Abstract:Two new sesquiterpene lactones, inulasalsolin and inulasalsolide, and the known compounds eupatolide and budlein B, were isolated from Inula salsoloides and their structures assigned unambiguously by COSY, ROESY, HMQC and selective INEPT experiments. The first and third compound showed cytotoxic activity against KB and P-388 cell lines.

Abstract:A novel trinorsesquiterpene (1), three new (2-4), and 10 known sesquiterpenes were isolated from the roots of Inula racemosa Hook. f. The structures and absolute configurations of the new sesquiterpenes were elucidated by extensive spectroscopic and computational methods. All compounds were evaluated for their inhibition of LPS-induced nitric oxide production in RAW264.7 macrophages, and the results indicated that compounds 9, 12, and 13 moderately inhibited nitric oxide production with IC50 values of 7.39 +/- 0.36, 6.35 +/- 0.26, and 5.39 +/- 0.18 mu M, respectively.

Abstract:Twenty-two new sesquiterpenes were isolated from the aerial parts of Inula japonica Thunb., together with fifteen known ones. Their structures were determined by detailed spectroscopic analysis, X-ray diffraction studies, and modified Mosher method. All 37 compounds were evaluated for the inhibition of LPS-induced nitric oxide (NO) production in RAW264.7 macrophages, and most of isolates significantly inhibited the NO production with IC50 values in the range of 3.5–20μM. Besides, results obtained in our studies provided a structure–activity relationship that would be used to design anti-inflammatory agents in the future.

Abstract:A phytochemical investigation to obtain new NO inhibitors resulted in the identification of six new (1-6) and four known (7-10) terpenoids from Salvia plebeia. Compounds 1 and 2 are new diterpenoids, 3-5 are new meroditerpenoids, 6-9 are sesquiterpenoids, and 10 is a known meroditerpenoid. The structures of these isolates were determined by routine NMR experiments and X-ray diffraction, as well as the electronic circular dichroism spectra. Compounds 1-4 are diterpenoids carrying an oxygen bridge, and 6 is a rare copane-type sesquiterpenoid with a bridged tricyclic framework. The isolates inhibited NO generation induced by lipopolysaccharide in BV-2 cells. The possible mechanism of NO inhibition of some bioactive compounds was also investigated using molecular docking, which revealed interactions of bioactive compounds with the iNOS protein.

Abstract:Fourteen sesquiterpenoids including six new analogues (1-6) were isolated from the aerial pails of Inula lineariifolia. The structures of the new sesquiterpenoids were determined by spectroscopic data analysis, and the structures of 1 and 6 were confirmed by single-crystal X-ray diffraction data and the modified Mosher method. All 14 compounds were examined for the inhibition of LPS-induced nitric oxide production in RAW264.7 macrophages. Compounds 6, 13, and 14 were found to inhibit nitric oxide production potently, with IC50 values of 0.11, 0.25, and 0.10 mu M, respectively.

Abstract:A phytochemical study performed on Inula japonica led to isolation of a new 1,10-so-sesquiterpene dimer Neolinulicin A (1) and 1,10-seco-sesquiterpene Neolinulicin B (2), together with nine known sesquiterpenes (3−11). Among them, Neolinulicin A (1), which has a new carbon skeleton, was a Diels-Alder [4 + 2] adduct of two sesquiterpene moieties. Their structures were established by extensive spectroscopic analysis. All of the isolated compounds showed inhibition of NO production in RAW 264.7 macrophages. The findings might supply information for the future design of anti-inflammatory agents from I. japonica.

Abstract:A new sesquiterpene lactone dimer (1) and a new eremophilane sesquiterpene (4) together with nine known sesquiterpenes have been characterized from Inula macrophylla. The structure of macrophyllidimer D (1) and macrophallic acid F (4) were established by comprehensive analysis of spectroscopic data. The absolute configurations were determined by single-crystal X-ray diffraction analysis and calculated electronic circular dichroism (ECD) spectra. In vitro assays results indicated that compounds 5, 7, 9 and 11 exhibited beneficial inhibitory effects on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated BV2 cells.

