Abstract:We report the first catalytic method for activating the acyl C-O bonds of methyl esters through an oxidative-addition process. The oxidative-addition adducts, formed using nickel catalysis, undergo in situ trapping to provide anilide products. DFT calculations are used to support the proposed reaction mechanism, to understand why decarbonylation does not occur competitively, and to elucidate the beneficial role of the substrate structure and the Al(OtBu)(3) additive on the kinetics and thermodynamics of the reaction.

Abstract:Although enzymes are able to cleave amide bonds in nature, it is difficult to selectively break the carbon–nitrogen bond of an amide using synthetic chemistry; now the activation and cleavage of these bonds using nickel catalysts is used to convert amides to esters. Although enzymes are able to cleave amide bonds in nature, it is difficult to selectively break the carbon–nitrogen bond of an amide using synthetic chemistry. In this paper the authors demonstrate that amide C–N bonds can be activated and cleaved using nickel catalysts. They used this methodology to convert amides to esters, which is a challenging and underdeveloped transformation. Amides are common functional groups that have been studied for more than a century1. They are the key building blocks of proteins and are present in a broad range of other natural and synthetic compounds. Amides are known to be poor electrophiles, which is typically attributed to the resonance stability of the amide bond1,2. Although amides can readily be cleaved by enzymes such as proteases3, it is difficult to selectively break the carbon–nitrogen bond of an amide using synthetic chemistry. Here we demonstrate that amide carbon–nitrogen bonds can be activated and cleaved using nickel catalysts. We use this methodology to convert amides to esters, which is a challenging and underdeveloped transformation. The reaction methodology proceeds under exceptionally mild reaction conditions, and avoids the use of a large excess of an alcohol nucleophile. Density functional theory calculations provide insight into the thermodynamics and catalytic cycle of the amide-to-ester transformation. Our results provide a way to harness amide functional groups as synthetic building blocks and are expected to lead to the further use of amides in the construction of carbon–heteroatom or carbon–carbon bonds using non-precious-metal catalysis.

Abstract:Ni-catalyzed C−O bond activation of esters provides an unprecedented synthetic approach to utilize esters. To understand the mechanistic details of the Ni-catalyzed C−O bond activation, computational chemists have dedicated remarkable efforts to elucidate the origins of reactivities and selectivities of this important process. In order to provide a general perspective of this exciting area, this review summarizes the recent advances of the computational studies involving Ni-catalyzed C−O bond activation of esters. The mechanistic models of C−O bond cleavage are categorized in three types, and the key mechanistic information regarding the control of reactivities and selectivities is presented.

Abstract:The direct conversion of alkyl esters to ketones has been hindered by the sluggish reactivity of the starting materials and the susceptibility of the product towards subsequent nucleophilic attack. We have now achieved a cross-coupling approach to this transformation using nickel, a bulky N-heterocyclic carbene ligand, and alkyl organoboron coupling partners. 65 alkyl ketones bearing diverse functional groups and heterocyclic scaffolds have been synthesized with this method. Catalyst-controlled chemoselectivity is observed for C(acyl)-O bond activation of multi-functional substrates bearing other bonds prone to cleavage by Ni, including aryl ether, aryl fluoride, and N-Ph amide functional groups. Density functional theory calculations provide mechanistic support for a Ni-0/Ni-II catalytic cycle and demonstrate how stabilizing non-covalent interactions between the bulky catalyst and substrate are critical for the reaction's success.
Mechanisms and Origins of Switchable Chemoselectivity of Ni-Catalyzed C(Aryl)-O and C(Acyl)-O Activation of Aryl Esters with Phosphine Ligands

Abstract:Many experiments have shown that nickel with monodentate phosphine ligands favors the C(aryl)-O activation over the C(acyl)-O activation for aryl esters. However, Itami and co-workers recently discovered that nickel with bidentate phosphine ligands can selectively activate the C(acyl)-O bond of aryl esters of aromatic carboxylic acids. The chemoselectivity with bidentate phosphine ligands can be switched back to C(aryl)-O activation when aryl pivalates are employed.To understand the mechanisms and origins of this switchable chemoselectivity, density functional theory (DFT) calculations have been conducted. For aryl esters, nickel with bidentate phosphine ligands cleaves C(acyl)-O and C(aryl)-O bonds via three-centered transition states. The C(acyl)-O activation is more favorable due to the lower bond dissociation energy (BDE) of C(acyl)-O bond, which translates into a lower transition-state distortion energy. However, when monodentate phosphine ligands are used, a vacant coordination site on nickel creates an extra Ni-O bond in the five-centered C(aryl)-O cleavage transition state. The additional interaction energy between the catalyst and substrate makes C(aryl)-O activation favorable. In the case of aryl pivalates, nickel with bidentate phosphine ligands still favors the C(acyl)-O activation over the C(aryl)-O activation at the cleavage step. However, the subsequent decarbonylation generates a very unstable tBu-Ni(II) intermediate, and this unfavorable step greatly increases the overall barrier for generating the C(acyl)-O activation products. Instead, the subsequent C-H activation of azoles and C-C coupling in the C(aryl)-O activation pathway are much easier, leading to the observed C(aryl)-O activation products.

