Abstract:Four novel nortriterpenoids, 24,25-epoxy-tirucall-7,20(E)-diene-3,23-dione (1), 24,25,26,27-tetranortirucall-1,7-diene-23(21)-lactone (2), 3 alpha-hydroxy-tirucall-7-ene-20-one (3), and 3-oxo-tirucall-7-ene-3,20-dione (4), together with one known compound mombasol (5) were isolated from the stem barks of Aphanamixis grandifolia. Their structures were elucidated on the basis of various spectroscopic analysis including electrospray ionization (ESI)-MS, high resolution (HR)-ESI-MS, IR, 1D- and 2D-NMR techniques (heteronuclear single quantum coherence (HSQC), heteronuclear multiple bond connectivity (HMBC) and rotating frame Overhauser enhancement spectroscopy (ROESY)), and by comparison of their spectral data with those reported. The absolute configuration of compound 1 was determined by circular dichroism (CD) exciton chirality.

Abstract:Phytochemical investigation on the stem bark of Aphanamixis grandifolia afforded five novel tirucallane-type triterpenoids, (13a,14 beta,17a,23Z)-25-methoxy-21,23-epoxylanosta-7,20(22),23-triene-3,21-dione (1), (13a,14 beta,17a,23Z)-21,23-epoxylanosta-7,20(22),23,25-tetraene-3,21-dione (2), (3R,5R, 9R,10R,13S,14S,17S)-17-{(2R,3S,5R)-5-[(2S)-3,3-dimethyloxiran-2-yl]-2,3,4,5-tetrahydro-2,5-dimethoxyfuran-3-yl}-4,4,10,13,14-pentamethyl-2,3,4,5,6,9,10,11,12,13,14,15,16,17-tetradecahydro-1H-cyclopenta[a]phenanthren-3-ol (3), (5R,9R,10R,13S,14S,17S)-17-{(2R,3S,5R)-5-[(2S)-3,3-dimethyloxiran-2-yl]-2,5-dimethoxytetrahydrofuran-3-yl}-1,2,4,5,6,9,10,11,12,13,14,15,16,17-tetradecahydro-4,4,10,13,14-pentamethyl-3H-cyclopenta[a]phenanthren-3-one (4), and (3a,13a,14 beta,17a,20S,23R)-23-ethoxy-3-hydroxy-21,23-epoxylanost-7-en-24-one (5). The 1H- and 13C-NMR spectra of all compounds were fully assigned using a combination of 2D-NMR experiments, including HSQC, HMBC, and ROESY sequences. The structure of 1 with the absolute configuration was determined by ECD calculation. Compounds 3 and 4 showed moderate activities against human MCF-7 and HeLa cancer cells.

Abstract:Aphanamgrandiol A (1), a novel triterpenoid with a bicyclo[3,2,1]octane ring skeleton produced by 2,3-ring opening and 2,6-ring closure, was isolated from the stems of Aphanamixis grandifolia. The structures were established on the basis of extensive spectroscopic methods, including 1D and 2D NMR techniques, and determined unambiguously by X-ray crystal diffraction. Aphanamgrandiol A showed moderate cytotoxicities against MCG-803, SKOV-3, HCT116 and HepG2 cell lines.

Abstract:From the fruits of the tropical tree Aphanamixis grandifolia, five new evodulone-type limonoids, aphanalides I-M (1-5), one new apo-tirucallane-type triterpenoid, polystanin E (6), and three new chain-like diterpenoids, nemoralisins A-C (7-9), along with 12 known compounds were identified. The absolute configurations were determined by a combination of single-crystal X-ray diffraction studies, Mo2(OAc)4-induced electronic circular dichroism (ECD) data, the Mosher ester method, and calculated ECD data. The cytotoxicities of all the isolates and the insecticidal activities of the limonoids were evaluated.

Abstract:Five tirucallane type C(26) triterpenoids, accompanied by two known compounds, 3α-hydroxy-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone and 3-oxo-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone, were isolated from the stem barks of Aphanamixis grandifolia. Their structures were established mainly by means of a combination of 1D and 2D NMR spectroscopic and mass spectrometry techniques as 3α-hydroxyl-21α-methoxy-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone, 3α-hydroxy-21β-methoxy-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone, 3-oxo-21α-methoxy-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone, 3-oxo-21β-methoxy-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone, and 3-oxo-21α-ethoxy-24,25,26,27-tetranortirucall-7-ene-23(21)-lactone. All isolates were in vitro evaluated for their cytotoxic activities against five tumor cell lines (MCF-7, HeLa, HepG2, SGC-7901 and BGC-823).

Abstract:One new sterol, (3 β ,21 α ,22a)-stigmast-5-ene-3,21,22-triol ( 1 ), together with 36 compounds ( 2 – 37 ), were isolated from the stems of Aphanamixis grandifolia . The structure of 1 was elucidated by physical, chemical, and spectroscopic methods, including 1D and 2D NMR and HR-ESI-MS. Besides, 33 compounds ( 2 , 4 , 5 , 7 – 10 , 12 – 30 , and 32 – 37 ) were isolated from the Aphanamixis genus for the first time, and 15 was isolated from the plant for the first time.

Abstract:A total of 14 new diterpene dimers, aphanamenes C-P (1-14), with four known homologous compounds were isolated from the root barks of Aphanamixis grandifolia Bl. The structures of these compounds were elucidated by spectroscopic analyses, and their absolute configurations were determined using the CD exciton chirality method. In addition, all the compounds exhibited significant inhibition of lipopolysaccharide-induced nitric oxide production in RAW264.7 macrophages, with IC50 values between 7.75 and 38.23 mu M.

Abstract:Two novel tirucallane C27-triterpenoid epimers, aphagranins A (1) and B (2), featuring an unprecedented enolized cyclopentenone presented in the side-chain at C-17, were isolated from the stem barks of Aphanamixis grandifolia. Extensive spectroscopic analyses helped the establishment of the structures of the two isolates, whose absolute configurations were determined using density functional theory (DFT) calculations of optical rotation, and electronic circular dichroism (ECD). Remarkable discrepancies in the inhibitory activities against the growth of six lines of human cancer cells (MCF-7, A549, HepG2, Bel-7402, SGC-7901, and BGC-823) were found for the two epimers: with IC50 less than 10μM, aphagranin A exhibited much stronger antiproliferative activity than aphagranin B, showing no such activities with IC50 over 20μM.

Abstract:In order to understand the antitumor active ingredients ofAphanamixis grandifoliaBl., nine compounds were isolated from 95% ethanol extract of its roots. They were identiifed as 7-hydroxycadalene (1), dregeana-1 (2), 4-oxopinoresinol (3), 4-ketopinoresinol (4), 6-deoxyjacareubin (5), schleicheol 1 (6), stigmasterol (7), β-sitosterol (8), and daucosterol (9) on the basis of spectral data. Compounds1–6 were isolated from the genusAphanamixis for the ifrst time, and the13C NMR spectral data of compound1 was reported for the ifrst time. Compounds1 and 5 showed inhibition of the proliferation of K562 cell line, and compounds1,5 and6 possessed inhibitory activity against SGC-7901 cell line.
