Abstract:We report the first catalytic method for activating the acyl C-O bonds of methyl esters through an oxidative-addition process. The oxidative-addition adducts, formed using nickel catalysis, undergo in situ trapping to provide anilide products. DFT calculations are used to support the proposed reaction mechanism, to understand why decarbonylation does not occur competitively, and to elucidate the beneficial role of the substrate structure and the Al(OtBu)(3) additive on the kinetics and thermodynamics of the reaction.

Abstract:The conjugate additions of oxygen-centered nucleophiles to conjugate acceptors are among the most powerful C–O bond formation reactions. The conjugate addition normally takes place at the β-position carbon to the electron-withdrawing group, resulting in the formation of a stabilized carbanion intermediate that can be quenched by proton or electrophiles to form the β-addition (i.e., hetero-Michael addition) products. On the contrary, the formation of α-hydroxyl or alkoxyl amides through conjugate addition needs an α,β-inverse addition. Nevertheless, a regio-inversed nucleophilic α-addition of oxygen-centered nucleophiles to α,β-unsaturated carbonyl compounds still remains less explored because of the electronic mismatch. In this research, we discovered the first α-specific nucleophilic addition of α,β-unsaturated amides with oxygen and fluoride nucleophiles. This region-inversed nucleophilic addition is enabled by the catalysis of a novel redox-neutral nondonor–acceptor organic photoreductant (CBZ6). As low as 0.5 mol % of visible light photoreductant was employed. The mechanistic insights were also explored. The oxidative potential of the excited state of CBZ6 is obtained in −1.92 V (vs SCE), presenting a stronger reductive potential than representative metal-cored or organic photoredox catalysts. Abstract:Carbon dioxide radical anion (CO2 center dot-) is a highly reactive nucleophilic radical species that has recently emerged in organic chemistry as a strong single electron donor (reductant) and as a reactant for the synthesis of carboxylic acids. In general, CO2 center dot- can be generated by either direct reduction of CO2 or HAT of formate salts. Achievements in reactions involving CO2 center dot- have been witnessed in recent years. This Review summarizes the recent advances in reactions involving CO2 center dot- by highlighting some challenges and identifying potential areas of improvement, which may offer inspiration for future studies.

Abstract:One of the most common reactions of diazo compounds with alkenes is cyclopropanation, which occurs through metal carbene or free carbene intermediates. Alternative functionalization of alkenes with diazo compounds is limited, and a methodology for the addition of the elements of Z-CHR2 (with Z = H or heteroatom, and CHR2 originates from N2═CR2) across a carbon–carbon double bond has not been reported. Here we report a novel reaction of diazo compounds utilizing a radical-mediated addition strategy to achieve difunctionalization of diverse alkenes. Diazo compounds are transformed to carbon radicals with a photocatalyst or an iron catalyst through PCET processes. The carbon radical selectively adds to diverse alkenes, delivering new carbon radical species, and then forms products through hydroalkylation by thiol-assisted hydrogen atom transfer (HAT), or forms azidoalkylation products through an iron catalytic cycle. These two processes are highly complementary, proceed under mild reaction conditions, and show high functional group tolerance. Furthermore, both transformations are successfully performed on a gram-scale, and diverse γ-amino esters, γ-amino alcohols, and complex spirolactams are easily prepared with commercially available reagents. Abstract:The synthetic strategies of oxime derivatives participating in radical-type reactions have been rapidly developed in the last few decades. Among them, the N–O bond cleavage of oxime esters leading to formation of nitrogen-centered radicals triggers adjacent C–C bond cleavage to produce carbon-centered free radicals, which has been virtually used in organic synthesis in recent years. Herein, we summarized the radical reactions involving oxime N–O bond and C–C bond cleavage through this special reaction form, including those from acyl oxime ester derivatives and cyclic ketoxime ester derivatives. These contents were systematically classified according to different reaction types. In this review, the free radical reactions involving acyl oxime esters and cyclic ketoxime esters after 2021 were included, with emphasis on the substrate scope and reaction mechanism.

Abstract:Aliphatic amides are selectively functionalized at the γ- and δ-positions through directed radical 1,5 and 1,6 H-abstractions, respectively. The initially formed γ- or δ-lactams are intercepted by N-iodosuccinimide and trimethylsilyl azide, leading to double and triple C-H functionalizations at the γ-, δ-, and ε-positions. This new reactivity is exploited to convert alkyls into amino alcohols and allylic amines.

Abstract:The straightforward strategy of building a chiral C-O bond directly on a general carbon radical center is challenging and stereocontrol of the reactions of open-chain hydrocarbon radicals remains a largely unsolved problem. Advance in this elementary step will spur the development of asymmetric radical C-O bond construction. Herein, we report a copper-catalyzed regioselective and enantioselective carboesterification of substituted dienes using alkyl diacyl peroxides as the source of both the carbon and oxygen substituents. The participation of external acids in this reaction substantially extends its applicability and leads to structurally diverse allylic ester products. This work represents the advance in the key elementary reaction of intermolecular enantioselective construction of C-O bond on open-chain hydrocarbon radicals and may lead to the discovery of other asymmetric radical reactions.

Abstract:Transition metal-catalyzed direct C-H functionalization has drawn great attention in the past several decades owing to its advantages compared to conventional organic transformations, including higher atom-, step-and cost-economy and the avoidance of tedious prefunctionalization and waste emission. At the current stage, to make the C-H functionalization more applicable, chemists have devoted themselves to expanding the substrate and reaction scope. In the past decade, we exerted ourselves to develop new transformations based on direct C-H functionalization. In this minireview we report on our recent achievements on the addition of C-H bonds to carbonyls and imines. The addition of organometallic reagents, such as Grignard reagents, toward carbon-heteroatom double bonds is one of the most powerful reactions in organic synthesis to produce secondary and tertiary alcohols and amines. This chemistry is broadly used in both laboratory and industry. However, this powerful transformation suffers from some drawbacks: (1) the preparation of initial organohalides from easily available fossil feedstocks is tedious and sluggish; (2) substantial amounts of metal halide salts are emitted as waste; (3) last but not least, the manipulation of organometallic reagents is complicated due to their sensitivity to air and moisture. In contrast, direct insertion of polar double bonds to C-H bonds via transition-metal catalysis is ideal from the viewpoint of atom-, step-and cost-economy and the avoidance of the waste emission, as well as of the complex manipulation of sensitive reagents. Starting from this point, we made a commitment to this project years ago and have made credible achievements in this field. We first carried out Ir-catalyzed addition of pyridinyl C-H bonds to aldehydes promoted by silane, showing an unusual C-3 selectivity. Later on, we developed Rh-catalyzed addition of aryl C-H bonds with aldimines in the absence of any additives with directing strategy with highest atom-and step-economy. The mechanism was investigated in depth by the isolation of key intermediates and systematic thermodynamic and kinetic studies. Such a concept was expanded to the coupling of aryl/alkenyl C-H bonds with aldehydes and imines. Notably, a tandem process of relayed C-H activation/alkyne insertion/ cyclization between benzoates/benzimide and alkynes was developed, indicating the potential of the direct coupling of esters and amides with C-H bonds. Ideally, this strategy opens a new window to approach the ideal reactions to produce amines and alcohols from hydrocarbons.
