Abstract:Pyridones/isoquinolones are a kind of important nitrogen-containing heterocyclic compounds, which have good biological activity and unique chemical properties, and play an important role in synthesis chemistry, functional materials, biomedicine and so on. Recently, transition-metal-catalyzed C-H activation has emerged as a more atom- and step-economical strategy for the construction of functionalized pyridones/isoquinolones, and it has attracted the attention of many organic chemists. Herein, the recent advances in transition-metal-catalyzed C-H activation of pyridone/isoquinolone derivatives are summarized according to the types of metal catalysis. An emphasis on the discussion of catalytic system, reaction mechanisms, substrate scopes and synthetic applications is also introduced. Finally, the limitations and development trend of this research field are analyzed and prospected.
Accelerated Metal-Free Hydration of Alkynes Within Milliseconds in Microdroplets

Abstract:Conversion of alkynes into ketones by hydration is one of the most fundamental organic addition reactions. Traditional methods for alkyne hydration suffer from usage of toxic and costly metals, excessive acids, harsh conditions, and long reaction times, bringing about an urgent desire to explore more efficient and green protocols to complete this transformation. A metal-free efficient protocol for the accelerated hydration of alkynes in microdroplets was successfully developed in this work to overcome these drawbacks. The hydration process was finished under room temperature using only 18 mol % sulfuric acid as the catalyst and acetonitrile as the solvent. Complete conversions were feasible on the low millisecond time scale in microdroplets for a wide range of alkynes bearing functional groups, including electron-withdrawing groups and electron-donating groups. This is in sharp contrast to trace amounts of ketones or none in 20 min in bulk phase. Compared to the bulk reaction under the same conditions, the reactions in microdroplets are accelerated by 4 to 5 orders of magnitude as seen by the increase in measured rate constants. Further, the accelerated microdroplet hydration can be scaled up to an acetophenone amount of 57.2 +/- 0.1 mg min(-1) (3.43 +/- 0.01 g h(-1)) by using 2 mol L-1 phenylacetylene sprayed at 300 L min(-1). The microdroplet protocol offers several advantages including ultrahigh acceleration, complete conversion, mild reaction conditions (metal, light, and heat free), and minimal post purification, as well as a wide substrate scope, making it attractive for green and sustainable chemistry.

Abstract:A general and convenient method for the direct transition-metal-free intermolecular chloro- difluoromethylthiolation of a variety of unactivated alkenes and alkynes has been described. This methodology exhibits good functional group tolerance and operationally simple. Mechanistic studies indicated that this is the electrophilic substitution mechanism, and the strong electrophilic difluoromethylthiolating reagent HCF 2 SCl was generated in situ under the standard reaction conditions. The developed protocol fulfills the requirements of sustainable and green chemistry.

Abstract:A general transition-metal-free photoinduced acetalation-pyridylation of alkenes using diethoxyacetic acid and cyanopyridine was developed under mild conditions. By employing 4CzIPN as the photocatalyst and Cs2CO3 as the base, a diverse range of styrene derivatives and cyanopyridines worked well to give the desired products. The versatility of this method is highlighted by its application in the construction of various functional groups and the late-stage modification of drugs. Importantly, some of the synthesized compounds showed good in vitro antitumor activity, indicating that this protocol is of significance and potential for antitumor drug development.

Abstract:A specific N-alkylation of 2-hydroxypyridines is achieved by reacting with organohalides under catalyst- and base-free conditions. The observed HX-facilitated conversion of pyridyl ether intermediates to 2-pyridone products may account for the success and specific N-alkylation of the reaction under the unexpectedly simple conditions. This new reaction may provide a useful alternative for the synthesis of 2-pyridones and analogous structures because of its >99% N-selectivity, relatively broad scopes of both substrates, and no mandatory use of catalysts and bases.

Abstract:A catalyst‐based switchable regioselective C−H activation/annulation of acrylamides with propargyl carbonates has been developed, delivering C5 or C6 alkenyl substituted 2‐pyridones. This robust protocol proceeds with a broad substrate scope and good functional group tolerance under redox‐neutral reaction conditions. More significantly, this reaction is highly effective with previously challenging unsymmetrical alkynes, including unbiased alkyl‐alkyl substituted alkynes, with perfect and switchable regioselectivity. Additionally, mechanistic studies and DFT calculations were performed to shed light on the switchable regioselectivity.
