Abstract:Ring fusion: The Pd(0)-catalyzed reaction of 2-allyl-3-iodo-1-tosyl-1H-indoles and propargylic bromides affords dihydrocycloocta[b]indoles (see scheme; M.S. = molecular sieves, TFP = tris(2-furyl)phosphine, Ts = 4-toluenemethanesulfonyl), and proceeds by carbon-carbon coupling, [1,5]-hydrogen migration, and electrocyclization. Abstract:A Lewis acid Sc(OTf)(3)-catalyzed annulation reaction of vinyl diazoacetates with in situ formed α,β-unsaturated imines for the preparation of indole-fused tricyclic rings has been reported. This strategy involves a sequential addition/rebound addition process of vinyl diazoacetates and an in situ dedinitrogenation. This annulation protocol features low-cost catalysts, mild reaction conditions, and facilely prepared substrates.

Abstract:A strategy for the synthesis of 6,9-dihydropyrido[1,2-a]indoles through a cascade iodocyclization of 4-(3-methyl-1H-indol-1-yl)-1,1-diphenylbut-2-yn-1-ol derivatives is presented. This reaction was conducted under very mild conditions and in a short time. The reactions are metal-free, are environmentally friendly and give up to 94% yield. Moreover, the obtained halides allow functional group diversification by palladium-catalyzed coupling reactions, which could act as potential intermediates for the synthesis of valuable compounds.

Abstract:A novel method for the synthesis of C2-quaternary indole-3-ones via palladium-catalyzed one-pot transformation of 2-alkynyl arylazides is described. The reaction produced in moderate to excellent yields under mild conditions. This novel rearrangement of 1-H-indole-3-sulfonates represents an important contribution to the application of aryl sulfonates.

Abstract:We report a palladium-catalyzed asymmetric [4+4] cycloaddition reaction between 2-alkylidenetrimethylene carbonate and electron-deficient indole-2,3-quinodimethanes (IQDMs). This reaction features exclusive regioselectivity, high yield (up to 98%), excellent enantioselectivity (up to 95% ee), and easy scale-up without any loss of efficiency, making it valuable for the synthesis of indole-fused eight-membered oxa-rings. The Pd-catalyzed asymmetric [4+4] cycloaddition reaction between 2-alkylidenetrimethylene carbonate and electron-deficient indole-2,3-quinodimethanes (IQDMs) results in the formation of indole-fused eight-membered oxa-rings with high enantioselectivity and excellent yield (up to 98% yield, 95% ee). The practicality of the method is evidenced by the successful gram-scale reaction and efficient product transformation. image

Abstract:An efficiently catalytic method toward the synthesis of indolin‐2‐one featuring an allylic derived C3‐quaternary stereocenter via an intramolecular Heck cyclization/Suzuki coupling of N‐substituted‐N‐(2‐bromophenyl)acrylamides and organoboron reagents was successfully developed by using a 1,3‐bis(2,6‐diisopropylphenyl)acenaphthoimidazol‐2‐ylidene (AnIPr)‐ligated oxazoline palladacycle. It enabled a very broad substrate scope tolerating different functional groups, electronic properties and steric bulkiness. Notably, it revealed great potential to build diverse heterocycle‐fused indoline alkaloids via the same intermediate 3‐allyl‐1,3‐dimethylindolin‐2‐one. Abstract:Convenient and regioselective syntheses of 1,2-annulated, and 1,2-, 5,6- and 1,2-, 7,8-bisannulated polycyclic indolizine derivatives have been achieved by one-pot tandem reactions of cyclic N-ylides derived from the corresponding N-substituted pyridinium, quinolinium, and isoquinolinium salts 1–3 with dichloro substituted α,β-unsaturated carbonyl compounds 4–7. The reactions of the N-ylides with 2,3-dichloroindenone 4, 3,4-dichlorocoumarin 5, and 4a,6,7,8a-tetrachloro-1,4-methanonaphthalene-5,8-dione 6 proceed by sequential [3+2] cycloaddition and elimination of hydrogen chlorides from the cycloadducts. On the other hand, reactions of the N-ylides with 2,3-dichloro-1,4-naphthoquinone 7 take place via a novel reaction sequence to give the products 15–17.

Abstract:An interesting sequential three-component copper-catalyzed coupling/propargyl-allenyl isomerization/[4 + 2] cyclization reaction, providing a facile synthesis of highly substituted tetrahydro-1H-isoindolones from conjugated vinylic alkynes, imines, and α,β-unsaturated enoic acid chlorides is reported. The most attractive feature of this transformation is that three stereogenic centers could be generated in one step with high diastereoselectivity.