Abstract:Two efficient protocols for the nickel-catalyzed aryl–alkyl cross-coupling reactions using esters as coupling components have been established. The methods enable the selective oxidative addition of nickel to acyl C–O and aryl C–O bonds and allow the aryl–alkyl cross-coupling via decarbonylative bond cleavage or through cleavage of a C–O bond with high efficiency and good functional group compatibility. The protocols allow the streamlined, unconventional utilization of widespread ester groups and their precursors, carboxylic acids and phenols, in synthetic organic chemistry.

Abstract:Ni-catalyzed selective C-O bond activation opens a door for the cross-coupling of aryl esters. The present study reports a thorough theoretical analysis of Ni-catalyzed cross-coupling between aryl esters and arylboronic acids, with an emphasis on explaining the cause for the surprising selectivity in C-O activation. The overall catalytic cycle is found to include three basic steps: oxidative addition, transmetalation, and reductive elimination. Oxidative addition of Ar-OAc to Ni(0) in the presence of PCy3 ligand proceeds through the monophosphine pathway (instead of the alternative two-phosphine pathway) with a relatively low barrier of +22.9 kcal/mol. Transmetalation proceeds via a base-assisted mechanism with a barrier of +31.2 kcal/mol. Reductive elimination is the most facile step in the whole catalytic cycle. Comparatively, oxidative addition of ArO-Ac to Ni(0) is a more facile process (barrier = +14.2 kcal/mol) than oxidative addition of Ar-OAc to Ni(0). However, the former process is associated with a fairly low reverse barrier, and its product does not transmetalate easily (barrier = +33.1 kcal/mol). By comparison, the latter process is an irreversible reaction, and its product transmetalates more readily. These results explain why only the cross-coupling products from the Ar-OAc activation (but not from the ArO-Ac activation) were observed in experiments.

Abstract:The nickel-catalyzed direct borylation and silylation of phenolic esters has been established. The key to this highly efficient acyl C–O bond borylative and silylative cleavage depends on the appropriate choice of different ligands and additives in the presence of nickel catalyst. Both transformations exhibit good functional group compatibility and can serve as powerful synthetic tools for late-stage functionalization of complex compounds. The elucidation of key mechanistic features of this newly developed acyl C–O bond activation in esters was confirmed by two well-characterized organonickel(II) intermediates.

Abstract:Amide, as a ubiquitous functional group, is essential in various aspects of chemistry and biology. Although the history of studying amide is rich and fruitful, the synthetic application of amide is very limited due to the inertness of amide C–N bond. Recently,significant advances have been achieved towards the nickel-mediated C–N activation of amides. This approach allows a facile generation of acyl-nickel intermediates, and a number of unique transformations have been designed and realized based on the amide C–N bond activation. Focused on the catalytic transformation, this review summarizes and categorizes the recent advances on the synthetic applications of Ni-mediated C–N bond activation of amides.

Abstract:An efficient nickel-catalyzed decarbonylative cyanation reaction which allows the direct functional-group interconversion of readily available esters into the corresponding nitriles was developed. This reaction successfully offers access to structurally diverse nitriles with high efficiency and excellent functional-group tolerance and provides a good alternative to classical synthetic pathways from diazonium salts or organic halide compounds.

Abstract:We report the amination of aryl carbamates using nickel-catalysis. The methodology is broad in scope with respect to both coupling partners and delivers aminated products in synthetically useful yields. Computational studies provide the full catalytic cycle of this transformation, and suggest that reductive elimination is the rate-determining step. Given that carbamates are easy to prepare, robust, inert to Pd-catalysis, and useful for arene functionalization, these substrates are particularly attractive partners for use in synthesis. The sequential use of carbamate functionalization/site-selective cross-coupling processes highlights the utility of this methodology.

Abstract:A ligand-controlled and site-selective nickel catalyzed Suzuki–Miyaura cross-coupling reaction with aromatic esters and alkyl organoboron reagents as coupling partners was developed. This methodology provides a facile route for C(sp2)–C(sp3) bond formation in a straightforward fashion by successful suppression of the undesired β-hydride elimination process. By simply switching the phosphorus ligand, the ester substrates are converted into the alkylated arenes and ketone products, respectively. The utility of this newly developed protocol was demonstrated by its wide substrate scope, broad functional group tolerance and application in the synthesis of key intermediates for the synthesis of bioactive compounds. Abstract:Herein we report a nickel/photo-cocatalyzed C(sp2)–H allylation of aldehydes and formamides wherein both allyl acetates and allyl alcohols can be used as the allylating agents. In this reaction, radical-type umpolung of the formyl moiety is enabled by tetrabutylammonium decatungstate as a hydrogen-atom-transfer photocatalyst, whereas nickel serves to cleave the C–O bond of allyl acetates or allyl alcohols. Abstract:A simple catalyst of Ni(OAc)(2) and P(t-Bu)(3) enables selective C-alkylation of thioacetamides and primary acetamides with alcohols for the first time. Monoalkylation of thioamides, amides and t-butyl esters occurs in excellent yields (>95%). Mechanistic studies reveal that the reaction proceeds via a hydrogen autotransfer pathway.
