Abstract:An enzymatic method which combines D-aminoacylase catalyzed nitroaldol reaction and lipase PS-IM catalyzed acyl resolution reaction was constructed to obtain optically pure beta-nitroalcohols in organic media under mild conditions. A series of beta-nitroalcohols were synthesized and then resolved, with excellent ee values (>95%) and yields mostly, giving enantiomerically pure acetate and alcohol of the opposite configuration simultaneously.

Abstract:1.1. Material. Lipase immobilized on acrylic resin from Candida antarctica (≥10,000 U g −1 , recombinant, expressed in Aspergillus oryzae), Amano lipase PS-C I (immobilized on ceramic), Amano lipase PS-IM (immobilized on diatomaceous earth) and Amano lipase M from M. javanicus (≥10,000 U g −1 enzyme activity, pH 7.0, 40 °C) were purchased from Sigma-Aldrich (Steinheim, Germany), Lipase from hog pancreas (2.4 U mg −1 , 1 U is the amount of immobilized enzyme which forms 1% octyl laurate from 0.5 mmol lauric acid and 1.0 mmol 1-octanol in 10 ml water-saturated isooctane in 1 h at 20 °C) was purchased from Fluka (Switzerland)., Lipozyme®, immobilized from Mucor miehei (42 U g −1 , 1 U corresponds to the amount of enzyme which liberates 1 mol oleic acid at pH 8.0 and 40 °C per minute), D-Aminoacylase from Escherichia. coli (10,000 U mg −1 , 1 U is defined as enzyme quantity which produces 1 μmol of D-Amino acid per 30 min). All solvents were analytical grade and were dried by storing over activated 3 A molecular sieves for 24 h prior to use. All other reagents were analytical grade and used as received.

Abstract:Aromatic aldehydes are very important organic intermediates,which are widely used in pharmaceuticals,pesticides and other areas.In this paper,an improved synthetic method of aromatic aldehydes containing electrowithdrawing groups in ortho or para(or both) position of the aromatic ring was introduced.4-choloro-2-nitrotoluene,4-nitrotoluene and 4-cyanotoluene were used as raw materials,respectively,via enamination with N,N-dimethylformamide dimethyl acetal(DMFDMA) in the present of N,N-dimethylformamide(DMF) as solvent and under reflux,the corresponding styrylamine compounds were produced;then the intermediates were further oxidized by H2O2 under different reaction conditions.The effects of solvent species,reaction temperature and the used amount of H2O2 on the product yield were studied,and the optimal reaction conditions were determined.Under those optimal conditions,the best yields of different aldehydes are 75.6%(4-chloro-2-nitrobenzaldehyde),58.3%(4-nitrobenzaldehyde) and 68.2%(4-cyanobenzaldehyde),respectively with the advantages of simple process,low cost and being environment-friendly..The main intermediates and objective products were confirmed by HPLC and 1H-NMR.

Abstract:A green and fully enzymatic procedure for the resolution of chiral alcohols through lipase/esterase-catalyzed acylation and subsequent lipase-catalyzed aminolysis using anhydrous ammonia was demonstrated. Both enantiomers can be obtained in high ee values (up to >99%) under ambient reaction conditions. The solvent and acyl donors can be recycled, and the enzyme can be reused for up to five times.

Abstract:Esterase BioH, which is obligatory for biotin synthesis in Escherichia coli, was found to exhibit a promiscuous ability to catalyse Aldol and Knoevenagel reactions with moderate to good yields. The reaction conditions including organic solvent, molar ratio of ketone to aldehyde, enzyme amount, and reaction time were investigated to evaluate the effect of different reaction conditions on yield. Target compounds were afforded in the best yield of 91.2% for Aldol reaction and 54.7% for Knoevenagel reaction. In addition, because the enzyme could be prepared with a low cost, this protocol could provide an economic route to conduct Aldol and Knoevenagel reactions, which expand the field of enzymatic promiscuity.

Abstract:The development and characterization of enantioselective catalytic kinetic resolution of allylic alcohols through asymmetric isomerization with chiral BINOL derivatives-based alkoxides as bifunctional Bronsted base catalysts were described in the study. A number of chiral BINOL derivatives-based alkoxides were synthesized, and their structure enantioselectivity correlation study in asymmetric isomerization identified a promising chiral Bronsted base catalyst, which afforded various chiral secondary allylic alcohols (ee up to 99%, S factor up to >200). In the mechanistic study, alkoxide species were identified as active species and the phenol group of BINOL largely affected the high reactivity and enantioselectivity via hydrogen bonding between the chiral Bronsted base catalyst and substrates. The strategy is the first successful synthesis strategy of various chiral secondary allylic alcohols through enantioselective transition metal-free base-catalyzed isomerization. The applicability of the strategy had been demonstrated by the synthesis of the bioactive natural product (+)-veraguensin.

Abstract:Readily available potassium alkynyltrifluoroborates were used for organocatalytic asymmetric conjugate alkynylation of β-enaminones. The interception of a modified binaphthol catalyst and in situ generated organodifluoroboranes proved important to access functionalized β-alkynyl-β-amino carbonyls and derivatives with improved chemo-reactivity and enantio-induction. Mechanistic studies revealed the impact of molecular sieves on efficiency and stereocontrol. Abstract:Alkylamine structures represent one of the most functional and widely used in organic synthesis and drug design. However, the general methods for the functionalization of the shielded and deshielded alkyl radicals remain elusive. Here, we report a general deoxygenative amination protocol using alcohol-derived carbazates and nitrobenzene under electrochemical conditions. A range of primary, secondary, and tertiary alkylamines are obtained. This practical procedure can be scaled up through electrochemical continuous flow technique. Secondary arylalkylamines represent important building blocks for organic synthesis, and as such their straightforward synthesis from readily available chemicals remains a priority in organic chemistry. Here, the authors report a synthesis of arylalkylamines from nitroarenes and carbazates via paired electrolysis.
