Abstract:Two new asymmetric eremophilane-type sesquiterpene dimers, ligulamulienin A (1) and B (2), were isolated from the rhizomes of Ligularia muliensis. Their structures were determined based on their spectroscopic data, including IR, EI-MS, HR-electrospray ionization (ESI)-MS, 1D and 2D-NMR spectroscopy. The cytotoxicity of compounds 1 and 2 was measured in in vitro human carcinoma (MGC-803), human hepatoma (HEP-G2), and murine sarcoma (S-180) cell lines.

Abstract:Virgaurols A-D (1-4), four novel asymmetric eremophilane sesquiterpene dimers were isolated from the roots of Ligularia virgaurea. Especially, virgaurols A (1) and B (2) possessed novel carbon skeleton in which the two sesquiterpene units are uniquely connected by a single C-C bond directly. The structures of these dimers were determined on the basis of comprehensive spectral analysis. Compounds 1 and 3 were found to exhibit weak cytotoxicity against human leukemia (HL-60), human hepatoma (SMMC-7721), and human cervical carcinoma (HeLa) cells.

Abstract:Two new compounds, a stigmasterol (1) and an eremophilenolide (2), were isolated from Ligularia dolichobotrys (Diels) together with ten known sesquiterpenoids, two known triterpenes and five known sterols. Their structures were elucidated by spectroscopic methods (IR, MS, 1H, 13C and 2D NMR). In addition, bakkenolide A (3) exhibited effective antitumor activity to human leukemia cells (HL-60), human hepatoma cells (Bel-7402) and human ovarian neoplasm cells (HO-8910).

Abstract:From the whole plants of Ligularia duciformis, four new sesquiterpenoids, 3 beta-acetoxy-6 beta-methoxyeremophila-7(11),9(10)-dien-12,8 beta-olide (1), 3 beta-acetoxy-8 alpha-hydroxy-6 beta-methoxyeremophila-7(11),9(10)-dien-12,8 beta-olide (2), 3 beta-acetoxy-10 beta-hydroxy-6 beta,8 beta-dimethoxyeremophil-7(11)-en-12,8 alpha-olide (3), and 3 beta-acetoxy-6 beta,8 beta,10 beta-trihydroxyeremophil-7(11)-en-12,8 alpha-olide (4) were isolated. Their structures were established by high-field NMR techniques ((1)H,(1)H-COSY, (13)C-APT, HMQC, HMBC, and NOESY) and HR-ESI-MS analysis, together with comparison of the spectroscopic data with those of structurally related compounds. In addition, the cytotoxicity of the new compounds against human hepatic cancer cells Bel-7402, human pneumonic cancer cells A-549, and human colonic cancer cells HCT-8 were evaluated, the new compounds showed no cytotoxicity against the three tumor cells (all IC(50) values > 200 mu M).

Abstract:Previous phytochemical study of Ligularia duciformis, a special weed in China, which has long been used traditionally in folk medicine for reducing inflammation and for curing apoplexy 1, dealt with the isolation of coniferyl and sinapyl alcohol derivatives. Further investigation of the chemical constituents of this plant afforded two new si napyl alcohol derivatives named as 4-0-[6,7-dihydro-5,6£-dehydro7(9)-dehydrogeranyl]-sinapyl alcohol 1 and 4-0-[5oxogeranyl]-sinapyl alcohol 2. The isolation and structure elucidation of the compounds are discussed here.

Abstract:A new eremophilane norsesquiterpenoid (1), together with a known eremophilane sesquiterpenoid (2), was isolated from the leaves of Ligularia virgaurea. The structure of 1 was elucidated by a combination of spectroscopic analysis (IR, 1D NMR, 2D NMR, and HR-ESI-MS), and its absolute configuration was determined by a single-crystal X-ray diffraction experiment (with copper radiation). The known compound 2 was identified by comparison of its physical and spectral data with those reported in the literature. Compound 1 was assayed for its cytotoxic activities against human cervical carcinoma cell (HeLa) and human small cell lung cancer cell (NCI-446) lines.

Abstract:One new triterpene and seven new eremophilane-type sesquiterpenes were isolated from the roots of Ligularia sagitta, among them la, an O-acetylated oleanane type triterpene, 2a and 3a, two 11-nor eremophilane-type sesquiterpenes, 4a, 5, and 6, three standard eremophilane-type sesquiterpenes, and 7 and 8, two tri-nor eremophilane-type sesquiterpenes. Their structures were established by extensive spectral analyses (1D, 2D-NMR, IR, and MS). Compound la showed moderate cytotoxicity against two tumor cell lines.

Abstract:Phytochemical investigation of the roots of Ligularia dentata has resulted in the isolation of three new bisabolane sesquiterpenes: 1alpha,3beta,10,11 -tetrahydroxy-2alpha,5alpha,8-triangeloyl-oxybisabola-(7)14-en-4-one, 1alpha,3beta,10-trihydroxy-11 -methoxy-2alpha,5alpha,8-triangeloyloxybisabola-7(14)-en-4-one, and lalpha, 3beta,10-trihydroxy-2alpha,5alpha,8-triangeloyloxybisabola-7(14),11(12)-dien-4-one. Their structures were elucidated with spectro-scopic methods and 2D-NMR techniques as well as chemical transformations. In addition, three known phenolic norsesquiterpenes, liguhodgsonal, ligujapone, and 2-hydroxy-platyphyllide have also been obtained.
