Abstract:An unprecedented cascade Heck-Aldol-Heck reaction was developed to form two C-C single bonds and one C=C double bond in one process by a combination of palladium(0) catalysis and aminocatalysis. Various aryl iodides could perform the cascade reaction with readily available propenol and formaldehyde to afford novel (E)-trisubstituted alkenes in 66-81% yields.

Abstract:Palladium-catalyzed enantioselective domino Heck/intramolecular C-H functionalization reaction, as a valuable strategy for creating molecular diversity, has remained a prominent challenge. Here, we describe a Pd/XuPhos catalyst for asymmetric domino Heck/intermolecular C-H alkylation of unactivated alkenes with diverse polyfluoro- and heteroarenes in a highly chemo- and enantioselective manner. This process enables efficient synthesis of various dihydrobenzofurans, indolines and indanes, which are of interest in pharmaceutical research and other areas. Late-stage modifications of the core structures of natural products are also well showcased. Moreover, synthetic transformations create a valuable platform for preparing a series of functionalized molecules. Several control experiments for mechanistic study are conducted to pursue a further understanding of the reaction.

Abstract:Abstract Heck-type C-H bond activation of unactivated alkenes has emerged as a powerful strategy for the construction of synthetically valuable spirocycles over past 30 years, however, the development of asymmetric version has lagged largely behind. Herein we demonstrate a robust Heck-type reaction of a broad range of unactivated alkenes enabled by the first palladium/Xu-Phos-catalyzed tandem Heck/remote C−H bond alkylation. Moreover, the synthesis of both enantiomers of the product using the same enantiomer of a chiral ligand via a position of the phenyl ring-dependent enantiodivergent synthesis. The salient features of this methodology include operational simplicity, high chemo- and enantioselectivity, broad substrate scope. In addition, we first revealed that the C(sp2)-H activation, alkene insertion and C−I reductive elimination steps are reversible by experiments.

Abstract:Elaborated molecular architectures including complex carbocycles, heterocycles, and polycyclic molecular structures represent an important class of organic compounds because they are ubiquitous structural units in a broad variety of biologically and pharmacologically relevant natural products, medicinal molecules, and functional materials. A major challenge in modern synthetic methodology is the development of strategically efficient and selective construction of these compounds from readily available starting materials. Transition‐metal‐catalyzed transformations involving Heck reaction as the pivotal step have afforded a powerful approach for accessing sophisticated polycyclic skeletons from a set of easy‐to handle starting materials, thus highlighting its potential significance in streamlining the drug discovery process. Over the past few decades, significant effort has been devoted to accessing complex polycyclic derivatives in efficient and versatile routes, and a number of powerful and direct synthetic strategies have been reported. In this review, the latest advances in transition‐metal‐catalyzed transformations involving Heck reaction as the critical step are summarized, which could be divided into six categories: 1) Heck/Sonogashira tandem reaction; 2) Heck/Suzuki tandem reaction; 3) Heck/Heck tandem reaction; 4) Heck/Hiyama tandem reaction; 5) Heck/Cacchi tandem reaction; 6) Heck/C‐H bond functionalization tandem reaction.

Abstract:Recent advances in Heck reaction are highlighted according to the types of carbon and heteroatom electrophiles. The last two decades have witnessed significant progress in asymmetric Heck reaction of aryl/alkenyl halides, sulfonates and aryldiazonium salts as in Heck Matsuda reaction. Heck-type reactions that used new types of carbon electrophiles also flourished, examples including unactivated alkyl halides possessing eliminable β-hydrogens, activated alkyl halides (e.g., α-haloesters and α-halomalonates), benzylic, allylic and propargylic electrophiles. Significant progress was also made in Heck reactions using heteroatom electrophiles, such as the innovation of enantioselective Narasaka-Heck cyclization of oxime derivatives, aza-Heck cyclization of hydroxamate derivatives and the emergence of Heck-type reactions of silyl and boryl halides.

Abstract:As an important unit of molecular structure and organic building blocks, the development of new synthetic methods of allene has received much attention. The Heck and related cascade reaction of alkenes is arguably one of the most synthetically versatile method. However, the Heck reaction of alkyl-alkynes leading to allenes has lagged behind due to the energetically unfavored beta-hydride elimination of vinyl palladium species. Several competitive reactions such as protonation, carbohalogenation and cascade reaction exist. Moreover, the isomerization of allene and the regioselectivity of unsymmetrical alkyl-alkynes also limited this research progress. To effectively promote beta-hydride elimination of vinyl palladium species, several commonly-used strategies are the increasing reaction temperature, introduction of ortho-substituent to aryl halides and the development of ligand. In this mini-review, Heck and related reaction of alkyl-alkynes including beta-hydride elimination reactions of vinyl palladium species, protonation reaction, carbohalogenation reaction, and cascade reaction are emphasized. Finally, the major limitation and an outlook this type of reaction are provided.

Abstract:An efficient approach to functionalized (E)-3-cinnamyl-3-methyl-2,3-dihydrobenzofurans and (E)-(3-methyl-2,3-dihydrobenzofuran-3-yl)but-2-enones has been developed through a Pd-catalyzed one-pot cascade process involving two sequential Heck reactions, that is, an intramolecular Heck reaction of olefin-tethered aryl iodides and an intermolecular Heck reaction with substituted styrenes and α,β-unsaturated ketones. As a result, a series of desired products were obtained in moderate to good yields and with exclusive E-form selectivities.

Abstract:Geminal bis(boronates) are versatile synthetic building blocks in organic chemistry. The fact that they predominantly serve as nucleophiles in the previous reports, however, has restrained their synthetic potential. Herein we disclose the ambiphilic reactivity of α-halogenated geminal bis(boronates), of which the first catalytic utilization was accomplished by merging a formal Heck cross-coupling with a highly diastereoselective allylboration of aldehydes or imines, providing a new avenue for rapid assembly of polyfunctionalized boron-containing compounds. We demonstrated that this cascade reaction is highly efficient and compatible with various functional groups, and a wide range of heterocycles. In contrast to a classical Pd(0/II) scenario, mechanistic experiments and DFT calculations have provided strong evidence for a catalytic cycle involving Pd(I)/diboryl carbon radical intermediates.

Abstract:An asymmetric cascade annulation between 2-hydroxystyrenes and 2-alkynylbenaldehyes or 1-(2-alkynylphenyl)ketones has been established with good to excellent enantioselectivities (up to >99.5% ee), on the basis of an enantioselective oxa-Diels-Alder cycloaddition of in situ generated metallo-isochromenylium intermediates, by cooperative binary catalysis of Pd(OAc)2 and (S)-Trip. The developed methodology is workable for a broad spectrum of substrates and shows great efficiency in establishing dense multiple chiral centers including quaternary carbons of variable bridged ring systems. The mechanism study suggests that (S)-Trip plays multiple roles in assembling the reactants and controlling the stereoselectivity.