Abstract:As a part of our ongoing search for plant-derived compounds that inhibit nitric oxide production, the methanol extract of the roots of Vladimiria souliei was found to show significant inhibitory effects on INF-γ-induced nitric oxide production in murine macrophage RAW264.7 cells. Bioactivity-guided isolation of the extract yielded two most active sesquiterpenes, including a new compound, named souliene A (1) and alismol (2). Their structures were elucidated by spectroscopic methods (IR, ESIMS, HRESIMS, 1D and 2D NMR). Two isolates showed promising inhibitory effects on INF-γ-induced nitric oxide production in murine macrophage RAW264.7 cells.

Abstract:Eight new (1-8) and two known (9 and 10) sesquiterpenic acids featuring α-methylene-γ-carboxyl units were isolated from the whole plants of Inula wissmanniana, along with two new germacranolides (11 and 12). Their structures were elucidated based on detailed spectroscopic analysis, including HRESIMS, 1D and 2D NMR, and X-ray crystallography. Notably, the skeleton of 1 was firstly discovered from nature, while that of 2 was discovered for the second time. All the compounds were evaluated for their inhibition against LPS-induced nitric oxide (NO) production in RAW264.7 macrophages. Compound 11 exhibited the strongest activity with the IC50 value of 1.04 μM.

Abstract:Two new inositol derivatives, 4-acetate-1,2,3,5-tetrakis(2-methyl-2-butenoate) inositol ( 1 ) and 3-acetate-1,2,4,5-tetrakis(2-methyl-2-butenoate) inositol ( 2 ), one new flavonolignan 23- o -acetylsilychristin A ( 3 ), and one new thymol derivative 3-hydroxy-4-(2-hydroxy-1-methylethyl)benzoic acid ( 4 ), along with 13 known compounds, including ten flavonolignans ( 5 – 14 ) and three thymol derivatives ( 15 – 17 ) were isolated from the aerial part of Inula wissmanniana . The structures of new compounds were elucidated by detailed spectroscopic analysis, including HRESIMS and 2D-NMR spectroscopic data. All the compounds were isolated from this plant for the first time. The anti-inflammatory activities of the compounds were tested against LPS-induced NO production in RAW 264.7 macrophages.

Abstract:Eight new pseudoguaianolides (1-8), two new guaianolides (9 and 10), and 14 known sesquiterpenes were isolated from the aerial parts of Inula hupehensis. The structures were elucidated using spectroscopic methods and circular dichroism analysis. All compounds were tested for inhibitory activities against LPS-induced nitric oxide production in RAW264.7 macrophages. Compounds 13 and 22 were found to inhibit nitric oxide production potently, with IC50 values of 0.9 and 0.6 mu M, respectively. Preliminary structure activity relationships for these compounds are proposed.

Abstract:Five new and seven known mono‐sesquiterpenoids (1–5 and 6–12, resp.) together with five known lindenane‐type disesquiterpenoids, 13–17, were isolated from the whole plant of Chloranthus henryi. Based on spectroscopic methods, the new structures were established to be (5S,6R,8S,10R)‐6‐hydroxyeudesma‐4(15),7(11)‐diene‐12,8‐olide (1), 6α‐hydroxyeudesma‐4(15),7(11),8(9)‐triene‐12,8‐olide (2), 8,12‐epoxy‐1β‐hydroxyeudesma‐4(15),7,11‐trien‐6‐one (3), 12‐oxochloraniolide A (4), and (4α)‐8‐hydroxy‐12‐norcardina‐6,8,10‐trien‐11‐one (5), respectively. Among the isolates, compound 2, zederone epoxide (8), spicachlorantin G (13), chloramultilide A (14), shizukaol B (15), and spicachlorantin B (17) showed significant anti‐neuroinflammatory effects by inhibiting nitric‐oxide (NO) production in lipopolysaccharide (LPS)‐stimulated murine BV‐2 microglial cells with relatively low cytotoxicity.

Abstract:Ten new ent-kaurane diterpenoids, including two pairs of epimers 1/2 and 4/5 and a 6,7-seco-ent-kauranoid 10, were obtained from the aerial parts of Isodon serra. The structures of the new compounds were confirmed by extensive spectroscopic methods and electronic circular dichroism (ECD) data analysis. An anti-inflammatory assay was applied to evaluate their nitric oxide (NO) inhibitory activities by using LPS-stimulated BV-2 cells. Compounds 1 and 9 exhibited notable NO production inhibition with IC50 values of 15.6 and 7.3 μM, respectively. Moreover, the interactions of some bioactive diterpenoids with inducible nitric oxide synthase (iNOS) were explored by employing molecular docking studies.
