Abstract:Aphanamgrandiol A (1), a novel triterpenoid with a bicyclo[3,2,1]octane ring skeleton produced by 2,3-ring opening and 2,6-ring closure, was isolated from the stems of Aphanamixis grandifolia. The structures were established on the basis of extensive spectroscopic methods, including 1D and 2D NMR techniques, and determined unambiguously by X-ray crystal diffraction. Aphanamgrandiol A showed moderate cytotoxicities against MCG-803, SKOV-3, HCT116 and HepG2 cell lines.

Abstract:Aphanapolystachones A–C ( 1 – 3 ), three undescribed sesquiterpene-diterpene heterodimers, were obtained from the fruits of Aphanamixis polystachya . Their structures and absolute configurations were identified by extensive analysis of HR-ESI-MS, NMR, experimental and TD-DFT calculated ECD spectra. The biosynthetic pathway of them was also proposed, which is produced by key intermolecular Diels-Alder [4 + 2]-cycloaddition reaction between a guaiane sesquiterpene and an acyclic diterpene. Compounds 1 – 3 inhibited NO production in LPS activated RAW264.7 cells with the IC 50 values of 1.7 ± 0.2, 3.0 ± 0.3, 5.3 ± 0.3 μM, respectively, lower than that of the positive control L-NMMA (31.5 ± 2.6 μM). In addition, compounds 1 – 3 significantly reduced IL-6 secretion at diluted concentration of 0.4 μM.

Abstract:A new prenylated coumarin (1) and a new anthranilamide derivative (2) were isolated from the aerial parts of Evodia lepta. Compound 2 represents the first anthranilamide derivative from the Evodia genus. Their structures were elucidated by spectroscopic data analysis (MS, UV, IR, and NMR). Compound 1 exhibited certain inhibition on nitric oxide (NO) production induced by lipopolysaccharide (LPS) in RAW264.7 macrophages with an IC50 value of 37.96±1.7 μM.

Abstract:In this article, two undescribed amides (1-2) with an unusual (2-formyl-5-hydroxymethyl)pyrroyl-butylamine moiety were obtained from the Physochlainae Radix. Comprehensive spectroscopic studies, including NMR and HR-ESI-MS, coupling with spectroscopic data comparisons were used to determine structures. Anti-inflammatory assay results showed that new amides possessed significant inhibitory activities of the NO production of LPS-induced RAW 264.7 cells, with IC50 values of 17.52±1.68 μM and 20.37±2.42 μM, respectively.

Abstract:Four new tirucallane-type triterpenoids, polystanins H–K ( 1 – 4 ), were obtained from the stems and leaves of Aphanamixis polystachya . Their structures were elucidated by analysis of the spectroscopic data and comparison with literature data. Compounds 1 and 2 showed week inhibitory effects against NO production in LPS-stimulated RAW264.7 cells. All the isolates were investigated for their antifungal activities against drug-resistant Candida albicans .

Abstract:A new chromone derivative, aspergione A (1), along with seven known metabolites, was isolated from a mangrove endophytic fungus, Aspergillus sp. GXNU-B1, which was collected from mangrove Acanthus ilicifolius L. Their structures and the absolute configuration of 1 were elucidated based on the analysis of HR-ESI-MS, NMR, and ECD calculation. Compounds 1-8 were evaluated for their anti-inflammatory effects on the production of nitricoxide (NO). Compounds 1 and 8 have potent inhibitory effects against NO production in activated macrophages with IC50 values of 38.26 and 44.30 μM, respectively.

Besides, 33 compounds ( 2 , 4 , 5 , 7 – 10 , 12 – 30 , and 32 – 37 ) were isolated from the Aphanamixis genus for the first time, and 15 was isolated from the plant for the first time.

Abstract:A new phytoecdysteroid compound, named Niuxixinsterone D (1), together with two known phytoecdysteroid compounds (2 and 3) were isolated from Achyranthes bidentata Bl.. The structure of the new compound was elucidated by extensive spectral analysis, including HR-ESI-MS, 1D and 2D NMR methods. Compounds 1-3 were tested for their inhibitory effects against LPS-induced NO production in RAW 264.7 macrophages, and compound 1 and 3 exhibited anti-neuroinflammatory activity with inhibited 29.7 and 26.0% NO production.

