Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:We report herein a synthesis of quinoxalin‐2(1 H )‐ones from 3‐hydroxyindolin‐2‐one derivatives, which can be synthesized from commercially available isatins in one or two steps. The reaction proceeded through a CF 3 CO 2 H‐mediated azidation via a carbocation, followed by acid‐induced N 2 extrusion and ring‐expansion. Both CF 3 CO 2 H and the solvent 1,1,1,3,3,3‐hexafluoro‐2‐propanol were found to be crucial for this rearrangement. The axial chirality in 3‐hydroxyindolin‐2‐one skeleton was fully preserved in the products.

Abstract:The present study provided an efficient synthesis pathway to the azepino[5,4,3‐cd]indoles using N‐sulfonyl‐1,2,3‐triazoles and 4‐vinylindoles through a cascade carbene insertion and intramolecular aza‐Michael addition. This method is distinguished by mild reaction conditions, straightforward operational steps, accessible starting materials, and broad substrate compatibility. Furthermore, the versatility of this reaction with various Michael acceptors significantly broadens the scope of potential substrates. The ability to further transform the resulting multifunctional products underscores the promising applications of this approach in indole nitrogen heterocyclic synthesis.

Abstract:Azetidines, being four-membered N-heterocycles, possess significant potential in contemporary medicinal chemistry owing to their favorable pharmacokinetic properties. Regrettably, the incorporation of functionalized azetidines into pharmaceutical lead structures has been impeded by the absence of efficient synthetic methods for their synthesis. In this study, a Rh-catalyzed one-carbon ring expansion of aziridines with vinyl-N-triftosylhydrazones is presented, which facilitates the synthesis of high value-added 2-alkenyl azetidine products. This research represents the first example of ring expansion of aziridines enabled by vinyl carbenes. Additionally, a one-pot two-step protocol, initiated from cinnamaldehyde, was successfully achieved, offering a step-economical and facile approach for the synthesis of these compounds. The pivotal aspect of this successful transformation lies in the in-situ formation of an alkenyl aziridinium ylide intermediate. Experimental investigations, coupled with computational studies, suggest that a diradical pathway is involved in the reaction mechanism.

Abstract:A novel Brønsted-acid-promoted selective C2-N1 ring-expansion reaction of indoles has been developed that provides a rapid and efficient protocol for the preparation of fused quinolines. A variety of corresponding quinolines were obtained in high yields. Controlled experiments revealed that C2-spiroindolenines might be intermediates of this C2-N1 ring-expansion reaction. The notable advantages of this process include excellent yields, good functional group tolerance, and operational simplicity.

Abstract:Gold-catalyzed and NaH-supported intramolecular cyclization of N-propargyl indole derivatives with pyrazole and pyrrole units attached to indole is described. An efficient route to the synthesis of pyrazolodiazepinoindole, pyrazolopyrazinoindole, and pyrrolopyrazinoindole has been established. First, N-propargyl 2-(1H-pyrazol-5-yl)-1H-indole and 2-(1H-pyrrol-2-yl)-1H-indole were synthesized. Introduction of various substituents into the alkyne functionality was accomplished by Sonogashira cross-coupling reaction. Gold-catalyzed cyclization of pyrazoles having a terminal alkyne afforded the 6-exo-dig cyclization product. However, exclusive formation of 7-endo-dig cyclization products was observed with internal alkynes. On the other hand, cyclization with NaH only resulted in the formation of 6-exo-dig cyclization products regardless of the substitution of the alkyne functionality. Allenic intermediates were postulated for this outcome.

Abstract:The chiral pyrrolo[1,2-a]indole represents a privileged stuctural motif in many natual products and pharmaceutical agents. Herein we developed an efficient [3+2] annulation of 2-nitrovinylindoles with azlactones via the cascade Michael addition and intramolecular acylation under the catalysis of bifunctioanl tertiary amine in combination with DABCO, delivering a wide range of pyrrolo[1,2-a]indoles in good yields with high diastereo- and enantioselectivities. Alternatively, 7-nitrovinylindoles are served as 4C-synthons to perform the enantioselective [4+2] annulation under the identical condition, smoothly giving the pyrrolo[3,2,1-ij]quinoline skeleton with good to high stereoselectivities. Furthermore, a novel enantio-enriched indole-based tetracyclic skeleton was facilely derived from the obtained pyrrolo[1,2-a]indoles.
