Abstract:Two new alkaloids, longphyllinesides A (1) and B (2), were isolated from Daphniphyllum longeracemosum. Their structures were elucidated using 1D and 2D NMR spectra. The key step in their biogenetic pathway is indicated to be Diels–Alder cycloaddition via asynchronous concerted mechanism by density functional theory calculations. The possibility of involvement of a Diels–Alderase is also discussed.

Abstract:The phytochemical investigation of extracts from the radix of Angelica sinensis yielded a new phthalide dimer, 3,3'Z-6.7',7.6'-diligustilide, along with a known dimer, levistolide A. Their structures were established on the basis of spectral data, particularly using 1D-NMR and several 2D shift-correlated NMR pulse sequences (¹H-¹H COSY, HSQC, HMBC and NOESY).

Abstract:Angelicae Sinensis Radix,dried roots of Angelicae sinensis,is a well-known traditional Chinese medicine. Six compounds were isolated and purified by silica gel and preparative HPLC. According to physicochemical properties and spectral data,the compounds were identified as senkyunolide H-7-acetate( 1),o-phthalic acid( 2),diisobutyl phthalate( 3),p-hydroxyphenylethanol ferulate( 4),ferulic acid( 5) and coniferylferulate( 6). Compound 1 was a new one.

Abstract:(-)-Daphnilongeranin B and (-)-daphenylline are two hexacyclic Daphniphyllum alkaloids, each containing a complex cagelike backbone. Described herein are the first asymmetric total synthesis of (-)-daphnilongeranin B and a bioinspired synthesis of (-)-daphenylline with an unusual E ring embedded in a cagelike framework. The key features include an intermolecular [3+2] cycloaddition, a late-stage aldol cyclization to install the F ring of daphnilongeranin B, and a bioinspired cationic rearrangement leading to the tetrasubstituted benzene ring of daphenylline.

Abstract:A phytochemical investigation of the fruits of Daphniphyllum longeracemosum K. Rosenthal led to the isolation of a novel fused-hexacyclic alkaloid, longeracemine (1), along with a new one, demethyl calyciphylline A (2). Their structures were established on the basis of comprehensive spectra associated with quantum chemical analysis. Bioassay firstly indicated that Daphniphyllum alkaloid 1 can stimulate wheat shoot elongation.

Abstract:Here, we wish to describe our detailed efforts in the total synthesis of three calyciphylline A-type alkaloids, namely (+)-daphlongamine E, (+)-calyciphylline R, and (–)-10-deoxydaphnipaxianine A. Abstract:Daphnezomines A and B are structurally unusual Daphniphyllum alkaloids that contain a unique aza-adamantane core skeleton. Herein, a modular approach to these alkaloids is presented that exploits a diverse array of reaction strategies. Commencing from a chiral pool terpene–(S)-carvone, the azabicyclo[3.3.1]nonane backbone, which occurs widely in Daphniphyllum alkaloids, was easily accessed through a Sharpless allylic amination and a palladium-catalyzed oxidative cyclization. A protecting group enabled a stereoselective B-alkyl Suzuki–Miyaura coupling sequence and an Fe-mediated hydrogen atom transfer (HAT)-based radical cyclization were then applied to construct C6 and C8 stereocenters. Abstract:Thirteen new benzamide alkaloids, delphiniumines A–M (1–13), together with one known analogue (14), were isolated from Delphinium anthriscifolium Hance. All of the structures were determined by spectroscopic and spectrometric analyses. Absolute configuration for 1 was established using experimental and calculated ECD data, as well as by X-ray crystallography analysis. Compound 1 possesses a previously undescribed polysubstituted cyclopentene carbon framework. Compound 2 was isolated as an artifact from 1 during the extraction process. Compound 7 is glycosylated with a β-D-glucose unit. Compound 13 bears a chlorine substituent. Abstract:The fruits, twigs and leaves of Daphniphyllum oldhamii (Hemsl.) K. Rosenthal, collected from Longshan County, Hunan Province, China, were chemically investigated. Three undescribed daphniphyllum alkaloids, namely longshanoldhamines A‒C, and six known related ones have been isolated from the fruits, whereas two undescribed triterpenoids and one undescribed lignan, along with six known triterpenoids, were found in the twigs and leaves. Their structures were elucidated by extensive spectroscopic analysis, X-ray diffraction analysis and comparison with the reported data.

Abstract:A new abietane-type diterpenoid, dalterpenoid A (1), a new long-chain alkenone derivative, (3E,5E,10E)-8-hydroxytrideca-3,5,10,12-tetraen-2-one (2), together with six known compounds (3-8), namelyepi-guaidiol A (3), xylaranol A (4), daldinone C (5),trans-3,4-dihydroxy-3,4-dihydro-anofinic acid (6), (R)-6-hydroxymellein (7), helicascolide A (8), were obtained from fungusDaldiniasp. TJ403-LS1, which was originally isolated from roots of the medicinally valuable plantAnoectochilus roxburghii. The structures of compounds1and2were established based on widespread spectroscopic methods, mainly including 1D & 2D NMR and HRESIMS analyses, and the absolute configuration of1was further confirmed by electronic circular dichroism (ECD) calculation. All new compounds were tested for the in vitro cytotoxicity against five human cancer cell lines.