Abstract:Our ongoing efforts to explore the chemical space associated with marine cyanobacteria from coral reefs of Guam have yielded two new members of the anaenamide family of natural products, anaenamides C (3) and D (4). These compounds were isolated from a novel Hormoscilla sp. (VPG16-58). Our phylogenetic profiling (16S rDNA) of this cyanobacterium indicated that VPG16-58 is taxonomically distinct from the previously reported producer of the anaephenes, VPG16-59 (Hormoscilla sp.), and other previously documented species of the genus Hormoscilla. The planar structures of 3 and 4 were determined via spectroscopic methods, and absolute configurations of the α-hydroxy acids were assigned by enantioselective HPLC analysis. To address the requirement for sufficient material for testing, we first adapted our published linear synthetic approach for 1 and 2 to generate anaenoic acid (7), which served as a point for diversification, providing the primary amides 3 and 4 from synthetic intermediates 5 and 6, respectively. The compounds were then tested for effects on HCT116 colon cancer cell viability and in an ARE-luciferase reporter gene assay for Nrf2 modulation using HEK293 human embryonic kidney cells. Our findings indicate that, in contrast to cytotoxic methyl esters 1 and 2, the primary amides 3 and 4 activate the Nrf2 pathway at noncytotoxic concentrations. Overall, our data suggest that the anaenamide scaffold is tunable to produce differential biological outcomes.

Their chemical structures were elucidated to be 11‐methoxysawaranospiroride C (1), 6α,9S,10,13‐tetrahydroxymegastigmane‐3‐one (2), 11‐hydroxyaphanamixin B (3), (2Z,6E,13E)‐2,6,13‐triene‐11,15‐dihydroxyphytanic acid (4), cinnacasside D (5), cinnacasside E (6), vilsonol F (7), (3S,5R,6S,7E,9R)‐3,5,6,9‐tetrahydroxy‐7‐en‐megastigmane (8), (3S,5R,6R,7E,9R)‐3,6,9,10‐tetrahydroxy‐7‐en‐megastigmane (9), citroside A (10), threo‐1‐(3,4,5‐trimethoxyphenyl)‐1,2,3‐propanetriol (11), 3,4,5‐trimethoxyphenyl‐1‐O‐β‐D‐glucopyranoside (12), p‐coumaric acid (13), ferulic acid (14) by HR‐ESI‐MS, ECD, 1D‐, and 2D‐NMR spectra. Compounds 1, 3, 4, and 9 showed NO production inhibitory activity in LPS activated RAW 264.7 cells with IC50 values of 42.0, 67.9, 20.5, and 78.6 μM, respectively, while the remaining compounds were inactive with IC50 values over 100 μM.

Abstract:Two new sulphur-containing amides, glylucidamides C-D (1-2), along with twelve known analogues (3-14) were isolated and characterised from the leaves of Glycosmis lucida. The chemical structures of 1 and 2 were elucidated by extensive spectroscopic methods and the known compounds were identified by comparisons their data with those reported in the literatures. All new compounds were evaluated for their anti-inflammatory activities via examining the inhibitory activity on nitric oxide (NO) production induced by lipopolysaccharide in mouse macrophage RAW 264.7 cells in vitro.

Abstract:Chemical examination of the gorgonian coral Junceella fragilis resulted in the isolation of four pairs of acetyl isomers belonging to briarane diterpenoids, including five new compounds. Their structures were determined on the basis of extensive spectroscopic (IR, MS, NMR, and single-crystal X-ray diffraction) analysis in association with the chemical conversion. Each pair of isomers featured by dynamical interconversion through as acetyl migration in 1,2-diol, which was postulated to be generated under the formation of a cyclic orthoacetate intermediate. All compounds exerted the inhibitory activities against the nitric oxide production in RAW264.7 macrophage cells.

