on chronic bronchitis and asthma, using the ethanol extract of I. japonica, are currently underway in South Korea. However, despite demonstrating good therapeutic effects, additional pharmacological and toxicological studies are still needed.
