Abstract:We report the first catalytic method for activating the acyl C-O bonds of methyl esters through an oxidative-addition process. The oxidative-addition adducts, formed using nickel catalysis, undergo in situ trapping to provide anilide products. DFT calculations are used to support the proposed reaction mechanism, to understand why decarbonylation does not occur competitively, and to elucidate the beneficial role of the substrate structure and the Al(OtBu)(3) additive on the kinetics and thermodynamics of the reaction.

Abstract:A rhodium(III)-catalyzed cascade reaction of phthalic anhydrides with cyclic 2-diazo-1,3-diketones and methanol for modular access to esterified cyclohexenone-fused isocoumarins has been developed. This strategy involves the process of one C-C and two C-O bonds formation through C-H activation, transannulative coupling and subsequent annulation process. Furthermore, no additive and base are needed in this transformation, and interestingly, the reaction could proceed well under air atmosphere with broad substrate and good functional groups tolerance.

Abstract:A carbene-catalyzed asymmetric access to chiral β-cyano carboxylic esters is disclosed. The reaction proceeds between β,β-disubstituted enals and aromatic thiols involving enantioselective protonation of enal β-carbon. Two main factors contribute to the success of this reaction. One involves in situ ultrafast addition of the aromatic thiol substrates to the carbon-carbon double bond of the enal substrate. This reaction converts almost all enal substrate to a Thiol-click Intermediate, significantly reducing aromatic thiol substrates concentration and suppressing the homo-coupling reaction of enals. Another factor is an in situ release of enal substrate from the Thiol-click Intermediate for the desired reaction to proceed effectively. The optically enriched β-cyano carboxylic esters from our method can be readily transformed to medicines that include γ-aminobutyric acids derivatives such as Rolipram. In addition to synthetic utilities, our control of reaction outcomes via in situ substrate modulation and release can likely inspire future reaction development.

Abstract:Esters are valuable commodity chemicals widely found in agrochemicals, pharmaceuticals, and advanced materials. They are also important synthetic building blocks for chemical transformation. Recent advances of ester synthesis via transition-metal-catalyzed C-H activation have provided highly efficient and atom-economical alternatives to the traditional methods. Herein, we summarize recent advances on ester synthesis via transition-metal-catalyzed C-H activation. On the basis of the modes of reactivity and the types of C-H bonds, transition-metal-catalyzed alkoxycarbonylation of C-H bonds with various esterification reagents will be discussed in sections 2 and 3. Finally, hydroesterification/hydroacylation of C-C or C-O double bonds via formate C-H activation will be discussed in sections 4 and 5.

Abstract:Chiral dialkyl carbinols and their derivatives are significant synthetic building blocks in organic chemistry and related fields. The development of convenient and efficient methods to access these compounds has long been an important endeavor. Herein, we report a NiH-catalyzed reductive hydroalkylation and hydroarylation of enol esters and ethers. alpha-Oxoalkyl organonickel species were generated in situ in a catalytic mode and then participated in cross-coupling with alkyl or aryl halides. This approach enabled C(sp(3))-C(sp(3)) and C(sp(3))-C(sp(2)) bond formation under mild reductive conditions with simple operations, thereby boosting a broad substrate scope and good functional compatibility. Esters of enantioenriched dialkyl carbinols were accessed in a catalytic asymmetric version. Mechanistic studies demonstrated that this reaction proceeded through a syn-addition of Ni-H intermediate to an enol ester with high regio- and enantioselectivity. [GRAPHICS] .

Abstract:The highly enantioselective preparation of spirooxindoles bearing α,α-disubstituted α-amino-β-keto esters was achieved through [4 + 1] annulation of oxindoles and α-imine-β-oxo-γ,δ-unsaturated esters under mild conditions in good yields (up to 82%) and stereoselectivities (up to >20:1 dr, 96% ee). The reaction is amenable to gram scale synthesis using catalyst loading as low as 1 mol %. The corresponding chiral α,α-disubstituted α-amino-β-keto esters could be easily transformed into cyclopenta[ b]indole derivatives without erosion of enantiopurity.

Abstract:The N-heterocyclic carbene catalyzed annulation of benzofuran-2,3-diones and enals via homoenolate intermediates is described. The reaction provided a direct and efficient method for the synthesis of spiro-bis-lactones. The ketone-carbonyl group annulated products and the ester-carbonyl group annulated products can be obtained as major products with good yields by convenient catalyst regulation. Furthermore, commercially available thiazolium salt can also catalyze this reaction with modest yield.