Abstract:Introduction: Dimeric natural products are widespread in plants and microorganisms, which usually have complex structures and exhibit greater bioactivities than their corresponding monomers. In this study, we report five new dimeric tetrahydroxanthones, aculeaxanthones A−E ( 4−8 ), along with the homodimeric tetrahydroxanthone secalonic acid D ( 1 ), chrysoxanthones B and C ( 2 and 3 ), and 4−4'-secalonic acid D ( 9 ), from different fermentation batches of the title fungus. Methods: A part of the culture was added to a total of 60 flasks containing 300 ml each of number II fungus liquid medium and culture 4 weeks in a static state at 28 ̊C. The liquid phase (18 L) and mycelia was separated from the fungal culture by filtering. A crude extract was obtained from the mycelia by ultrasound using acetone. To obtain a dry extract (18 g), the liquid phase combined with the crude extract were further extracted by EtOAc and concentrated in vacuo. The MIC of anaerobic bacteria was examined by a broth microdilution assay. To obtain MICs for aerobic bacteria, the agar dilution streak method recommended in Clinical and Laboratory Standards Institute document (CLSI) M07-A10 was used. Compounds 1−9 was tested against the Bel-7402, A-549 and HCT-116 cell lines according to MTT assay. Results and Discussion: The structures of these compounds were elucidated on the base of 1D and 2D NMR and HR-ESIMS data, and the absolute configurations of the new xanthones 4−8 were determined by conformational analysis and time-dependent density functional theory-electronic circular dichroism (TDDFT-ECD) calculations. Compounds 1–9 were tested for cytotoxicity against the Bel-7402, A549, and HCT-116 cancer cell lines. Of the dimeric tetrahydroxanthone derivatives, only compound 6 provided cytotoxicity effect against Bel-7402 cell line (IC50, 1.96 μM). Additionally, antimicrobial activity was evaluated for all dimeric tetrahydroxanthones, including four Gram-positive bacteria including Enterococcus faecium ATCC 19434, Bacillus subtilis 168, Staphylococcus aureus ATCC 25923 and MRSA USA300; four Gram-negative bacteria, including Helicobacter pylori 129, G27, as well as 26,695, and multi drug-resistant strain H. pylori 159, and one Mycobacterium M. smegmatis ATCC 607. However, only compound 1 performed activities against H. pylori G27, H. pylori 26695, H. pylori 129, H. pylori 159, S. aureus USA300, and B. subtilis 168 with MIC values of 4.0, 4.0, 2.0, 2.0, 2.0 and 1.0 μg/mL, respectively.

Abstract:Four pairs of undescribed chromane and chromene meroterpenoid scalemic mixtures (1a/1b-4a/4b), together with three pairs of known chromane meroterpenoid ones (5a/5b-7a/7b) were isolated from the twigs and leaves of Rhododendron dauricum L. Among them, 1a/1b-3a/3b and 5a/5b-7a/7b were the chromane ones derived from an intramolecular [2 + 2] cyclic addition of their respective chromene precursors, forming a 6/6/6/4 and 6/6/5/4 ring fused scaffold. The absolute configurations of the chiral center at C-15 of 2a/2b were determined by Snatzke's method, and comparing the experimental and calculated electronic circular dichroism (ECD) data. The inhibitory effects of the isolated compounds were tested against lipopolysaccharide (LPS)-induced nitric oxide production in RAW264.7 macrophage cells to evaluate their anti-inflammatory activity. Compounds 4a, 4b and 6a displayed inhibitory effects on nitric oxide (NO) production, and compound 4b exhibited the obvious anti-inflammatory activity, with an IC50 value of 6.91 ± 0.97 μM, by downregulating nuclear factor kappa B (NF-κB) and reducing the expression of inducible nitric oxide synthase (iNOS) in LPS-induced RAW264.7 cells. These results intimated that 4b could be used as a leading compound to develop anti-inflammatory drugs and is worthy of further investigated.

Abstract:Three new C10 and C12 aliphatic δ‐lactones (1–3), three new fatty acid methyl esters (4–6), and eight known compounds (7–14) were isolated from the marine Aureobasidium sp. LUO5. Their structures were established by detailed analyses of the NMR, HRESIMS, optical rotation, and ECD data. All isolates were tested for their inhibitory effects on nitric oxide production in LPS‐induced BV‐2 cells. Notably, compound 4 displayed the strongest inhibitory effect with the IC50 value of 120.3 nM.

Abstract:Members of the Meliaceae family have been studied extensively because they contain a series of interesting, structurally diverse and biologically important triterpenoids. Plants from the genus Aphanamixis of Meliaceae have a long history of medicinal use. Substantial investigations into their chemical and pharmacological properties have been performed. Recently, interest in this genus has been centred on their production of complex, highly oxidised tetranortriterpenoids (also known as limonoids or meliacins), which exhibit potent cytotoxic and antitumour activities against various human cancer cell lines and in preclinical animal models. This review summarises triterpenoids isolated from Aphanamixis plants and those triterpenoids' bioactivities, structure-activity relationships and biosyntheses.

Abstract:Six new sesquiterpene quinone/hydroquinone meroterpenoids, arenarialins A-F (1-6), were isolated from the marine sponge Dysidea arenaria collected from the South China Sea. Their chemical structures and absolute configurations were determined by HRMS and NMR data analyses coupled with DP4+ and ECD calculations. Arenarialin A (1) features an unprecedented tetracyclic 6/6/5/6 carbon skeleton, whereas arenarialins B-D (2-4) possess two rare secomeroterpene scaffolds. Arenarialins A-F showed inhibitory activity on the production of inflammatory cytokines TNF-alpha and IL-6 in LPS-induced RAW264.7 macrophages with arenarialin D regulating the NF-kappa B/MAPK signaling pathway.
